Connexin subunits are proteins that form gap junction channels, and play an important role in communication between adjacent cells. This review article discusses the function of connexins/hemichannels/gap junctions under physiological conditions, and summarizes the findings re-garding the role of connexins/hemichannels/gap junctions in the physiological and pathological mechanisms underlying central nervous system diseases such as brain ischemia, traumatic brain and spinal cord injury, epilepsy, brain and spinal cord tumor, migraine, neuroautoimmune disease, Alzheimer’s disease, Parkinson’s disease, X-linked Charcot-Marie-Tooth disease, Peli-zaeus-Merzbacher-like disease, spastic paraplegia and maxillofacial dysplasia. Connexins are considered to be a potential novel target for protecting the central nervous system.

Most of the phosphorite deposits in the world contain isomorphism rare earths （RE） which are considerably difficult to be leached into solution in the wet phos- phoric acid process. In this work, a systematic study of leaching RE using sulfuric acid, phosphoric acid, mixed acid and two-step leaching of phosphoric acid and sulfuric acid was performed. The aims are to illuminate the main factors that inhibit RE leaching and to provide insights into the further enrichment of RE in the wet phosphoric acid solution. The results indicate that H2SO4 is not an effective acid for leaching isomorphism RE from phosphorite ore. The low RE leaching efficiency attributes to the RE cocrystallized and encapsulated by phosphogypsum （PG） as well as the precipitation of RE by RE sulfates or phosphates. High concentration of H3PO4 can enhance the dissolution and diffusion of RE ions. Hence, the optimized leaching mode of improving RE leaching efficiency is to adequately dissolve phosphorite ores in high concentration of H3PO4 solution and then add H2SO4 to crystallize PG. The effect of corystallization or encapsulation of PG on RE can be decreased due to the crystallizing mode of PG in the bulk solution instead of on the interface of solid reactants. RE leaching efficiency can be high up to 65% by the optimized leaching mode.

We analyzed 20 chemosensory protein （CSP） genes of the silkworm Bombyx mori. We found a high number of retrotransposons inserted in introns. We then analyzed expression of the 20 BrnorCSP genes across tissues using quantitative real-time polymerase chain reaction （PCR）. Relatively low expression levels of BmorCSPs were found in the gut and fat body tissues. We thus tested the effects of endectocyte insecticide abamectin （B 1 a and Blb avermectins） on BmorCSP gene expression. Quantitative real-time PCR experi- ments showed that a single brief exposure to insecticide abamectin increased dramatically CSP expression not only in the antennae but in most tissues, including gut and fat body. Furthermore, our study showed coordinate expression of CSPs and metabolic cytochrome P450 enzymes in a tissue-dependent manner in response to the insecticide. The function of CSPs remains unknown. Based on our results, we suggest a role in detecting xenobiotics that are then detoxified by cytochrome P450 anti-xenobiotic enzymes.

Objective To review the mechanisms and current clinical application of pharmacological interventions for phantom limb pain. Data sources Both Chinese and English language literatures were searched using MEDLINE （1982-2011）, Pubmed （1982-2011） and the Index of Chinese Language Literature （1982-2011 ）. Study selection Data from published articles about pharmacological management of phantom limb pain in recent domestic and foreign literature were selected. Data extraction Data were mainly extracted from 96 articles which are listed in the reference section of this review. Results By reviewing the mechanisms and current clinical application of pharmacological interventions for phantom limb pain, including anticonvulsants, antidepressants, local anaesthetics, N-methyI-D-aspartate receptor antagonists, non-steroidal anti-inflammatory drugs, tramadol, opioids, calcitonin, capsaicin, beta-adrenergic blockers, clonidine, muscle relaxants, and emerging drugs, we examined the efficacy and safety of these medications, outlined the limitations and future directions. Conclusions Although there is lack of evidence-based consensus guidelines for the pharmacological management of phantom limb pain, we recommend tricyclic antidepressants, gabapentin, tramadol, opioids, local anaesthetics and N-methyI-D-aspartate receptor antagonists as the rational options for the treatment of phantom limb pain.

