乙型肝炎病毒(HBV)所致的肝细胞坏死,被认为是与人类白细胞抗原(hla)有关的mHC限制的T细胞为主的免疫应答的结果。本研究证实,转染HBV基因组或X基因后的人肝癌细胞系可诱导hlaⅡ(即hla DR)表达。核转录及RNA杂交分析提示表达X基因的细胞其hla DR mRNA水平增加,X蛋白增加该基因的转录。氯霉索乙酰基转移酶(CAT)基因分析进一步提示,X蛋白通过作用于hla DR基因上游的调节序列诱导hla DR表达。结果表明:X蛋白可能通过调节hla DR表达参与HBV感染的免疫发病机理。

Differential Effects of Alternative Glycoforms of IgG on Human monocytes and macrophages: Sialylated IgG Induces Novel Expression Signatures of Cell Surface markers, Cytokines, and Chemokines

The effector functions elicited by the fragment crystallizable (Fc) region of immunoglobulin G (IgG) antibodies are subject to variation by the presence of terminal sialic acid (Sia) residues at asparagine-297 (Asn-297). We have previously shown that the sialic acid-containing (Sia+) fraction of intravenous immune globulin (IVIG) influences cell surface marker expression and cytokine/ chemokine secretion during the differentiation and maturation of human dendritic cells (DC). The present study examined the effects of Sia+ IgG on human peripheral blood mononuclear cell (PBmC)-derived monocyte and macrophage surface marker expression and cytokine/chemokine secretion. Sia+ IgG induced increased expression of CD80 and dendritic cell immunoreceptor (DCIR) on monocytes, whereas the expression of hla-DR was decreased. In addition, the production of IL-6, TNFα, IL-1β, and CXCL1 by monocytes was profoundly increased by treatment with Sia+ IgG. Sia+ IgG also increased the expression of cell surface markers associated with macrophage polarization (e.g. CD40 and CD206) on monocytes. In macrophage-colony stimulating factor (mCSF) generated macrophages, Sia+ IgG induced increased production of numerous cytokines/ chemokines including IL-6, TNFα, CXCL1, and IL-10, and the expression of the macrophage surface marker CD163. Our data extended prior observations of Sia+ IgG on DC function and showed that Sia+ IgG was able to differentially modulate multiple pathways in monocytes and macrophages. Our data indicate that the Sia+ fraction of IVIG possesses the ability to influence inflammatory processes in multiple immune cell types and induces novel signatures in cell surface marker expression and cytokine/chemokine production.

Renal transplantation provides the best long-term treatment for chronic renal failure. Single-nucleotide polymorphisms (SNPs) play a major role in the understanding of the genetic basis of many complex human diseases. Also, the genetics of human phenotype variation could be understood by knowing the functions of these SNPs. It is still a major challenge to identify the functional SNPs in a disease-related gene. This work explored how SNPs mutations in hla-DRB1 gene could affect renal transplantation rejection. This study was carried out in Ahmed Gasim Hospital, Renal Dialysis Center during the period, from September 2012 to November 2013. Blood samples from five Sudanese patients (different families) with known renal transplantation rejection were collected before hemodialysis, furthermore one blood sample for control. DNA sequences results and detected SNPs were analyzed using bioinformatics tools (BLAST, SIFT, nsSNP Analyzer, PolyPhen, I-mutant, BioEdit, CPH, Chimera, Box shade and Project Hope). In addition, international databases were used for datasets [NCBI, Uniprot]. Results showed that, three SNPs were detected;two of three SNPs were predicted as tolerant or benign (rs1059575, novel) and one was deleterious (rs17885437). This study concluded that the identification of pathological SNPs could be an answer to unknown causes for a lot of organ transplantation rejection cases.

本研究以子宫颈角化上皮为靶细胞，用试管培养技术及非同位素细胞毒性试验，探讨了干扰素（ＩＦＮ－γ）对淋巴细胞介导的对宫颈角化上皮细胞毒作用的影响及细胞间吸附分子１（ＩＣＡＭ－１）与白细胞功能相关抗原１（ＬＦＡ－１）在细胞毒作用中的意义。结果显示ＣａＳｋｉ、ＳｉＨａ、ＨｅＬａ、Ｗ１２及ＮＣｘ均为ＮＫ抵抗、ＬＡＫ敏感细胞。ＩＦＮ－γ预处理靶细胞明显增加ＬＡＫ细胞的杀伤活性，但对ＮＫ活性影响不大；ＩＦＮ－γ预处理效应细胞可增强ＬＡＫ细胞对上述靶细胞的溶解作用，但并不改变ＮＫ对靶细胞的选择性；抗ＩＣＡＭ－１与ＬＦＡ－１单克隆抗体能有效地降低ＬＡＫ细胞对靶细胞的杀伤作用，而抗ＨＬＡＡＢＣ及ＨＬＡＤＲ则无此作用。提示ＩＦＮ－γ仅对ＬＡＫ细胞介导的对宫颈角化上皮的细胞毒作用有明显增强作用，而对ＮＫ细胞无明显影响；ＩＣＡＭ－１－ＬＦＡ－１通路为ＬＡＫ细胞结合并溶解靶细胞的主要通路，且这种细胞毒作用是非ＭＨＣ限制的。

Autoimmune Thyroid, Celiac and Addison’s Diseases Related to hla-DQ Types in Young Patients with Type 1 Diabetes in Belgium

Objective: To define the prevalence of auto antibodies (Ab) to different organs in young patients with type 1 diabetes (type 1Dm). methods: Ab to thyroid, celiac and adrenal disease was analyzed in 831 type 1 Dm patients. Results: Hundred twenty-three (14.8%) had positive thyroid Ab. The risk of developing thyroid Ab was increased in girls (HR 2.3;95% CI 1.6 - 3.2, p mysium Ab (3.1% in girls and 3.5% in boys, p = NS). Adrenal Ab was detected in 5 patients. The DQA1*0301-DQB1*0301 haplotype was more prevalent in patients with thyroid Ab (p = 0.0281);DQA1*301-DQB1*302 and DQA1*501-DQB1*201 in patients with endo- mysium Ab (p = 0.0251 and p mmunity and celiac disease. The DQA1*0301-DQB1*0301 haplotype seems to confer susceptibility to thyroid autoimmunity, DQA1*301-DQB1*302 and DQA1*501-DQB1*201 to celiac disease and DQA1*301-DQB1*302 to adrenal autoimmunity.
