Novel rapid tissue lysis method to evaluate cancer proteins: Correlation between elevated Bcl-X_L expression and colorectal cancer cell proliferation

AIM： We optimized a rapid and efficient tissue lysis method using the MagNA Lyser （Roche, Germany）. Using this novel method combined with immunoblot analysis, we investigated the correlation between abnormal Bcl-XL expression and clinicopathological characteristics in colorectal cancer. METHODS： Tissue samples from Sprague-Dawley rats were tested to determine optimal lysis conditions for use with MagNA Lyser. We next used the new method to extract tissue proteins from the tumor tissue of a colorectal cancer patient. The availability of extractable tissue proteins for proteomic study was demonstrated by two-dimensional （2D） gel electrophoresis and subsequent matrix-assisted laser desorption/ionization time-of-flight （MALDI-TOF） mass spectrometry. In addition, we prepared tissue lysates from paired tumor tissues and adjacent nontumor tissues of 50 colorectal carcinoma patients. Ensuing immunoblot analyses were performed to detect the level of Bcl-X, expression. RESULTS： The optimal sample sizes processed were found to be around 200 mg, with oscillation frequency of 6 500 r/rain for 80 s. Test of the first human tissue lysate confirmed that the MagNA Lyser method was adequate for protein extraction and subsequent identification by current proteomic protocols. The method was also applicable to immunoblot analysis. Thirty of 50 （60%） colorectal patients exhibited higher level of Bcl-XL expression in their tumor tissues. Raised level of Bcl-XL expression correlated with patients＇ gender and tumor cell proliferation index （P= 0.037 and P〈0.001, respectively）, but was independent of clinicopathological characteristics and overall survival. CONCLUSION： We report a novel tissue lysis method applicable to proteomic and immunoblot analyses, which can facilitate the discovery and detection of cancer protein alterations.

Comparison of Vs and SPT Soil Liquefaction Assessments of NCEER: Including Hypothesis Testing

Soil liquefaction is one of the complex research topics in geotechnical engineering and engineering geology. Especially after the 1964 Niigata earthquake (Japan) induced many soil liquefaction incidents, a variety of soil liquefaction studies were conducted and reported, including the liquefaction potential assessment methods utilizing the shear wave velocity (Vs) or SPT-N profiles (SPT: standard penetration test). This study used the Vs and SPT methods recommended by the National Center for Earthquake Engineering Research (NCEER) to examine which is more conservative according to the assessment results on 41 liquefiable soil layers at sites in two major cities in Taiwan. Statistical hypothesis testing was used to make the analysis more quantitative and objective. Based on three sets of hypothesis tests, it shows that the hypothesis—the SPT method is more conservative than the Vs method—was not rejected on a 5% level of significance.

Earthquake Magnitude Probability and Gamma Distribution

The earthquake magnitude probability distribution is one of the underlying input data for certain earthquake analyses, such as probabilistic seismic hazard analysis. Nowadays, the method proposed by McGuire and Arabasz (1990) is commonly used for obtaining the (simulated) earthquake magnitude probability distributions. However, based on the observed earthquake data in 5 regions (Taiwan, Japan, California, Turkey, and Greece), the model did not fit the observation well. Instead, all of the case studies show that using the newly proposed gamma distribution can improve the simulation significantly compared to the conventional method.

Percentage of peak-to-peak pulsatility of portal blood flow can predict right-sided congestive heart failure

