＜正＞ WC-Co cermet is widely used in industrial applications such as cutting tools, dies, wear parts and so on. It is of great importance to establish the recycling process for the precious metal resources contained in WC-Co cermet, because all these metals used in Japan are imported. In this paper we reported a hydrothermal oxidation technique using nitric acid for the reclamation of WC and Co. The WC-Co cermet specimens with various WC particle sizes and Co contents were hydrothermally treated in HNO3 aqueous solutions at temperatures of 110-200℃for durations of 6-240 h. The Co was preferentially leached out into the acidic solution, while the WC was oxidized to insoluble WO3 hydrate which was subsequently separated by filtration. The hydrothermal treatment parameters such as solvent concentrations, treatment temperatures, holding time were optimized in respect to different kinds of WC-Co cermets. A hydrothermal oxidation treatment in 3M HNO3 aqueous solution at 150℃for 24 h was capable of fully disintegrating the cermet chip composed of coarse WC grains of 1-5 μm in size with 20 wt% of Co as binder. While the more oxidation resistant specimen composed of fine WC grains of 0.5-1.0 urn in size with 13 wt% of Co, was completely disintegrated by a treatment in 7 M HNO3 aqueous solution at 170℃for 24 h. The filtered solid residues were composed of fine WO3.0.33H2O powder and a small amount of WO3. The recovered WO3.0.33H2O powder can be easily returned to the industrial process for the synthesis of WC powder so that the overall recycling cost can be possibly lowered.

It is known to be that lactic acid bacteria induce the IL-12. IL-12 activates NK cells and promotes the production of IFN-γ. IFN-γ activates macrophages, resulting in enhanced phagocytosis and bactericidal activity. We have been investigating fermented foods that activate the immune function. For that purpose, a specific antibody is required. We tried to express IL-12p35 by the usual method, but IL-12p35 was not expressed at all. In the present study, we constructed, purified human IL-12p35 and obtained a specific antibody against IL-12p35. We also purified human IFN-γ and obtained specific antibody against IFN-γ. We have established a method for expressing poorly expressed proteins. The method we have established can be applied to the purification of poorly expressed proteins and antibody production.

The ATPase activity of molecular chaperone HSP60 is inhibited by immunosuppressant mizoribine

The molecular chaperone HSP60 is a chaperonin homolog of GroEL. We had previously shown that the immunosuppressant mizoribine is bound directly to HSP60 and inhibited its chaperone activity. However, the inhibitory mechanisms of HSP60 by mizoribine have not yet been fully understood. In the present study, we investigated the influence of mizoribine on a folding cycle of HSP60 and co-chaperone HSP10. Our results showed that mizoribine inhibited the folding cycle of HSP60/HSP10. The ATPase activity of HSP60/HSP10 was decreased in the presence of mizoribine and the dissociation of HSP10 from HSP-60 was also decreased by mizoribine. The same functions of GroEL and/or GroES were slightly affected by mizoribine. Based on our findings, we discuss the inhibitory mechanisms of HSP60 by mizoribine.

日本雅马哈发动机株式会社最近成功开发了一种小型轿车用新一代柴油机。该项目已进行了若干年,旨在创造一种能满足21世纪环保要求的用于欧洲市场的汽车发动机,要求低燃油耗、低排放、高功率、尺寸小、质量轻。开发的发动机是一种采用曲轴箱压缩、回流扫气的二冲程柴油机,装有半直喷式燃烧室。此外,通过可变压缩比机构、扫气容积控制、电控分配式油泵(控制喷油量和喷油定时)、计算机控制的润滑系统和可溶性有机成分(SOF)还原催化器的结合,实现了低燃油耗和低排放。发动机装在小型轿车上,然后对车辆的各种性能进行了评定。试验结果表明,发动机实现了3L/100 km的低燃油耗,排放达欧4标准。

Cisplatin Inhibits AhR Activation

The AhR binds to contain ligands, such as 2, 3, 7, 8-tetrachlorodibenzo-p-dioxin, 3-methylcholantrene, or β-naphthoflavone. The activation mechanism of AhR is not yet fully understood, but it is known that AhR associates with the molecular chaperone HSP90 in the cytoplasm. There are a few reports about the association or dissociation of AhR and HSP90, and which domain of HSP90 binds to AhR. We reported the association and activation mechanisms between HSP90 and AhR-PAS or AhR-bHLH. In the current study, we found that cisplatin inhibits the AhR activation. Although ATP and 17-DMAG have no effect on the dissociation of HSP90 from AhR, some contents of HSP90 were dissociated from AhR in the presence of cisplatin. We could detect the increase of CYP1A in the presence of 3-MC. On the contrary, the induction of CYP1A1 was inhibited in the presence of cisplatin. We couldn’t detect AhR in the HeLa cell soluble fraction in the presence of 50 μM cisplatin. In the presence of MG-132, we could detect AhR. These results suggested that AhR was dissociated from the HSP90 chaperone complex and processed during the protein proteasome degradation system in the presence of cisplatin.

Cytosolic chaperonin CCT possesses GTPase activity

Cytosolic chaperonin CCT (also known as TRiC) is a hetero-oligomeric cage-like molecular chaperone that assists in protein folding by ATPase cycle-dependent conformational changes. However, role of the nucleo-tide binding and hydrolysis in CCT-assisted protein folding is still poorly understood. We purified CCT by using ATP-Sepharose and other columns, and found that CCT possesses ability to hydrolyze GTP, with an activity level very similar to the ATPase activity. CCT was more resistant to proteinase K treatment in the presence of GTP or ATP. These results suggest that the GTPase activity of CCT may play a role in chaperone-assisted protein folding.
