Strains from the Cryptococcus gattii species complex(CGSC)have caused the Pacific Northwest cryptococcosis outbreak,the largest cluster of lifethreatening fungal infections in otherwise healthy human hosts known to *** this study,we utilized a pan-phenome-based method to assess the fitness outcomes of CGSC strains under 31 stress conditions,providing a comprehensive overview of 2,821 phenotype-strain associations within this pathogenic *** clustering analysis revealed a strong correlation between distinct types of stress phenotypes in a subset of CGSC strains,suggesting that shared determinants coordinate their adaptations to various ***,a specific group of strains,including the outbreak isolates,exhibited a remarkable ability to adapt to all three of the most commonly used antifungal drugs for treating cryptococcosis(amphotericin B,5-fluorocytosine,and fluconazole).By integrating pan-genomic and pan-transcriptomic analyses,we identified previously unrecognized genes that play crucial roles in conferring multidrug resistance in an outbreak strain with high multidrug *** these genes,we identified biomarkers that enable the accurate prediction of highly multidrug-adapted CGSC strains,achieving maximum accuracy and area under the curve(AUC)of 0.79 and 0.86,respectively,using machine learning ***,we developed a pan-omic approach to identify cryptococcal multidrug resistance determinants and predict highly multidrug-adapted CGSC strains that may pose significant clinical concern.

国家癌症中心发布的最新统计数据显示,肺癌仍是我国发病率和死亡率最高的恶性肿瘤。同时,肺癌的发病率及死亡率逐年攀升,对癌症患者及社会造成了严重的负担。非小细胞肺癌(non-small cell lung cancer,NSCLC)是肺癌的主要病理类型,约占全部肺癌的85%左右。晚期NSCLC患者的5年生存率较低,目前,化疗仍然是晚期肺癌治疗的基石。细胞毒类药物会损伤骨髓内的造血干细胞,导致骨髓抑制,使患者因不能耐受而中止治疗。针对骨髓抑制的传统治疗手段可能会引发其他不良反应,如骨髓造血储备功能耗竭等。2022年7月,曲拉西利获批用于预防小细胞肺癌化疗引起的骨髓抑制。曲拉西利能有效预防化疗引起的骨髓抑制,并且不会拮抗化疗的抗肿瘤作用。本文特报道1例曲拉西利联合免疫联合化疗一线治疗晚期NSCLC的病例,以期为临床实践提供进一步的参考。

目的对有关(制)何首乌中英文文献进行文献计量分析,探讨(制)何首乌的研究重点和趋势,为今后的科学研究提供依据。方法利用中国知网、Web of Science数据库检索(制)何首乌的有关文献,并基于CiteSpace 6.1.R6软件利用文献计量学方法,对文章的发表年份、作者、作者国家、关键词等内容进行可视化分析。结果本研究总共纳入中文文献1131篇、英文文献386篇,共计1517篇。近30年以来,在该领域的中文文献发文总量先上升后下滑,英文文献的发文总量则呈现波动性增长态势。发文量较高的作者为肖小河和王伽伯及其团队,该团队主要从事何首乌肝损伤机制与炮制减毒研究以及临床中药学与安全用药研究。关键词分析表明,本研究领域的研究热点和发展方向为(制)何首乌的效毒同源成分以及肝毒性机制及早期预警监测的研究。结论(制)何首乌效毒同源化学成分的研究一直很活跃,对于效毒同源成分转换关系与机制研究以及肝毒性机制的研究将是该领域的研究重点与发展趋势。

The cytochrome P4501A1(CYP1A1)inhibitor bergamottin enhances host tolerance to multidrug-resistant Vibrio vulnificus infection

