Comparison of Chromatographic Performance for L-Phenylalanine-Derived Organic Phases on Silica by “Grafting from” and “Grafting to” Strategies

L-phenylalanine-derived polymerizable organogel, N’-octadecyl-Nα-(4-vinyl)-benzoyl-L-phenylalanineami- de (4) has been prepared according to the procedure described elsewhere. Compound 4 was successfully polymerized by surface initiated atom transfer radical polymerization (ATRP) from the initiator grafted silica particles (sil-poly4). It was also telomerized with 3-mercaptopropyltrimethoxysilane (MPS) and the telomer (T4) was grafted on to silica (sil-T4). TGA and elemental analysis measurement revealed that higher amount of polymer can graft by ATRP process than that of “grafting to” strategy. The results of 13C CP/MAS NMR measurement showed that the N-alkyl chain of the grafted polymers for both sil-poly4 and sil-T4 remained as less ordered gauche conformational form on silica surface and no inversion to trans form was occurred until temperature is increased up to 50?C. The retention of alkylbenzene samples showed that sil-poly4 prepared by “grafting from” method yielded extremely higher retention than conventional C18 phase however, sil-T4 prepared by conventional “grafting to” method showed lower retention than C18 phase. Aspects of molecular recognition were evaluated by the retention studies of a series of polycyclic aromatic hydrocarbons (PAHs) and aromatic positional isomers. We have observed sil-T4 yielded slightly higher selectivity for PAHs than sil-poly4 regardless the fact that it has low surface coverage and lower hydrophobic interactions. The enhanced selectivity observed for sil-T4 than C18 phases and sil-poly4 can be explained by the π - π interactions between the guest PAHs and carbonyl groups present in the polymer chain. In addition the aromatic moieties of compound 4 that aggregates through π - π interactions also contribute to the separation of PAHs for both sil-poly4 and sil-T4. Heat-Killed <i>Lactobacillus brevis</i>SBC8803 Induces Serotonin Release from Intestinal Cells

Previously, we reported that changes induced in autonomic neurotransmission in rats by Lactobacillus brevis SBC8803 may be mediated by serotonin 3 (5-HT3) receptors. In this study, we evaluated the effects of heat-killed L. brevis SBC8803 on serotonin (5-HT) releasing from intestinal cells. In the in vitro study, L. brevis SBC8803 stimulated 5-HT release from cultured rat endocrine RIN-14B cells (SBC8803 vs. sterile water;P in vivo study, 2 mg of heat-killed L. brevis SBC8803 was administered using a stomach sonde (feeding needle) to C57BL/6J mice. Analysis of plasma by ELISA showed gradually increase in 5-HT concentrations (0 min vs. 60 min;P ex vivo cultured intestinal loops composed of duodenum and part of the jejunum, from C3H/HeN and C57BL/6J male mice indicated that L. brevis SBC8803 effectively induced 5-HT release (SBC8803 vs. sterile water;P L. brevis SBC8803 may stimulate 5-HT release from mouse intestinal cells such as enterochromaffin cells.

Improvement in Surface Roughness and Hardness for Carbon Steel by Slide Burnishing Process

Slide burnishing process, which is a surface severe plastic deformation technique, offers an attractive post-machining alternative due to its chip-less and relatively simple operations. The purpose of the present work is to investigate effects of initial turned surface roughness on the burnished surface roughness and hardness in slide burnishing. The carbon steel samples those have different roughness surfaces being treated were prepared by turning by varying the feed. Slide burnishing was then carried out by a silicon nitride ceramic ball that was loaded and fed on the turned surface of a rotating specimen using a lathe machine. It was found that the turned surfaces were smoothed drastically by the burnishing process, and that the Ra and Rz values were reduced at most by a factor of 52 and 21, respectively. However, the smoothing effect of burnishing has limit, and the limited maximum height roughness (Rz*) for burnishing smoothing increased under a higher burnishing force and with a larger ball diameter. When the Rz values of initial turned surfaces were less than the Rz*, the roughness of the burnished surfaces did not depend on the roughness of the initial turned surface and the burnishing force. There was no significant difference in the burnished microstructure and hardness under a specific burnishing force among the initial turned surface roughness, while a higher burnishing force caused a greater increase in surface hardness.

