Identification of an immune-related gene signature for predicting prognosis and immunotherapy efficacy in liver cancer via cell-cell communication

BACKGROUND Liver cancer is one of the deadliest malignant tumors worldwide.Immunotherapy has provided hope to patients with advanced liver cancer,but only a small fraction of patients benefit from this treatment due to individual differences.Identifying immune-related gene signatures in liver cancer patients not only aids physicians in cancer diagnosis but also offers personalized treatment strategies,thereby improving patient survival rates.Although several methods have been developed to predict the prognosis and immunotherapeutic efficacy in patients with liver cancer,the impact of cell-cell interactions in the tumor microenvir-onment has not been adequately considered.AIM To identify immune-related gene signals for predicting liver cancer prognosis and immunotherapy efficacy.METHODS Cell grouping and cell-cell communication analysis were performed on single-cell RNA-sequencing data to identify highly active cell groups in immune-related pathways.Highly active immune cells were identified by intersecting the highly active cell groups with B cells and T cells.The significantly differentially expressed genes between highly active immune cells and other cells were subsequently selected as features,and a least absolute shrinkage and selection operator(LASSO)regression model was constructed to screen for diagnostic-related features.Fourteen genes that were selected more than 5 times in 10 LASSO regression experiments were included in a multivariable Cox regression model.Finally,3 genes(stathmin 1,cofilin 1,and C-C chemokine ligand 5)significantly associated with survival were identified and used to construct an immune-related gene signature.RESULTS The immune-related gene signature composed of stathmin 1,cofilin 1,and C-C chemokine ligand 5 was identified through cell-cell communication.The effectiveness of the identified gene signature was validated based on experi-mental results of predictive immunotherapy response,tumor mutation burden analysis,immune cell infiltration analysis,survival analysis,and expression an

BACKGROUND Gender consciousness directly affects the development of gender identity,which is a continuous and lifelong process.Meanwhile,hospitalization is a part of many children's lives and has an impact on their gender development.AIM To investigate the current situation of gender identity in lower primary school children by conducting a survey of 202 hospitalized children in the lower grades and to provide a theoretical basis and foundation for the cultivation of gender identity and medical treatment of children based on the results.This study aims to inspire clinical medical staff to scientifically and reasonably arrange hospital wards for lower primary school children and pay attention to gender protection during the medical treatment process and to help children shape a unified and clear gender identity,which will enable them to better integrate into society and promote their personality development.METHODS The gender consciousness scale for elementary and middle school students was RESULTS Gender identity was already present in lower primary school children.The children's gender roles and gender equality consciousness were strong,exceeding the critical value,but their gender characteristics,gender identity,and gender ideal consciousness were weak.Children aged 6 had the weakest gender identity,and girls had significantly stronger gender identity than boys.CONCLUSION Gender identity is already present in lower primary school children,providing a basis and inspiration for the cultivation of gender identity and medical treatment of lower primary school children.Clinical medical staff should be aware of and understand these results and should scientifically and reasonably arrange hospital wards for lower primary school children.

Data-driven mapping-relationship mining between hardness and mechanical properties of dual-phase titanium alloys via random forest and statistical analysis

In order to accelerate the research on the property optimization of titanium alloy based on high-throughput methods,it is necessary to reveal the relationship between hardness and other mechanical properties which is still unclear.In this work,taking Ti20C alloy as research object,almost all the microstructure of dual-phase titanium alloys were covered by traversing over 100 heat treatment schemes.Then,massive experiments including microstructure characterization and performance test were conducted,obtaining 51,590 pieces of microstructure data and 3591 pieces of mechanical property data.Subsequently,based on large-scale data-driven technology,the quantitative mapping relationship between hardness and other mechanical properties was deeply discussed.The results of random forest models showed that the correlation between hardness(H)and Charpy impact energy(A_(k))(or elongation,A)was hardly dependent on the microstructure types,while the relationship between H and tensile strength(R_(m))(or yield strength,R_(p0.2))was highly dependent on microstructure types.Specifically,combined with statistical analysis,it was found that the relationship between H and Ak(or A)were negatively linear.Interestingly,the relationship between H and strength was positively linear for equiaxed microstructure,and strength was linked to d^(−1/2)(d,equivalent circle diameter)ofα-grains in the form of classical Hall–Petch formula;but for other microstructures,the relationships were quadratic.Furthermore,the above rules were nearly the same in the rolling direction and transverse direction.Finally,a"four-quadrant partition map"between H and R_(p0.2)/R_(m) was established as a versatile material-screening tool,which can provide guidance for on-demand selection of titanium alloys.