Purpose:Vibrio vulnificus (V. Vulnificus) infection is characterized by rapid onset, aggressive progression, and challenging treatment. Bacterial resistance poses a significant challenge for clinical anti-infection treatment and is thus the subject of research. Enhancing host infection tolerance represents a novel infection prevention strategy to improve patient survival. Our team initially identified cytochrome P4501A1 (CYP1A1) as an important target owing to its negative modulation of the body''s infection tolerance. This study explored the superior effects of the CYP1A1 inhibitor bergamottin compared to antibiotic combination therapy on the survival of mice infected with multidrug-resistantV. Vulnificus and the protection of their vital ***:An increasing concentration gradient method was used to induce multidrug-resistantV. Vulnificus development. We established a lethal infection model in C57BL/6J male mice and evaluated the effect of bergamottin on mouse survival. A mild infection model was established in C57BL/6J male mice, and the serum levels of creatinine, urea nitrogen, aspartate aminotransferase, and alanine aminotransferase were determined using enzyme-linked immunosorbent assay to evaluate the effect of bergamottin on liver and kidney function. The morphological changes induced in the presence of bergamottin in mouse organs were evaluated by hematoxylin and eosin staining of liver and kidney tissues. The bacterial growth curve and organ load determination were used to evaluate whether bergamottin has a direct antibacterial effect on multidrug-resistantV. Vulnificus. quantification of inflammatory factors in serum by enzyme-linked immunosorbent assay and the expression levels of inflammatory factors in liver and kidney tissues by real-time quantitative polymerase chain reaction were performed to evaluate the effect of bergamottin on inflammatory factor levels. Expression changes of activin A in the development of hepatic fibrosis

AIM: To examine the expression of activin A, a member of the transforming growth factor (TGFbeta) superfamily, recently has been reported to be overexpressed in liver cirrhosis, in the course of carbon tetrachloride-induced rat hepatic fibrosis. METHODS: Hepatic fibrosis was induced in rats by subcutaneous injections of 40% carbon tetrachloride oily solution for a period of 1 to 7 weeks. At the end of 1, 2, 3, 4, 5, 6 and 7 weeks after carbon tetrachloride injections, the rats were killed in group (6-10 rats each time) for study. The activin A messenger RNA expression and its protein localization were assessed by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) and immunohistochemistry. RESULTS: The normal rat liver expressed activin A mRNA and protein, and its expression was transiently decreased and became undetectable after carbon tetrachloride injections for 2 or 3 weeks and then increased gradually. After injection of carbon tetrachloride for 6 and 7 weeks, activin A mRNA and protein expressions were significantly enhanced in rat liver. Compared with that of the normal rat liver. Activin A mRNA expression levels in rats receiving carbon tetrachloride injections for 6 and 7 weeks were 1.6 and 2.2 times that of those in normal rat liver respectively (0.456 +/- 0.094 vs 0.2860.0670, P20 +/- 0.134 vs 0.286 +/- 0670, Pliver, and its expression was decreased in rats receiving carbon tetrachloride for 2 or 3 weeks. Compared with normal liver, activin A expression distribution mode changed in fibrotic liver, being increased significantly in hepatocytes around fibrotic areas. CONCLUSION: Activin A expression was increased in late stage of hepatic fibrosis, and this may be involved in hepatic fibrosis formation in this period.

Low-Dose Ethanol Preconditioning Protects Against Oxygen-Glucose deprivation/Reoxygenation-Induced Neuronal Injury By Activating Large Conductance,Ca2+-Activated K+Channels In Vitro

Increasing evidence suggests that low to mod- erate ethanol ingestion protects against the deleterious effects of subsequent ischemia/reperfusion; however, the underlying mechanism has not been elucidated. In the present study, we showed that expression of the neuronal large-conductance, Ca2＋-activated K＋ channel （BKca） α- subunit was upregulated in cultured neurons exposed to oxygen-glucose deprivation/reoxygenation （OGD/R） compared with controls. Preconditioning with low-dose ethanol （10 mmol/L） increased cell survival rate in neurons subjected to OGD/R, attenuated the OGD/R-induced elevation of cytosolic Ca2＋ levels, and reduced the number of apoptotic neurons. Western blots revealed that ethanol preconditioning upregulated expression of the anti-apoptotic protein Bcl-2 and downregulated the pro-apoptotic protein Bax. The protective effect of ethanol precondi- tioning was antagonized by a BKα channel inhibitor, paxilline. Inside-out patches in primary neurons also demonstrated the direct activation of the BKCa channel by 10 mmol/L ethanol. The above results indicated that low- dose ethanol preconditioning exerts its neuroprotective effects by attenuating the elevation of cytosolic Ca2＋ and preventing neuronal apoptosis, and this is mediated by BKca channel activation.
