BACKGROUND In recent.years,t.e emergence of mult.plex t.chnology t.at.can simult.neously measure mult.ple ant.-islet.aut.ant.bodies has become part.cularly valuable for t.e st.ging and early diagnosis of immune-mediat.d t.pe 1 diabet.s(t.D).While it.has been est.blished t.at.20%-30%of t.D pat.ent. suffer from aut.immune t.yroid disease(AIt.),t.ere is limit.d available dat. regarding t.e presence of ant.-islet.aut.ant.bodies in AIt. *** commercially available ant.-islet.aut.ant.bodies,glut.mic acid decarboxylase 65 aut.ant.bodies(GADAs)are oft.n t.e first.marker measured in general clinical *** t. invest.gat. t.e frequency of ant.-islet.aut.ant.bodies in AIt. *** Our st.dy involved four hundred ninet.-five AIt. pat.ent.,cat.gorized int. t.ree dist.nct.groups:AIt. wit. t.D(n=18),AIt. wit. phenot.pic t.pe 2 diabet.s(t.D)(n=81),and AIt. wit.out.diabet.s(n=396),and t.e enzyme-linked immunosorbent.assay(ELISA)was employed t. det.rmine t.e frequencies of 3 Screen Islet.Cell Aut.ant.body(3 Screen ICA),GADA,insulinoma-associat.d ant.gen-2 aut.ant.bodies(IA-2As),and zinc t.ansport.r 8 aut.ant.bodies(Znt.As)wit.in t.ese *** t.e frequency of 3 Screen ICA in AIt. pat.ent. wit. t.D,t.D,and t.ose wit.out.diabet.s were 88.9%,6.2%,and 5.1%,respect.vely,wit. no significant.difference seen bet.een t.e lat.er t.o ***,t.e frequency of 3 Screen ICA was 11.1%higher in AIt. pat.ent. wit. t.D,1.3%higher in AIt. pat.ent. wit. t.D,and 1.1%higher in AIt. pat.ent. wit.out.diabet.s compared t. AIM： t. clarify differences in mucin phenot.pe, prolif- erat.ve act.vit. and oncogenet.c alt.rat.on among sub- t.pes of colorect.l lat.rally spreading t.mor （LSt.. MEt.ODS： LSt., defined as superficial elevat.d lesions great.r t.an 10 mm in diamet.r wit. a low vert.cal axis, were macroscopically classified int. t.o subt.pes：（1） a granular t.pe （Gr-LSt. composed of superficially spread- ing aggregat.s of nodules forming a fiat.based lesion wit. a granulonodular and uneven surface; and （2） a non-granular t.pe （NGr-LSt. wit. a flat.smoot. surface and an absence of granulonodular format.on. A t.t.l of 69 LSt., comprising 36 Gr-LSt. and 33 NGr-LSt., were immunohist.chemically st.ined wit. MUC2, MUC5AC, MUC6, CD10 （markers of gast.oint.st.nal cell lineage）, p53, 13-cat.nin and Ki-67 ant.bodies, and examined for alt.rat.on in exon 1 of v-Ki-ras2 Kirst.n rat.sarcoma viral oncogene homolog （KRAS） and exon 15 of v-raf murine sarcoma viral oncogene homologue B1 （BRAF） by poly- merase chain react.on followed by direct.sequencing. RESULt.： Hist.logically, 15 Gr-LSt.samples were ad- enomas wit. low-grade dysplasia （LGD）, 12 were high- grade dysplasia （HGD） and 9 were adenocarcinomas invading t.e submucosa （INV）, while 12 NGr-LSt. demonst.at.d LGD, 14 HGD and 7 INV. In t.e proximal colon, MUC5AC expression was significant.y higher in t.e Gr-t.pe t.an t.e NGr-t.pe. MUC6 was expressed only in NGr-LSt. MUC2 or CD10 did not.differ. P53 ex- pression demonst.at.d a significant.st.pwise increment.in progression t.rough LGD-HGD-INV wit. bot. t.pes of LSt. Nuclear β-cat.nin expression was significant.y higher in t.e NGr-t.pe. Ki-67 expression was signifi- cant.y higher in t.e Gr-t.pe in t.e lower one t.ird zone of t.e t.mor. In proximal, but.not.dist.l colon t.mors, t.e incidence of KRAS provided mut.t.on was signifi- cant.y higher in t.e Gr-t.pe harboring a specific mut.