Charge Transfer Mechanism and Spatial Density Correlation of Electronic States of Excited Zinc (3d<sup>9</sup>) Films

In material science, half filled 3d orbital of transition metals is essentially an important factor controlling characteristics of alloys and compounds. This paper presents a result of the challenge of excitation of inner-core electron system with long lifetime of zinc films. The advanced zinc films with excited inner-core electron, 3dn (n = 9, 8). We report experimental results of XPS measurements of 9 points in the sample along vertical direction, respectively. The most pronounced futures are existence of satellites, which are about 4 eV higher than the main lines. According to the charge transfer mechanism proposed by A. Kotani and K. Okada, it was clarified that the origins of these peaks are c3d9L for the main peak and c3d9 for the satellite, respectively. From the energy difference, δEB, and peak intensity ratio, I+/I-, between c3d9 and c3d10L, the energy for charge transfer, Δ, and mixing energy, T, were estimated. In the region where the intensity of c3d10L becomes large, Δ becomes small, 1.2 Δdc = 5.5 eV and Udd = 5.5 eV. In the analysis along vertical direction, intensity profile of Zn3d9 showed odd functional symmetry and that of Zn3d10L showed even functional symmetry. Only the intensity profile of C1s (288 eV) showed the same spatial correlation with Zn3d9. In our experiment, the sample also showed high mobility of the constituting elements. These suggest that charge conservation in excited zinc atom suggests combination between Zn3d9 and C2-.

Detection of fusion gene in cell-free DNA of a gastric synovial sarcoma

Synovial sarcoma(SS) is genetically characterized by chromosomal translocation, which generates SYT-SSX fusion transcripts. Although SS can occur in any body part, primary gastric SS is substantially rare. Here we describe a detection of the fusion gene sequence of gastric SS in plasma cell-free DNA(cf DNA). A gastric submucosal tumor was detected in the stomach of a 27-year-old woman and diagnosed as SS. Candidate intronic primers were designed to detect the intronic fusion breakpoint and this fusion sequence was confirmed in intron 10 of SYT and intron 5 of SSX2 by genomic polymerase chain reaction(PCR) and direct sequencing. A locked nucleic acid(LNA) probe specificto the fusion sequence was designed for detecting the fusion sequence in plasma and the fusion sequence was detected in preoperative plasma cfD NA, while not detected in postoperative plasma cfD NA. This technique will be useful for monitoring translocation-derived diseases such as SS.

Endoscopic submucosal dissection for early esophageal neoplasms using the stag beetle knife

AIM To determine short-and long-term outcomes of endoscopic submucosal dissection(ESD) using the stag beetle(SB) knife, a scissor-shaped *** Seventy consecutive patients with 96 early esophageal neoplasms, who underwent ESD using a SB knife at Kure Medical Center and Chugoku Cancer Center, Japan, between April 2010 and August 2016, were retrospectively evaluated. Clinicopathological characteristics of lesions and procedural adverse events were assessed. Therapeutic success was evaluated on the basis of en bloc, histologically complete, and curative or non-curative resection rates. Overall and tumor-specific survival, local or distant recurrence, and 3-and 5-year cumulative overall metachronous cancer rates were also *** Eligible patients had dysplasia/intraepithelial neoplasia(22%) or early cancers(squamous cell carcinoma, 78%). The median procedural time was 60 min and on average, the lesions measured 24 mm in diameter, yielding 33-mm tissue defects. The en bloc resection rate was 100%, with 95% and 81% of dissections deemed histologically complete and curative, respectively. All procedures were completed without accidental incisions/perforations or delayed bleeding. During follow-up(mean, 35 ± 23 mo), no local recurrences or metastases were observed. The 3-and 5-year survival rates were 83% and 70%, respectively, with corresponding rates of 85% and 75% for curative resections and 74% and 49% for noncurative resections. The 3-and 5-year cumulative rates of metachronous cancer in the patients with curative resections were 14% and 26%, *** ESD procedures using the SB knife are feasible, safe, and effective for treating early esophageal neoplasms, yielding favorable short-and long-term outcomes.
