氨基酸类化合物在现代医药化学研究和药物设计开发方面具有非常重要的作用,一直以来,开发高效的合成方法学来制备这些氨基酸类化合物的研究吸引了化学家们广泛的研究兴趣.另一方面,近些年电化学反应逐渐兴起成为一种绿色可再生的合成手段,是有机合成化学领域的研究热点,其在氨基酸的合成方面的应用也受到了极大的关注.本综述第一次较为系统全面地总结了近些年有关电化学手段合成氨基酸的研究进展,讨论了这些电化学方法在合成氨基酸类化合物方面的实际应用的可行性、具备的优势以及存在的一些局限,为氨基酸绿色合成方面的研究提供了一定的参考.

Chemoprevention of colorectal cancer by targeting obesity-related metabolic abnormalities

Obesity and its related metabolic disorders, including insulin resistance and chronic inflammation, increase the risk of colorectal cancer(CRC). This observation suggests that the metabolic abnormalities associated with obesity can be effective targets for preventing the development of CRC in obese individuals. In recent years, many studies using obese and diabetic animal models have been conducted to investigate the chemoprevention of CRC using pharmaceutical or nutritional interventions. Pitavastatin, a medicine used to treat hyperlipidemia, prevents the development of obesityrelated colorectal carcinogenesis by attenuating chronic inflammation. Anti-hypertensive medicines, such as captopril and telmisartan, also suppress the formation of colonic preneoplastic lesions in obese and diabetic mice. In addition, several phytochemicals, including green tea catechins, have been reported to improve metabolic disorders and prevent the development of various cancers, including CRC. Moreover, the administration of branched-chain amino acids, which improves protein malnutrition and prevents the progression of hepatic failure, is effective for suppressing obesityrelated colon carcinogenesis, which is thought to be associated with improvements in insulin resistance. In the present article, we summarize the detailed relationship between metabolic abnormalities and the development of CRC. This review also outlines recent evidence, in particular drawing from basic and clinical examinations using either pharmaceutical or nutritional intervention that suggests that targeting metabolic alterations may be an effective strategy for preventing the development of CRC in obese individuals.

＜正＞The present paper describes a new method for numerical analysis of tooth contact area in computer simulation for plunge cut shaving process. A computer simulation program has been developed to reduce trial-and-error processes that are required to improve a shaved gear tooth flanks. In the simulation, stock removals of shaved gear tooth flanks are evaluated from depth of cut of cutting edge. The depths of cut are calculated at all of referred points on the calculating tooth flanks. In order to reduce a calculation time, the depth of cut was calculated only for positions near geometrical contact area. As a result, the calculation time is remarkably reduced.

A new method is proposed to suppress chatter, in which the ultrasonic elliptical vibration is added on the cutting tool edge. It results in the fact that the rake face of tool is separated from the chip and the direction of the frictional force between the rake face and the chip is reversed in each cycle of elliptical vibration cutting. The experimental investigations show that the chatter can be suppressed effectively by adding ultrasonic elliptical vibration on the cutting tool edge. In order to make clear the reason of chatter suppression, the mechanism of chatter suppression is analyzed theoretically from the viewpoint of energy.

We investigated the effectiveness and usefulness of a novel tool: the Power Tree?, for body massage in 10 healthy female volunteers (age range, 24 - 55 years;mean age, 40.5 years) by evaluating several dermatological and psychological parameters, such as the amount of dermal collagen, the skin temperature, the level of salivary amylase and the scores on the State-Trait Anxiety Index (STAI). After 60-minute Power Tree?-mediated body massage, both the dermal collagen score determined from the DermaLab? images and skin temperature measured by infrared thermography were found to have increased significantly in several body sites compared to those before the treatment (p < 0.01 and p < 0.001, respectively). Although the level of stress estimated by the amount of amylase in the saliva was not significantly different pre- and post-procedure (p = 0.3), the treatment significantly reduced both the state anxiety (SA) and trait anxiety (TA) scores on the STAI (p < 0.001 for the SA, p < 0.01 for the TA). The treatment with this device was smoothly performed without any burden on the therapists during the present study. These data suggest that the newly-developed device is a powerful and useful tool for reflexology when used for full body massage therapy, and massage therapy using this device may produce beneficial, physiological effects as well as psychosocial improvements.

The molecular clock component Brain and Muscle Arnt-Like protein-1 (BMAL-1) affects various biologic processes, including cell survival, in numerous cell types. We previously demonstrated that BMAL1 positively regulates cell proliferation in Vascular Smooth Muscle Cells (VSMCs). However, its role in VSMC inflammation remains unelucidated. Because doxorubicin causes phlebitis associated with vascular inflammation, the present study used cultured VSMCs to investigate whether BMAL1 affected doxorubicin-induced vascular inflammation. Doxorubicin treatment led to Increased Interleukin (IL)-6 mRNA expression with an increase in BMAL1 expression in VSMCs. BMAL1 knockdown significantly increased IL-6 mRNA and further enhanced doxorubicin-induced IL-6 mRNA expression in VSMCs. BMAL1 knockdown also significantly decreased cell viability and affected the expression of other clock genes, including Per1 and Clock. Furthermore, the levels of nuclear factor erythroid 2-related factor 2, which has anti-inflammatory effects, increased in VSMCs with BMAL1 knockdown. Finally, BMAL1 knockdown increased NADPH oxidase 4 mRNA, p38α mRNA, and p38β mRNA levels, leading to increased total p38 Mitogen-Activated Protein Kinase (MAPK) and phosphorylated p38 MAPK. Fucose (6-deoxy-L-galactose) is a monosaccharide that is found on glycoproteins and glycolipids in verteb rates, invertebrates, plants, and bacteria. Fucosylation, which comprises the transfer of a fucose residue to oligosaccharides and proteins, is regulated by many kinds of molecules, including fucosyltransferases, GDP-fucose synthetic enzymes, and GDP-fucose transporter(s). Dramatic changes in the expression of fucosylated oligosaccharides have been observed in cancer and inflammation. Thus, monoclonal antibodies and lectins recognizing cancer-associated fucosylated oligosaccharides have been clinically used as tumor markers for the last few de cades. Recent advanced glycomic approaches allow us to identify novel fucosylation-related tumor markers. Moreover, a growing body of evidence supports the functional significance of fucosylation at various pathophysiological steps of carcinogenesis and tumor progression. This review highlights the biological and medical signifi cance of fucosylation in gastrointestinal cancer.
