Effects of aqueous extract from red Liriope platyphylla on phthalic-anhydride-induced atopic dermatitis in Interleukin-4/Luciferase/Consensus non-coding sequence-1 transgenic mice evaluated in terms of luciferase signal and general phenotype biomarkers

OBJECTIVE: To quantitatively evaluate the therapeutic effects of Red Liriope platyphylla(RLP) on atopic dermatitis(AD), alterations in the luciferase(Luc) signal and general phenotype biomarkers were compared in phthalic anhydride(PA) treated Interleukin-4(IL-4)/Luc/Consensus non-coding sequence-1(CNS-1) transgenic(Tg) mice following treatment with aqueous extract of RLP(AEt RLP) for4 ***: Alterations in AD phenotypes were measured in IL-4/Luc/CNS-1 Tg mice following treatment with AEt RLP using inflammation parameter analysis, bioluminescence imaging analysis, histological analysis, reverse transcription-polymerase chain reaction, enzyme linked immunosorbent assay and Western blot ***: RLP contained high concentrations of total phenolic compounds, total flavonoid compounds and 5-HNE related to AD therapy. The Luc signal was only detected in the abdominal region and the submandibular lymph node(SL), mesenteric lymph node(ML), thymus and pancreas of the PA treated group. This signal was significantly decreased by 28%-73% throughout the body and in the four organs in PA + AEt RLP treated group. Furthermore, the lymph node weight, immunoglobulin E concentration and dermal thickness were decreased by 37%-67% in the PA + AEt RLP treated ***: Our findings suggest that the therapeutic effect of AEt RLP on PA induced AD could be successfully quantified by comparison of Luc signals and AD phenotype markers in IL-4/Luc/CNS-1Tg mice, and that the Luc signal was as sensitive as the general AD phenotypes, enabling detection of effects without euthanasia.

Background:Immune checkpoint inhibitors(ICIs)are effective in a subset of patients with metastatic solid ***,the patients who would benefit most from ICIs in biliary tract cancer(BTC)are still *** and methods:We molecularly characterized tissues and blood from 32 patients with metastatic BTC treated with the ICI pembrolizumab as second-line ***:All patients had microsatellite stable(MSS)type *** of the 32 patients achieved partial response(PR),with an objective response rate(ORR)of 9.4%(95%confidence interval[CI],2.0–25.2)and nine showed stable disease(SD),exhibiting a disease control rate(DCR)of 37.5%(95%CI,21.1–56.3).For the 31 patients who had access to PD-1 ligand 1(PD-L1)combined positive score(CPS)testing(cut-off value≥1%),the ORR was not different between those who had PD-L1-positive(PD-L1+;1/11,9.1%)and PDL1-(2/20,10.0%)tumors(p=1.000).The tumor mutational burden(TMB)of PD-L1+BTC was comparable to that of PD-L1-BTC(p=0.630).TMB and any exonic somatic mutations were also not predictive of pembrolizumab *** analysis of blood and tumor samples demonstrated a relatively high natural killer(NK)cell proportion in the peripheral blood before pembrolizumab treatment in patients who achieved tumor ***,the tumors of these patients presented high enrichment scores for NK cells,antitumor cytokines,and Th1 signatures,and a low enrichment score for cancer-associated ***:This study shows the molecular characteristics associated with the efficacy of pembrolizumab in BTC of the MSS type.

Effect of CaF2 content in refining slag on formation behavior of spinel inclusion in Ni-Cr-V-alloyed special steel

The influence of CaF2 content on the formation behavior of spinel inclusions in the Ni-Cr-V-alloyed special steel was presented. The spinel was not formed at CaF2 content lower than 10 wt%. However, it was formed at early stage when the CaF2 content was greater than 30 wt%, followed by a modification to the aluminosilicate-type inclusions. Because the slag was saturated by MgO, the activity of MgO was unity irrespective of CaF2 content in the slag. Thus, Mg was transferred from slag to metal phase. Mg transferred to molten steel reacted with Al2O3-rich inclusions to form ***2O3 spinel. However, the spinel inclusion was modified to aluminosilicate-type inclusions by the reaction with Si and Ca transferred from slag to molten steel about 2-3 h later.

Deformation prediction of porcelain-enameled steels with strain history by press forming and high-temperature behavior of coating layer

Porcelain enamel coatings were widely applied for the protection of steel products because they offered high corrosion protection,resistance to heat and abrasion,high hardness,hygiene and ease of *** typical process to produce enameled steels is roughly divided into two stages:the first stage consists of a forming process to give the desired shape to a steel substrate,and the second stage consists of a firing process to bond enamel frits on the *** firing process requires a high temperature above 800 °C,which may lead to austenitic transformation and severe thermal deformation of the steel *** aim of this study is to develop a finite element analysis (FE analysis) technique to predict the mechanical and thermal deformations of the enameled steels during forming and any further enameling process,including *** FE analysis involves analyzing the strain history of the steel substrate,which comprises the stress and thickness distributions of the substrate and its deformed shape,and the high-temperature behavior of the enamel coating *** validity of the FE analysis is verified through the U-bending test and firing test with various numbers and positions of enamel coating layers on the *** results reveal that the FE analysis results agree well with the experimental results with 8% error.

