HA/TCP and HA rods （φ5 mm×10 mm） were made for implantation in New Zealand white rabbit with different condition. Sixty three rabbit were divided into three groups： group 1 （n=18）, group 2 （n=27） and group 3 （n=18）. In group 1, 10 mm radius was defected, and one HA/TCP rod was implanted in the muscle a distant away from the bone defect area. In group 2, also, 10 mm radius was defected, one HA rod was implanted in the muscle a distant away from the bone defect area. In group 3, two HA/TCP rods were implanted in the dorsal muscle of the rabbit with bone intact. Histological observation showed that in group 1, some new bone was found only two months after implantation （n=2）, and obvious immature woven bone could be observed in these bioceramics from the 3rd month on. However, in group 3, bone began to be found 6 months after implantation （n=2）. In group 2, we could not find any bone tissue up to 9 month’s observation. These results suggest that, first, the bone defect model could significantly accelerate bone formation at non-osseous sites in rabbits; second,. HA/TCP bioceramics were confirmed with osteoinductive property while HA bioceramics without osteoinductive property nearly. Thus, bone defect might be a good animal model for further researches for osteoinductive bioceramics.

The Effect of Foods Containing Refined Konjac Meal on Human Lipid Metabolism

A total of 110 elderly people with hyperlipidemia were randomly assigned to one of two groups. The experimental group consumed an ordinary diet plus foods containing refined Konjac meal, and the control group consumed only the ordinary diet. The experiment was carried out for 45 days. The results indicate that for the experimental group blood levels of triglyeendes (TG), total cholesterol (TC), and low-density lipoprotein cholesterol (LDL-C) were significantly lowered (P < 0.01) at the end of the trial, whereas high-density lipoprotein cholesterol (HDL-C) and apoprotein (Al) levels were significantly elevated (P<0.01). In contrast, for the control group, the changes in the above parameters were insignificant. The differences in TC. TG, LDL-C, and HDL-C levels between the two groups were statistically significant. The effects of refined Konjac meal on lipid levels in the blood were somewhat different between patients with hyperlipidemia and subjects with risk critical values only. For the former, TG and TC were decreased by 83.8±133.5 mg/dl, and 42.4±23.4 mg/dl. respectively: but for the latter, they are decreased only by -1.1±23.1 mg/dl and 8.3± 18.2 mg/dl, respectively; the difference mentioned above is Statistically significant (P<0.01). 1990 Academic Press. AIM: Hepatic stellate cell (HSC) plays a pivotal role inliver fibrosis and is considered as the therapeutic targetfor the treatment of hepatic fibrosis. Tyrosine proteinkinase plays an important role in the proliferation,activation of HSC. The purpose of the study is toinvestigate the effects of the tyrosine protein kinaseinhibitor genistein onthe proliferation and activationof cultured rat HscMETHODS: Rat HSC were isolated from Wistar rats by insitu perfusion of collagenase and pronase and single-stepdensity Nycodenz gradient. Culture-activated HSC wereserum-starved and incubated with 10-3 to 10-5 mol/Lconcentration of genistein for 24, 48 or 72 h. In PDGF-induced HSC proliferation, HSC were stimulatel with L0μg@L-1PDGF-BB for 15 min, and then treaeed with genisteinfor the same time. Cell proliferation was measured byMTr assay and based on flow cytometric analysis of cellcycle. The a-smooth muscle actin (α-SMA) expression inHSC was studied with confocal laser microscopy and flowcytometry. c-fos c-jun and cyclin D1 expression in HSCwas also detected by flow ***: Genistein inhibited basal and PDGF-inducedproliferation of HSC at the concentration of 10-8 to 10-5mol/L, and treatment with 10-7 mol/L concentration ofgenistein for 48 h inhibited the HSC proliferationsignificantly (the inhibition rate was 70.3 %, P＜0.05).Immunofluorescence detected by confocal lasermicroscopy and flow cytometry showed that treatmentwith 10-7 mol/L genistein for 48 h suppressed theexpression of α-SMA significantly in HSC (the specificfluorescence intensity were 60.2±21.5 vs 35.3±11.6and 12.8± 10.4 vs9.54±6.39, respectively, both P＜0.05).The intensity of c-fos, c-jun and cydin D1 expression ofHSCs treated with 10-7 mol/L genistein for 48 h wasalso significantly decreased compared with the ***: Genistein influences proliferation of HSC,suppresses the expression of α-SMA in HSC and tinhibits the intensity of c-fos, c-jun and cyclin D1expression of HSCs. AIM: To explore the role of focal adhesion kinase (FAI)in the apoptosis in culture-activated rat hepatic stellatecells (HSCs) using a specific anti-FAK ***: Rat HSCs were prepared from Wistar rats byin situ perfusion of collagenase and pronase and single-step density Nycodenze gradient. Culture-activatedHSCs were serum-starved and treated with the anti-FAK antibodies for 24, 48 or 72 h. The apoptosis of HSCwas detected by DNA-fragment assay, flow cytometryand caspase-3 activity determination. The expressionof tissue inhibitor of metalloproteinase-1 (TIMP-1)mRNA was assessed by reverse transcriptionpolymerase chain reaction (RT-PCR).RESULTS: The experiment showed that anti-FAKantibodies induced apoptosis of culture-activated ratHSCs. This phenomenon displayed the classical featuresof apoptotic cell death (DNA fragmentation, cell cycleanalysis) after treated with 30 mg@L-1 FAK antibody for72 h, and accompanied by a significant increase ofcespase-3 activity(1208±76) vs (309±28) nmol@min-1@ g-1, t=-208.5, P＜0.05. Meanwhile, treatment with theFAK antibody in HSCs could markedly decrease theTIMP-1 mRNA expression (0.07±0.01 vs 0.38±0.03, t=2.72, P＜0.05).CONCLUSION: FAK plays an important role in the survivalof HSCs and the specific anti-FAK antibody could inducethe apoptosis in rat HSCs.