Predicting the prognosis of primary orbital lymphoma by clinical characteristics and imaging features

AIM: To analyze the 5-year disease-free survival(DFS) of primary orbital lymphoma(POL) by clinical characteristics and imaging features.METHODS: A total of 72 patients, 43 males and 29 females, with histologically confirmed POL, were retrospectively recruited between January 2012 and May 2017. The information on clinical characteristics, imaging features, and 5-year DFS was obtained. Univariate and multivariate forward logistic regression analyses were used to identify the variables significantly associated with 5-year DFS. Kaplan-meier was applied for survival analysis. RESULTS: Univariate analysis revealed that uni-or bilateral orbital involvement, single or multiple lesions, treatment methods, and contrast enhancement pattern on images were significant for 5-year DFS(P=0.022, 0.042, <0.001, and 0.028, respectively), while in multivariate logistic regression analysis, only uni-or bilateral orbital involvement, treatment methods and contrast enhancement pattern on images were significant(r=0.453, 0.897, and 0.556, P=0.038, <0.001 and 0.022, respectively). The survival curves for DFS were obtained. CONCLUSION: The majority of POL are B-cell lymphomas. Unilateral orbital involvement, homogeneous contrast enhancement on images, and the appropriate treatment schemes result to be significant factors for a good prognosis for POL.

Time trends in myopia and high myopia prevalence in young university adults in China

AIM:To investigate time trends in myopia and high myopia prevalence over 6y among young university adults in China.METHODS:This is a 6-year series cross-sectional study from 2016 to 2021.Totally 4910 freshmen were enrolled and completed a questionnaire concerning age,gender,and disease history.Students with eye diseases were excluded after a detailed eye examination.The refractive status was measured by non-cycloplegic objective refraction and ocular parameters were measured by Lenstar 900.The examination followed the same protocol each year.Trends over time in myopia and high myopia prevalence,as well as ocular biometry parameters,were analyzed.RESULTS:From 2016 to 2021,the axial length(AL)and corneal radius(CR)increased significantly(P=0.002 for AL;P=0.04 for CR).However,the spherical equivalent(SE)and the ratio of axial length to the corneal radius(AL/CR)did not change significantly(P=0.59 for SE;P=0.24 for AL/CR).The frequency of AL≥26.0 mm increased from 26.6%in 2016 to 29.3%in 2021(P=0.05 for trend).The prevalence of myopia and high myopia did not change significantly in our study(P≥0.18).Compared to a similar cross-sectional study conducted 10 years ago,the prevalence of myopia decreased significantly(94.9%vs 91.8%,P<0.001).Whereas the prevalence of high myopia increased largely(18.12%vs 27.6%,P<0.001).CONCLUSION:The prevalence of high myopia increases in young university adults during 10y period.Myopia control should begin earlier in childhood.However,these interventions are still needed for high myopia even in young adulthood.

Lymphocytes≥2.9(10^(9)/L)in Newly Diagnosed Diabetes Are A Predictor of Future CVD Events

Objective Atherosclerosis is considered a chronic inflammatory condition.The immune system is a key mediator in the initiation and progression of atherosclerosis.In a previous study,we found that the immune system was activated in diabetes and that total white blood cell(WBC)counts were elevated significantly in diabetic patients.To investigate whether WBC subtype counts in newly diagnosed diabetes are risk factors for future cardiovascular disease(CVD)events,we conducted a prospective population-based cohort study.Methods A total of 1498 newly diagnosed diabetic patients aged 40 to 70 years old were followed up for three years.Participants with previous CVD history and abnormal WBC counts were excluded.CVD events were recorded during follow-up.Results We found that the baseline lymphocyte counts were independently associated with cardiovascular events during follow-up,with the Exp(β)(95%CI)at 1.749(1.084-2.821).Lymphocyte count≥2.9(10^(9)/L)was significantly associated with the development of CVD(HR,2.29;95%CI,1.12-4.67).The corresponding incidence of CVD per 1000 person-year for the lymphocyte count≤2.8(10^(9)/L)and lymphocyte count≥2.9(10^(9)/L)groups were 11.26 and 26.38,respectively.Conclusion We concluded that even in a normal range,higher lymphocyte levels may result in a significantly higher CVD risk among diabetic patients.Lymphocyte count≥2.9(10^(9)/L)is an independent predictor of developing future CVD events.