Preoperative risk factors for early recurrence in patients with resectable pancreatic ductal adenocarcinoma after curative intent surgical resection

Background: Postoperative early recurrence(ER) in patients with pancreatic ductal adenocarcinoma(PDAC) is frequently encountered after curative intent surgery. Nonetheless, clinical significance and risk factors of ER after surgery for PDAC have not been extensively investigated. The aim of this study was to determine preoperative risk predictors for ER in patients with PDAC after upfront surgery. Methods: Eighty-one consecutive patients with PDAC who underwent curative intent surgical resection at Kangbuk Samsung Hospital between January 2004 and May 2015 were enrolled. ER was defined as tumor relapse within 6 months after surgery. Results: ER occurred in 26 patients(32.1%), whereas 49 patients(60.5%) had late recurrence( ≥ 6 months after surgery), and 6 patients had no recurrence(7.4%). Univariate analysis showed that C-reactive protein(CRP) > 3.0 mg/dL, modified Glasgow prognostic score(mGPS) = 2, decrease of total lymphocyte count by > 50% of baseline value in the preoperative period, prognostic nutritional index(PNI) 3.0 mg/dL, decrease of total lymphocyte count by > 50% of baseline value, and preoperative SUVmax were significant and independent contributors of ER in patients with resectable PDAC who underwent curative intent surgery. Conclusions: Postoperative ER for resectable PDAC was frequent with poor prognosis after curative intent upfront surgery. It is reasonable to suggest that there is a subgroup of resectable PDAC patients at highrisk of ER and neoadjuvant therapy should be considered in these patients in a clinical trial setting.

Changes in microorganisms and metabolites during long-term fermentation of commercially manufactured anchovy sauce

This study was performed to investigate the changes in microbial communities and metabolites during the long-term fermentation of commercially manufactured anchovy *** of commercial anchovy sauce were collected from large-scale fermentation tanks with fermentation periods of up to 7.87 *** complex bacterial community was simplified to two genera,Tetragenococcus and Halanaerobium,after approximately 0.55 years of *** genera,such as Saccharomyces,Cladosporium,Candida,and Aspergillus,were relatively dominant,no clear pattern was identified in fungal community *** longitudinal metabolite profile demonstrated that approximately half(55.8%)of the metabolites present in anchovy sauce were produced within a year of fermentation due to rapid *** the static microbial community,the contents of several metabolites including amino acids and biogenic amines changed continuously during the long-term fermentation of anchovy *** study provides novel insights into the changes in microbiota and metabolites in fish sauce produced without any starter inoculation.

Management of antiviral drug resistance in chronic hepatitis B

Rescue antiviral treatment for patients with resistance to preexisting nucleos(t)ide analogues remains a clinical challenge. The correct choice of a first-line treatment of high potency and with a high genetic barrier to achieve sustained long-term suppression of viral replication provides the best chance of preventing treatment failure and the emergence of drug resistance. The management of treatment failure and drug resistance requires a precise and accurate clinical and virologic monitoring. Combination treatment with antiviral drugs that belong to different groups is associated with a lower chance of developing resistance to rescue drugs. To guarantee better control of viral replication in patients with drug resistance, the addition of another drug without a cross resistance profile should be given as early as possible, preferably at the time when genotypic resistance emerges. Long-term surveillance for treatment efficacy and possible emergence of drug resistance should be continued to prevent the emergence of multidrug-resistant strains.

Distribution of Chancelloriids in a Middle Cambrian Carbonate Platform Deposit, Taebaek Group, Korea

The onset of the Cambrian witnessed the diversification of ＂small shelly fossils （SSF）＂, which affected carbonate depositional system. One of the problematic SSF, chancelloriids, are common components of the early to middle Cambrian carbonate and shale, and their contributions toward Cambrian carbonates are not yet fully understood. This study assesses distribution patterns of chancelloriid sclerites in the Cambrian Series 3 Daegi Formation based on microfacies analysis and discusses their sedimentologic implications. In the lower part of the formation, partially articulated chancelloriid sclerites occur mainly in bioclastic packstone and grainstone facies, with isolated sclerite rays in nodular packstone to grainstone facies. In the middle part of the formation, chancelloriid fragments occur only sporadically in bioclastic wackestone to packstone, bioclastic grainstone and oolitic packstone to grainstone facies, whereas boundstone facies are nearly devoid of their fragments. There are no chancelloriid fragments in the upper part of the formation, which consists of oolitic packstone to grainstone facies. Chancelloriids are interpreted to have primarily occupied platform margin shoal environments, shedding their sclerites to surrounding areas, and thus contributed as sediment producers. The distribution of Daegi chancelloriid sclerites is similar to other Cambrian examples, with the exception of common chancelloriids in Cambrian Series 2 reefs and their apparent near absence in the Daegi and other Cambrian Series 3 reefs. This disparity resulted from changes in the ecologic niche of chancelloriids after the end-Cambrian Series 2 reef crisis, coupled with an overall decline of chancelloriids in the middle Cambrian.