- t.onal pat.ern （G12V）. AIM： t. compare t.e prognosis of pat.ent. who underwent.hepat.ct.my and ablat.on using t.e modified Japan Int.grat.d St.ging score （mJIS）. MEt.ODS： We examined t.e clinicopat.ologic records and pat.ent.out.omes in 278 HCC pat.ent. including 226 undergoing hepat.ct.my and 52 undergoing ablat.on t.erapy. RESULt.： Cirrhosis was more frequent.in t.e ablat.on group. t.mor size, number and presence of vascular invasion were significant.y higher in t.e operat.on group compared t. t.e ablat.on group. t.e local recurrence rat. adjacent.t. t.eat.d lesions was significant.y higher in t.e ablat.on group compared t. t.e operat.on group （P 〈 0.05）. t.e 3- and 5-year survival rat.s in t.e ablat.on and t.e operat.on group were 66% and 78%, and 50% and 63%, respect.vely, but.not.significant.y different. Over 50% survival rat.s were observed in pat.ent. wit. a *** score of 0-2 in bot. groups. However, survival rat.s wit. a score of 3-5 in bot. groups were significant.y lower. CONCLUSION： According t. t.e mJIS syst.m, bot. local t.eat.ent. could be select.d for pat.ent. wit. a score of 0-2. However, for pat.ent. wit. a score more t.an 3, liver t.ansplant.t.on might.be a bet.er opt.on in pat.ent. wit. HCC.

We previously demonst.at.d t.at.polypod-like st.uct.red DNA, or polypodna, const.uct.d wit. t.ree or more oligodeoxynucleot.des （ODNs）, is efficient.y t.ken up by immune cells such as dendrit.c cells and macrophages, depending on it. st.uct.ral complexit.. t.e ODNs comprising t.e polypodna should bend t. form t.e polypod-like st.uct.re, and may do so by adopt.ng eit.er a bend- t.pe conformat.on or a cross-t.pe conformat.on. Here, we t.ied t. elucidat. t.e orient.t.on and bending of ODNs in polypodnas using at.mic force microscopy （AFM）. We designed t.o t.pes of pent.podnas （i.e., a polypodna wit. five pods） using 60- t. 88-base ODNs, which were t.en immobilized on DNA origami frames. AFM imaging showed t.at.t.e ODNs in t.e pent.podna adopt.d bend-t.pe conformat.ons. t.t.apodna and hexapodna also adopt.d bend-t.pe conformat.ons when t.ey were immobilized on frames under unconst.ained condit.ons. t.ese findings provide useful informat.on t.ward t.e coherent.design of, and t.e st.uct.re-act.vit. relat.onships for, a variet. of DNA nanost.uct.res.

AIM: t. elucidat. t.e relat.onship bet.een t.e microvessel count.(MVC) by CD34 analyzed by immunohist.chemical met.od and prognosis in hepat.cellular carcinoma (HCC) pat.ent. who underwent.hepat.ct.my based on our preliminary st.dy. MEt.ODS: We examined relat.onships bet.een MVC and clinicopat.ological fact.rs in 128 HCC pat.ent.. t.e modifi ed Japan Int.grat.d St.ging score (mJIS) was applied t. examine subset. of HCC pat.ent.. RESULt.: Median MVC was 178/mm^2, which was used as a cut.off value. MVC was not.signif icant.y associat.d wit. any clinicopat.ologic fact.rs or post.perat.ve recurrent.rat.. Lower MVC was associat.d wit. poor disease-free and overall survivals by univariat. analysis (P = 0.039 and P = 0.087, respect.vely) and lower MVC represent.d an independent.poor prognost.c fact.r in disease-free survival by Cox’s mult.variat.analysis (risk rat.o, 1.64; P = 0.024), in addit.on t. t.mor size, vascular invasion, macroscopic fi nding and hepat.c dysfunct.on. Signifi cant.differences in disease-free and overall survivals by MVC were observed in HCC pat.ent. wit. mJIS 2 (P = 0.046 and P = 0.0014, respect.vely), but.not.in t.ose wit. ot.er scores. CONCLUSION: t.mor MVC appears t. offer a useful prognost.c marker of HCC pat.ent.survival, part.cularly in HCC pat.ent. wit. mJIS 2.
