Relationship between therapeutic efficacy of arterial infusion chemotherapy and expression of P-glycoprotein and p53 protein in advanced hepatocellular carcinoma

AIM： To investigate the relationship between the chemotherapeutic drug efficacy and the expression of P-glycoprotein （PGP） and p53 protein in advanced hepatocellular carcinoma （HCC）. METHODS： The study was conducted on 41 patients with advanced HCC who were treated by repeated arterial infusion chemotherapy. Biopsy specimens from the tumor were collected before the start of treatment in all the patients, and the specimens were stored frozen until immunohistochemical staining, which was performed after the start of treatment, to detect PGP and p53 protein expressions. Twenty of the fortyone patients were treated with an anthracycline drug （epirubicin hydrochloride; anthracycline group）, and the remaining 21 were treated with a non-anthracycline drug （mitoxantrone hydrochloride in 11 patients and carboplatin in 10 patients; non-anthracycline group）. The relationship between the chemotherapeutic efficacy and the results of immunostaining were compared between the two groups. RESULTS： Before the start of the treatment, PGPpositive rate was 90.2% （strongly-positive, 36.6%） and p53 protein-positive rate was 34.1% （strongly-positive, 19.5%）. In the anthracycline group, the response rate was 40.0%. The number of patients showing poor response to the treatment was significantly larger in the patients with strongly positive PGP expression （P= 0.005）, and their prognoses were poor （P= 0.001）. in the nonanthracycline group, the response rate was 42.9%,and there was no significant relationship between the chemotherapeutic drug efficacy and the PGP or p53 protein expression. When only the data from the 11 patients treated with anthraquinone drug, mitoxantrone, were analyzed, however, the number of patients who showed poor response to treatment was significantly higher among the p53-positive patients （P= 0.012）, irrespective of the survival outcome. During vascular development, procambial and cambial cells give rise to xylem and phloem cells. Because the vascular tissue is deeply embedded, it has been difficult to analyze the processes of vascular development in detail. Here, we establish a novel in vitro experimental system in which vascular development is induced in Arabidopsis thaliana leaf-disk cultures using bikinin, an inhibitor of glycogen synthase kinase 3 proteins. Transcriptome analysis reveals that mesophyll cells in leaf disks synchronously turn into procambial cells and then differentiate into tracheary elements. Leaf-disk cultures from plants expressing the procambial cell markers TDRpro：GUS and TDRpro：YFP can be used for spatiotemporal visualization of procambial cell formation. Further analysis with the tdr mutant and TDIF （tracheary element differentiation inhibitory factor） indicates that the key signaling TDIF-TDR-GSK3s regulates xylem differentiation in leaf-disk cultures. This new culture system can be combined with analysis using the rich material resources for Arabidopsis including cell-marker lines and mutants, thus offering a powerful tool for analyzing xylem cell differentiation.

We have developed a loop thermosyphon for cooling electronic devices. The cooling performance of a thermosyphon deteriorates with an increasing amount of non-condensable gas (NCG). Design of a thermosyphon must consider NCG to provide guaranteed performance for a long time. In this study, the heat transfer performance of a thermosyphon was measured while changing the amount of NCG. The resultant performances were expressed as approximations. These approximations enabled us to predict the total thermal resistance of the thermosyphon by the amount of NCG and input heating. Then, using the known leakage in the thermosyphon and the amount of dissolved NCG in the water, we can predict the amount of NCG and the total thermal resistance of the thermosyphon after ten years. Although there is a slight leakage in the thermosyphon, we are able to design a thermosyphon with a guaranteed level of cooling performance for a long time using the proposed design method.

Immune checkpoint and inflammation as therapeutic targets in pancreatic carcinoma

This paper investigates the occupational attainment and job mobility of permanent rural- to-urban migrants and compares them with migrants who were born with an urban hukou. Using data from the 2003 China General Social Survey, we examine how much of the gaps in occupational-prestige scores between rural- and urban-born migrants can be explained by differences in observable characteristics up to the time of migration. We find that, with controls for these characteristics, the difference in occupational attainment between rural and urban migrants becomes statistically insignificant or even positive for some subgroups. In contrast, our analysis of job mobility reveals that rural migrants are generally more mobile and also more likely to move to better jobs by changing work- units, whereas urban migrants are more likely to be promoted within a work-unit.

Pancreatic adenocarcinoma(PAC) is one of the most deadly malignant neoplasms,and the efficacy of conventional cytotoxic chemotherapy is far from satisfactory. Recent research studies have revealed that immunosuppression and inflammation are associated with oncogenesis,as well as tumor development,invasion,and metastasis in PAC. Thus,immunosuppressionrelated signaling,especially that involving immune checkpoint and inflammation,has emerged as novel treatment targets for PAC. However,PAC is an immuneresistant tumor,and it is still unclear whether immune checkpoint or anti-inflammation therapies would be an ideal strategy. In this article,we will review immune checkpoint and inflammation as potential targets,as well as clinical trials and the prospects for immunotherapy in PAC.

The genomic DNA of bacteria is highly compacted in one or a few bodies known as nucleoids. In order to understand the overall configuration and physiological activities of the cyanobacterial nucleoid under various growth conditions and the role(s) of each nucleoid protein in clock function, thylakoid membrane-associated nucleoids from the Synechococcus elongatus (se) PCC 7942 strain were isolated and purified in presence of spermidine at low salt concentrations by sucrose density gradient centrifugation. The sedimentation rates, protein/DNA composition and microscopic appearances as well as variation in structural components of clock proteins from the isolated nucleoids were compared under identical conditions. Microscopic appearances of the nucleoids were consistent with the sedimentation profiles. The nucleoid structure in the wild type was more tightly compacted than that in the KaiABC mutant strain. Western immunoblot analyses revealed that the KaiC was associated with the nucleoid fraction whereas maximum KaiA was localized in the cytosolic fraction, supposedly in association with the translation machinery.
