In order to compare the difference between young and old intervertebral disc cells and theirresponsiveness to recombinant human bone morphogenetic protein-2 (rhBMP-2),disc cells were isolatedfrom the anulus fibrosus (AF) and transition zones of lumbar discs from eight old and eight young NewZealand white *** with the ceils from the young rabbits,cells from old rabbits respond less torhBMP-2 treatment with respect to sulfated-glycosaminoglycan (sGAG) synthesis and aggrecan *** in collagen Ⅰ and collagen Ⅱ gene expressions,there are no significant differences betweenthe old and the *** comparing sGAG content,aggrecan,and collagen Ⅱ gene expression of the oldAF cells after rhBMP-2 treatment with that of the young AF cells without rhBMP-2 treatment,the old AFcells with rhBMP-2 treatment have a greater capacity to synthesize sGAG bound in the cells and to releasesGAG in the media,as well as to express aggrecan and collagen Ⅱ *** can be concluded that old AF cellsafter rhBMP-2 treatment have a greater capacity to synthesize sGAG and express aggrecan and collagen Ⅱ ascompared to young AF cells without rhBMP-2 *** rhBMP-2 can reverse the decline in theanabolic capacity of the disc cells with *** it seems that rhBMP-2 has potential for use as an agent toretard a key component of disc degeneration and loss of disc matrix.

This paper described the use of fy ash for soil amelioration of acidic soils to promote plant *** mining sites,acid sulfate soils/rocks,which contain sulfde minerals(*** FeS_(2)),have appeared as a result of overburden *** excessively acidic condition inhibits plant growth due to the dissolution of harmful elements,such as Al,Fe,and *** ash,an alkaline byproduct of coal combustion generated in thermal power plants is expected to be adopted to ameliorate acidic ***,the mixing ratio of fy ash must be considered because excessive addition of fy ash can have a negative impact on plant growth due to its physical/chemical *** pot trials using Acacia mangium demonstrate the evolution of plant growth with a 5%–10%addition of fy ash into acidic *** the acidic soil has a high potential for metal dissolution,the metal ions leached from the acidic soil are large,making it difcult to improve plant growth due to osmotic and ionic *** work suggests that the efects of fy ash on metal ions leached from the soil have to be considered for the amelioration of acidic soil.

This paper analyzes the correctly-expanded supersonic jet from a convergent-divergent axisymmetric nozzle by using numerical simulation of turbulent *** the calculated density distributions in this flow are compared with the present experimental data using rainbow schlieren *** value of the density from the experimental data agrees well with the results calculated by this ***,the present method of the measurement using rainbow schlieren deflectometry is useful for the measurement of the density of the correctly-expanded supersonic jet.

ALG-2(a gene product of PDCD6) is a 22-kD protein containing five serially repetitive EF-hand structures and belongs to the penta-EF-hand(PEF) family,including the subunits of typical ***-2 is the most conserved protein among the PEF family members and its homologs are widely found in eukaryotes.X-ray crystal structures of various PEF proteins including ALG-2 have common features:presence of eightα-helices and dimer formation via paired EF5s that are positioned in anti-parallel ***-2 forms a homodimer and a heterodimer with its closest paralog *** calmodulin,a well-known four-EF-hand protein,ALG-2 interacts with various proteins in a Ca2+-dependent fashion,but the binding motifs are completely *** some exceptions,ALG-2-interacting proteins commonly contain Pro-rich regions,and ALG-2 recognizes at least two distinct Pro-containing motifs:PPYP(X) nYP(X,variable;n=4 in ALIX and PLSCR3) and PXPGF(represented by Sec31A) .A shorter alternatively spliced isoform,lacking two residues and designated ALG-2 GF122,does not bind ALIX but maintains binding capacity to Sec31A.X-ray crystal structural analyses have revealed that binding of calcium ions induces the configuration of the side chain of R125 so that it opens Pocket 1,which accepts PPYP,but Pocket 1 remains closed in the case of ALG-2 ***-2 dimer has two ligand-binding sites,each in a monomer molecule,and appears to function as a Ca2+-dependent adaptor protein to either stabilize a preformed complex or to bridge two proteins on scaffolds in systems of the endosomal sorting complex required for transport(ESCRT) and ER-to-Golgi transport.