Tumor suppressor p53-mediated cell cycle arrest and DNA damage repair may exert cytoprotective effects against cancer therapies, including WEE1 inhibition. Considering that p53 activation can also lead to multiple types of cell death, the role of this tumor suppressor in WEE1 inhibitor-based therapies remains disputed. In this study, we reported that nucleolar stress-mediated p53 activation enhanced the WEE1 inhibitor AZD1775-induced ferroptosis to suppress lung cancer growth. Our findings showed that AZD1775 promoted ferroptosis by blocking cystine uptake, an action similar to that of Erastin. Meanwhile, inhibition of WEE1 by the WEE1 inhibitors or si RNAs induced compensatory upregulation of SLC7A11, which conferred resistance to ferroptosis. Mechanistically, AZD1775 prevented the enrichment of H3K9me3, a histone marker of transcriptional repression, on the SLC7A11 promoter by repressing the expression of the histone methyltransferase SETDB1, thereby enhancing NRF2-mediated SLC7A11 transcription. This finding was also validated using the H3K9me3 inhibitor BRD4770. Remarkably, we found that the nucleolar stress-inducing agent Actinomycin D(Act. D) inhibited SLC7A11 expression by activating p53, thus augmenting AZD1775-induced ferroptosis. Moreover, the combination of AZD1775 and Act. D synergistically suppressed wild-type p53-harboring lung cancer cell growth both in vitro and in vivo. Altogether, our study demonstrates that AZD1775 promotes ferroptosis by targeting cystine uptake but also mediates the adaptive activation of SLC7A11 through the WEE1-SETDB1 cascade and NRF2-induced transcription, and inhibition of SLC7A11 by Act. D boosts the anti-tumor efficacy of AZD1775 by enhancing ferroptosis in cancers with wild-type p53.

Objective:Inquiry occupies an important position in the Traditional Chinese Medicine(TCM)diagnostic *** the characteristics of inquiry symptoms and the correlation with TCM syndromes can determine the important guiding significance to build recognition model and diagnostic criteria of ***:In this study,we collected 3707 valid cases,analyzed the inquiry symptoms of patients with cardiovascular disease,then to explore the distribution of inquiry information and the correlation with TCM syndromes based on the methods,such as frequency analysis,expert score,and correlation ***:We found the symptoms of frequency analysis and expert symptom score all in the top 50,add up to *** to the results by correlation analysis,the correlation coefficient between each symptom and each syndrome are sorted from high to low,the same with the experts score to each inquiry symptom basing on its importance of in diagnosing each ***:Inquiry occupies an important effect in differentiation *** research could provide certain objective basis for establishment of TCM syndromes diagnostic criteria of cardiovascular disease,thus provide some ideas and methods for the research of establishing TCM syndrome identification model,then to provide a reference for the syndrome standardization and objectification.

Selective modification of peptides has always been a challenging issue. The selective alkylation of peptides can alter the biological activity and physical properties of molecules, and it has gained significant research interest. We herein disclose an intriguing gold-catalyzed alkylation of peptides using bench-stable unactivated alkyl bromides under photoredox catalysis. A wide range of structurally diverse primary, secondary and tertiary alkyl bromides serve as effective coupling partners to precisely connect with the α-C(sp3)–H in glycine residue of peptides. This protocol demonstrates an exceptionally broad scope for alkyl halides and excellent tolerance for a wide range of useful functional groups. A series of relay transformations to construct cyclic peptides, to proceed click reaction and to realize peptide fluorescence labeling further enhance its synthetic practicality. In addition, mechanistic studies and density functional theory(DFT) calculations reveal an inner-sphere and subsequent outersphere single electron transfer(SET) mechanism.