BACKGROUND Percutaneous drainage(PCD)and endoscopic approaches have largely replaced surgical drainage as the initial approach for(peri)pancreatic fluid collections(PFC)s,while complications associated with endoscopic stent implantation are common.AIM To introduce a novel endoscopic therapy named endoscopic transgastric fenestration(ETGF),which involves resection of tissue by endoscopic accessory between gastric and PFCs without stent implantation,and to evaluate its efficacy and safety compared with PCD for the management of PFCs adjacent to the gastric wall.METHODS Patients diagnosed with PFCs adjacent to the gastric wall and who subsequently received ETGF or PCD were restrospectively enrolled.Indications for intervention were consistent with related guidelines.We analyzed patients baseline characteristics,technical and clinical success rate,recurrence and reintervention rate,procedure-related complications and adverse events.RESULTS Seventy-two eligible patients were retrospectively identified(ETGF=34,PCD=38)from October 2017 to May 2021.Patients in the ETGF group had a significantly higher clinical success rate than those in the PCD group(97.1 vs 76.3%,P=0.01).There were no statistically significant differences regarding recurrence,reintervention and incidence of complication between the two groups.While long-term catheter drainage was very common in the PCD group.CONCLUSION Compared with PCD,ETGF has a higher clinical success rate in the management of PFCs adjacent to the gastric wall.ETGF is an alternative effective strategy for the treatment of PFCs adjacent to the gastric wall.

Hydrothermal calcification surface modification of biomedical tantalum

Tantalum(Ta) as a new type of medical metal is now utilized in several orthopedic applications due to its excellent fracture toughness and workability,high corrosion resistance,good biocompatibility and X-ray visibility,but it lacks bioactivity.Therefore,pure Ta was hydrothermally calcified in CaHPO4 solution at 120-200℃ here for bioactive surface modification.Scanning electron microscopy(SEM),energy-dispersive X-ray spectroscopy(EDX),X-ray photoelectron spectroscopy(XPS),X-ray diffraction(XRD) and Raman spectroscopy analyses show that thin tantalum oxide film with a few calcium phosphate precipitates is formed at the surface after the treatment at 200℃.The hydrothermally treated sample has good hydrophilicity and corrosion resistance evidenced by the water contact angle measurement and the potentiodynamic polarization test,respectively,and it can induce the formation of apatite layer after soaked in the simulated body fluid for 1 week.The present method would be applicable for bioactive surface modification of tantalum implants with irregular shapes and even porous structure.

hTFtarget:A Comprehensive Database for Regulations of Human Transcription Factors and Their Targets

Transcription factors(TFs)as key regulators play crucial roles in biological processes.The identification of TF-target regulatory relationships is a key step for revealing functions of TFs and their regulations on gene expression.The accumulated data of chromatin immunoprecipitation sequencing(ChIP-seq)provide great opportunities to discover the TF-target regulations across different conditions.In this study,we constructed a database named hTFtarget,which integrated huge human TF target resources(7190 ChIP-seq samples of 659 TFs and high-confidence binding sites of 699 TFs)and epigenetic modification information to predict accurate TF-target regulations.hTFtarget offers the following functions for users to explore TF-target regulations:(1)browse or search general targets of a query TF across datasets;(2)browse TF-target regulations for a query TF in a specific dataset or tissue;(3)search potential TFs for a given target gene or noncoding RNA;(4)investigate co-association between TFs in cell lines;(5)explore potential coregulations for given target genes or TFs;(6)predict candidate TF binding sites on given DNA sequences;(7)visualize ChIP-seq peaks for different TFs and conditions in a genome browser.hTFtarget provides a comprehensive,reliable and user-friendly resource for exploring human TF-target regulations,which will be very useful for a wide range of users in the TF and gene expression regulatiol community.hTFtarget is available at bttp://***/hTFtarget.

Primary central nervous system lymphomas （PCNSLs） are rare non-Hodgkin tumors defined as lymphomas of the central nervous system （CNS） without primary tumor elsewhere. It was reported that PCNSLs represented only 3-7% of primary brain tumors and 1-5% of all lymphomas. Most of PCNSLs are B-cell lymphomas, while T-cell PCNSL （T-PCNSL） is extremely rare, the majority of the reported T-PCNSL cases are clinically sporadic that focus on the treatment, and its imaging features have rarely been described. Herein, we report a case of T-PCNSL and comprehensively summarize its magnetic resonance imaging （MRI） characteristics. It is the first time in China to locus on the MRI diagnosis of T-PCNSL.