AIM: To study the change of portal blood flow for the prediction of the status of right-sided heart failure by using non-invasive ***: We studied 20 patients with rheumatic and atherosclerotic heart diseases. All the patients had constant systemic blood pressure and body weight 1 week prior to the study. Cardiac index (CI), left ventricular end-diastolic pressure (LVEDP), mean aortic pressure (AOP), pulmonary wedge pressure (PWP), mean pulmonary arterial pressure (PAP), mean right atrial pressure (RAP), right ventricular end-diastolic pressure (RVEDP) were recorded during cardiac catheterization. Ten patients with RAP＜10 mmHg were classified as Group 1. The remaining 10 patients with RAP ≥ 10 mmHg were classified as Group 2. Portal blood velocity profiles were studied using an ultrasonic Doppler within 12h after cardiac ***: CI, AOP, and LVEDP had no difference between two groups. Patients in Group 1 had normal PWP (14.6±7.3mmHg), PAP (25.0±8.2 mmHg), RAP (4.7±2.4 mmHg), and RVEDP (6.4±2.7 mmHg). Patients in Group 2 had increased PWP (29.9±9.3 mmHg), PAP (46.3±13.2 mmHg), RAP (17.5±5.7 mmHg), and RVEDP (18.3±5.6 mmHg) (P＜0.001).Mean values of maximum portal blood velocity (Vmax), mean portal blood velocity (Vmean), cross-sectional area (Area)and portal blood flow volume (PBF) had no difference between 2 groups. All the patients in Group 1 had a continuous antegrade portal flow with a mean percentage of peak-topeak pulsatility (PP) 27.0±8.9 % (range: 17-40 %). All the patients in Group 2 had pulsatile portal flow with a mean PP 86.6±45.6 (range: 43-194 %). One patient had a transient stagnant and three patients had a transient hepatofugal portal flow, which occurred mainly during the ventricular systole. Vmax, Vmean and PBF had a positive correlation with CO (P＜0.001) but not with AOP, LVEDP, PWP, PAP,RAP, and *** showed a good correlation (P＜0.001)with PWP, PAP, RAP, and RVEDP but not with CI, AOP, and LVEDP. The building integrated photovoltaics （BIPV） application is one of the main study topics in the sustainable building field. In this paper, the high color rendering index （HCRI）-BIPV window system is developed to be used in the indoor environmental control, whose module material has appeared to be effective in improving the visible transmittance and reducing the absorption. This paper describes the performance of grid-connected HCRI-BIPV window system with 0.75 kWp capacity installed in an office building for a natural ventilation solution. The experimental results indicate that accumulative power generation of the HCRI-BIPV window system is 157.60 WKh during the 7-month experiment period. For consideration of each evaluated factors, the HCRI-BIPV window system not only offers the passive energy situation for its power loading but also improves the indoor thermal environment by natural ventilation.

Predictive Meteorological Factors for Elevated PM2.5 Levels at an Air Monitoring Station Near a Petrochemical Complex in yunlin County, Taiwan

Since 1991, air pollution has gained special attention in Taiwan after a petrochemical complex was constructed in Mailiao Township, yunlin County. We explored the association between the magnitude of PM2.5 and meteorological factors during 2012-2016. Our findings revealed that 1) mean PM2.5 levels gradually decreased from 30.70 μg/m3 in 2013 to 25.36 μg/m3 in 2016;2) wind speed is the main determinant of air quality—air quality significantly improved when it was faster than 4 m/sec;and 3) wind direction is another determinant of air quality—when the wind direction was southerly, air quality improved. Elevated PM2.5 levels were defined as those hourly levels higher than the third quartile (36 μg/m3). The significantly negative predictive factors for elevated PM2.5 levels were the summer or autumn seasons, rainfall, increased wind speed, and wind direction from 150° to 230° from the north. The significantly positive predictive factors for elevated PM2.5 levels were working hours from 6 a.m. to 2 p.m., a temperature between 11°C and 25°C, relative humidity between 40% and 68%, and wind direction (e.g., northerly wind, northeasterly wind, and easterly wind). The predictive formula is attached in the Appendix. Therefore, people should protect themselves on these high-risk days.

Minimally Invasive Open Reduction and Internal Fixation for Osteoporotic Vertebral Compression Fractures: Technical Report and Mid-Term Outcomes

Background: Traditional open instrumentation may cause surgical complications due to fragile bones and induce medical comorbidities in senile patients. Vertebroplasty and kyphoplasty are palliative augmentation procedures that have been associated with increased risks of cement leakage, adjacent fractures and non-union. Objective: The aim of this study was to describe a novel approach for the union of osteoporotic vertebral compression fractures with minimally invasive open reduction and internal fixation. Patients and Methods: Seven consecutive patients with intractable back pain without neurological deficits due to osteoporotic vertebral compression fractures were treated using minimally invasive fixation with intra-vertebral expandable pillars and artificial bone substitute. The clinical symptoms and image findings were recorded. Results: All of the patients reported relief of back pain, and the height of the vertebral bodies was well restored. X-ray findings obtained 2 to 4 years after the procedures showed fracture healing and favorable formation of the callus confirmed in the anterior longitudinal ligament. Conclusion: This mini-open procedure with intravertebral devices is an effective and reliable technique for osteoporotic vertebral compression fractures and may avoid complications related to traditional open spinal instrumentation procedures and augmentation with bone cement.

Experimental Demonstration of gm/IDBased Noise Analysis

Recent studies using BSIM3 models have suggested that noise depends on the transconductance-to-drain ratio gm/ID of a transistor. However, to the best of our knowledge, no experimental result demonstrating gm/ID dependent noise previously observed in simulation is available in the literature. This paper examines the underlying principles that make it possible to analyze noise using gm/ID based noise analysis. Qualitative discussion of normalized noise is presented along with experimental results from a 130 nm CMOS process. A close examination of the experimental results reveals that the device noise is width independent from 1 Hz to 10 kHz. Moreover, noise increases as gm/ID is reduced. The experiment observation that noise is width independent makes it possible for circuit designers to generate normalized parameters that are used to study noise intuitively and accurately.