Molecular Weight and Aggregation of Erwinia Gum in Aqueous Solutions

Erwinia(E) gum is composed of glucose, fucose, galactose and glucuronic acid. The weightaverage molecular weights M w, numberaverage molecular weights M n and intrinsic viscosities[η] of the four fractions and the unfractionated E gum in aqueous solutions at desired temperatures were studied by light scattering, membrane osmometry, size exclusion chromatography(SEC) and viscometry. The experimental results prove that E gum formed aggregates in the aqueous solution at 25 ℃ and the aggregates were broken gradually with increasing temperature. The dissociation of the aggregates of E gum in the aqueous solution started at 36 ℃, and was completed at around 90 ℃. The [η] values of E gum and its fractions are much higher than those of the conventional polymers with the similar molecular weights, and decrease with increasing NaCl concentration.

Erwinia (E) gum, an extracellular polysaccharide, is composed of fucose, galatose, glucose and glucuronic acid. Its viscosity behavior was investigated by a low-shear-rate multiball viscometer and a rotational viscometer. Its weight-average molecular weight M-w and intrinsic viscosity [eta] in 0.2 mol/L NaCl aqueous solution were measured by light scattering method at 35 degrees C and viscometry at 25 degrees C and found to be 1.06 x 10(6) g/mol and 1050 mL/g, respectively, and its aggregates in aqueous solution were proved by gel permeation chromatography (GPC). These results indicated that E gum in water has exceedingly high viscosity and exhibits Binham fluid behavior, owing to its aggregation. The viscosity of E gum decreased with increasing temperature, and the turning point appeared at 38 degrees C for dilute solution and 80 degrees C for concentrated solution suggesting that the aggregates of E gum in water started to disaggregate under these temperatures. In addition, the aggregates can be disrupted by adding either acid or base. The experimental results indicated that the E gum is a good thickening agent, and its fluid behavior is similar to xanthan.

Gene expression analysis in the larval silk gland of the eri silkworm Samia ricini

Insects produce silk for a range of purposes. In the Lepidoptera, silk is utilized as a material for cocoon production and serves to protect larvae from adverse environmental conditions or predators. Species in the Saturniidae family produce an especially wide variety of cocoons, for example, large, golden colored cocoons and those with many small holes. Although gene expression in the silk gland of the domestic silkworm （Bombyx mori L.） has been extensively studied, considerably fewer investigations have focused on members of the saturniid family. Here, we established expression sequence tags from the silk gland of the eri silkworm （Samia ricini）, a saturniid species, and used these to analyze gene expression. Although we identified thefibroin heavy chain gene in the established library, genes for other major silk proteins, such asfibroin light chain andfibrohexamerin, were absent. This finding is consistent with previous reports that these latter proteins are lacking in saturniid silk. Recently, a series offibrohexamerin-like genes were identified in the Bombyx genome. We used this information to conduct a detailed analysis of the library established here. This analysis identified putative homologues of these genes. We also found several genes encoding small silk protein molecules that are also present in the silk of other Lepidoptera. Gene expression patterns were compared between eri and domestic silkworm, and both conserved and nonconserved expression patterns were identified for the tested genes. Such differential gene expression might be one of the major causes of the differences in silk properties between these species. We believe that our study can be of value as a basic catalogue for silk gland gene expression, which will yield to the further understanding of silk evolution.

The broad availability of high throughput nanostructure fabrication is essential for advancement in nanoscale science. Large-scale manufacturing developed by the semiconductor industry is often too resource-intensive for medium scale laboratory prototyping. We demonstrate the inexpensive wafer scale direct- write of Ge and Si nanostructures with a 4-inch mask aligner retrofitted with a conducting microstructured stamp. A bias applied between the stamp and an underlying silicon substrate results in the reaction of diphenylgermane and diphenylsilane precursors at the stamp--substrate interface to yield the direct- write of Ge and Si nanostructures in determined locations. With the increasing number of outdated mask aligners available from the semiconductor industry and an extensive library of liquid precursors, this strategy provides facile, inexpensive, wafer scale semiconductor direct-write for applications such as electronics, photonics, and photovoltaics.