BACKGROUND:While suppressor of cytokine signaling 3(SOCS3) plays a crucial role in suppressing dysplasia and tumorigenesis,it also offers a typical instance of DNA methylation in the regulation of gene transcription,since the promoter region of the SOCS3 gene is rich in CpG islands(CGIs).During liver regeneration initiated by partial hepatectomy,SOCS3 acts as a suppressor to balance the acute-phase response and terminate the *** study aimed to determine whether the variation of SOCS3 expression throughout liver regeneration is also regulated by its DNA ***:We established a 70% partial hepatectomy mouse model and the animals were sacrificed at indicated times to assess the SOCS3 *** performed bisulfite sequencing PCR and DNA sequencing to investigate the detailed cytosine methylation in the SOCS3 ***:Within the promoter sequence,58 CGIs were identified and 30 were found variously methylated before or after operation;however,methylation remained at a very low *** evidence indicated that the total methylation level or the methylation of any CpG site regularly changed throughout liver ***:DNA methylation or demethylation seems to be a relatively stable modification of cytosine,but not a dynamic and reversible process to regulate gene transcription in daily and acute pathophysiological events.

The effects of surface acidity-basicity and surface oxidation reduction property of Li-La-Mn/TiO2 (Ⅰ) and Li-La-Mn-W/TiO2 (Ⅱ) catalysts on oxidative coupling of methane were studied by CO2-temperature programmed desorption (CO2-TPD) and temperature programmed reduction temperature programmed oxidation (TPR-TPO). The results show that there exist strong basic sites on catalysts Ⅰ and Ⅱ, but the quantity of these sites on catalyst Ⅱ is more than that on catalyst Ⅰ. Besides, the strength of basics site on catalyst Ⅱ is stronger than that on catalyst Ⅰ. The surface of catalyst Ⅱ is easier to reduce and re-oxidize than that of catalyst Ⅰ, and the extent of reduction and reoxidation of catalyst Ⅱ is more intensive than catalyst Ⅰ,which results in a lowering of the reaction theperature and enhances the activity and C2 hydrocarbon yield as well as gas hourly space velocity (GHSV) Catalyst Ⅱ is excellent for the oxidative coupling of methane (OCM).

BACKGROUND: β-catenin has two distinct roles in E-cadherin mediated cell adhesion and carcinogenesis by activating the wnt/β-catenin signaling pathway. One occurs at the cell-adhesion site, where cadherins are linked to the actin-based cytoskeleton. The other takes place in the cytoplasm and nuclei and is thought to regulate cell transformation. We studied the role of β-catenin in hepatocarcinogenesis of rats. METHODS: Fresh liver specimens were obtained from normal rats. and atypical hyperplasia livers and hepatocarcinoma tissues from model rats. The changes of β-catenin in gene expression levels were detected by reverse transcriptase polymerase chain reaction (RT-PCR) in the different specimens separately. At the same time, their localization was observed immunohistochemically. RESULTS: In the normal liver specimens, β-catenin staining was seen in the cell membrane. In liver specimens of atypical byperplasia, β-catenin staining occurred in the cell cytoplasm of some cells as well as in the cell membrane of others. Immunohistochemically cancerous tissues showed the presence of β-catenin in the cytoplasm and nuclei. RTPCR revealed that the gene expression levels of β-catenin were same in all samples. CONCLUSIONS: The accumulation of β-catenin in the cytoplasm and/or nuclei frequently occurs in hepatocarcinogenesis of rats. It may be an early event in the development of hepatocarcinoma of rats.
