为了得到高灵敏度的生物传感器,设计了一种可以激发出四重法诺共振的非通金属-绝缘体-金属(Metal-Insulator-Metal,MIM)波导传感器.采用了上下对称结构,由一个竖放矩形谐振腔和两个横放矩形谐振腔与总线波导共同作用而得到了四重法诺共振.采用了有限时域差分方法计算了此结构的传输特性与Z方向的归一化磁场,并详细分析了其法诺共振的产生机理.通过计算结构几何参数调节后的透射光谱,从而得出了四处法诺共振峰透射率和共振位置可以通过改变结构几何参数而获得独立调节.此外还发现在结构中添加不同浓度的葡萄糖溶液可以实现高折射率传感灵敏度(S),获得最高灵敏度2736.98nm/RIU,可以看出此结构对于折射率变化有很高的灵敏度,这为设计和优化纳米级别的折射率传感器提供了新的思路,还可以应用于电路构建前进行性能测试.

Background:Simiaowan(SMW),a well-known traditional Chinese medicine,has been employed to treat hyperuricemia(HUA)and gout for ***,the bioactive components and underlying mechanisms have not been *** objective of this study was to identify the active components and potential mechanisms of SMW by integrating pharmacological experimentation,serum pharmacochemistry,network pharmacology and molecular ***:HUA rats modelling by high-fat/high-sugar diet and potassium oxonate/adenine oral administration were used to evaluate the pharmacodynamic effects of ***-Q-Exactive-MS/MS was employed to detect the bioactive components present in SMW-containing *** pharmacology and molecular docking were utilized to elucidate the potential targets and underlying ***:SMW effectively ameliorated HUA rats via the inhibition of uric acid(UA)production,promotion of UA excretion,improvement of lipid and glucose metabolic abnormalities,antioxidant,anti-inflammatory and anti-insulin resistance effects.A total of 73 compounds detected in SMW-containing serum were identified as potential active components,with alkaloids,flavonoids,organic acids,and terpenoids emerging as the primary active *** 203 corresponding targets were obtained as SMW anti-HUA/gout targets,which mainly participated in apoptosis,insulin resistance,TNF,PI3K-Akt,HIF-1,NF-κB,MAPK,IL-17 and TLR signaling *** docking indicated that active compounds(***,phellodendrine,quercetin,formononetin,ferulic acid)had superior binding abilities to the key targets(*** carrier family 22 member 12(URAT1),solute carrier family 22 member 6(OAT1),ATP-binding cassette sub-family G member 2(ABCG2),solute carrier family 2,facilitated glucose transporter member 9(GLUT9),xanthine dehydrogenase/oxidase(XDH),transcription factor p65(RELA),toll-like receptor 4(TLR4),prostaglandin G/H synthase 2(PTGS2),caspase-3(CASP3),insulin(INS)).Conclusion:S

中华民族是一个连续性的民族,历史虽有断裂但没有断层。在周人的眼中,夏商是言之凿凿的信史,汉代人又把信史推进到黄帝时代。商代已经被考古学所证明,黄帝、尧、舜、禹(夏)是铭刻在族群或者民族心里的文化记忆。春秋时代的孔子对于夏商文明的关注与探索为儒家三统理论奠定了思想与理论基础,经过思孟学派及其流裔的理论演化,三统论在汉代得到了确认与弘扬,且在阴阳学的赋义下成为汉代经学的核心理论。三统循环、周合五行之道成为汉儒笃信的历史发展理论。在三统论动态演绎过程中,形成了传统社会“皇权+孔子+经学”的话语语境以及话术,同时凝固成一种思维模式、叙事方式。然而,三统论并不是形而上的唯物史观,从历法发展乃至成熟的角度看,是汉儒在政治愿景驱动下依附于皇权而形成的一种理论假说。

菊粉(inulin,INU)作为一种植物多糖,因具有广泛的应用价值以及增强免疫、抗炎、抗肿瘤等生物学功能而备受关注,本研究旨在探究INU对巨噬细胞RAW 264.7免疫活性的影响。采用细胞增殖检测试剂盒(cell counting kit-8,CCK8)、中性红试验检测INU对巨噬细胞RAW 264.7增殖活性和吞噬活性的影响。采用格里斯试剂法和免疫酶联吸附试验(enzyme-linked immunosorbent assay,ELISA)检测细胞中一氧化氮(nitric oxide,NO)和细胞因子白细胞介素-1β(interleukin-1β,IL-1β)、白细胞介素-6(interleukin-6,IL-6)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)的释放量。实时荧光定量PCR(real-time fluorescence quantitative PCR,qPCR)检测细胞因子(IL-1β、IL-6、TNF-α)的基因表达量以及免疫蛋白印迹(Western blotting,WB)检测INU对巨噬细胞RAW 264.7中环氧化酶2(cyclooxygenase 2,COX2)、诱导型一氧化氮合酶(inducible nitric oxide synthase,iNOS)和丝裂原蛋白活化激酶(mitogen activated protein kinase,MAPK)相关通路关键蛋白表达的影响。实验结果表明,INU显著增强RAW 264.7细胞的增殖活性、吞噬活性并促进NO的分泌及IL-1β、IL-6、TNF-α的表达水平(P<0.01)。此外,INU还以浓度依赖的方式显著上调了COX2、iNOS及MAPK通路中磷酸化细胞外调节蛋白激酶(phosphorylated extracellular regulatory protein kinase,p-ERK)、磷酸化p38丝裂原活化蛋白激酶(phosphorylated p38 mitogen-activated protein kinase,p-p38)的蛋白表达(P<0.01)。综上所述,INU可通过ERK/p38MAPK通路调控巨噬细胞的免疫活性,具有良好的免疫调节能力。

目的探讨脓毒症患儿外周血可溶性凋亡相关因子配体(soluble factor-related apoptosis ligand,sFasL)、单核细胞微小RNA(micro-ribonucleic acid,miR)-147b表达情况及对预后的评估价值。方法前瞻性纳入脓毒症患儿124例(脓毒症组)、普通感染儿童60例(感染组)和健康体检儿童60例(健康对照组),分析脓毒症患儿预后不良的独立危险因素,评价血清sFasL、单核细胞miR-147b对脓毒症患儿预后不良的预测价值。结果健康对照组、感染组、脓毒症组血清sFasL水平、单核细胞miR-147b相对表达量逐渐升高(P<0.05)。多因素logistic回归分析显示,血清sFasL水平高、单核细胞miR-147b相对表达量高均与脓毒症患儿预后不良密切相关(P<0.05)。受试者操作特征曲线分析显示,血清sFasL水平、单核细胞miR-147b相对表达量联合预测脓毒症患儿预后的曲线下面积高于各指标单独预测(P<0.05)。结论脓毒症患儿外周血sFasL水平、单核细胞miR-147b相对表达量明显升高,两项指标联合应用在脓毒症患儿预后评估中具有较高的临床价值。
