AAV6 as an effective gene delivery vector for prolonged transgene expression in intervertebral disc cells in vivo

Intervertebral disc degeneration is the main contributor to low back pain,now the leading cause of disability *** transfer,either in a therapeutic attempt or in basic research to understand the mechanisms of disc degeneration,is a fascinating and promising tool to manipulate the complex physiology of the *** vectors based on the adeno-associated virus(AAV)have emerged as powerful transgene delivery vehicles yet a systematic investigation into their respective tropism,transduction efficiency,and relative toxicity have not yet been performed in the disc in ***,we used in vivo bioluminescence imaging to systematically compare multiple AAV serotypes,injection volumes,titers,promoters,and luciferase reporters to determine which result in high transduction efficiency of murine nucleus pulposus(NP)cells in *** find that AAV6 using a CAG promoter to drive transgene expression,delivered into the NP of murine caudal discs at a titer of 1011 GC/mL,provides excellent transduction efficiency/kinetics and low toxicity in *** also show,for the first time,that the transduction of NP cells can be significantly boosted in vivo by the use of small cell permeabilization ***,to our knowledge,we are the first to demonstrate the use of optical tissue clearing and three-dimensional lightsheet microscopy in the disc,which was used to visualize fine details of tissue and cell architecture in whole intact discs following AAV6 *** together,these data will contribute to the success of using AAV-mediated gene delivery for basic and translational studies of the IVD.

The effect of the tilt angle on mixed convection and related heat transfer in a“T”shaped double enclosure with four heated obstacles on the bottom surface is numerically *** considered obstacles are constantly kept at a relatively high(fixed)temperature,while the cavity’s upper wall is *** finite volume approach is used to solve the mass,momentum,and energy equations with the SIMPLEC algorithm being exploited to deal with the pressure-velocity *** is put on the influence of the tilt angle on the solution symmetry,flow structure,and heat exchange through the *** following parameters and related ranges are considered:Rayleigh number 104≤Ra≤5.105,tilt angle 0°≤φ≤90°,Reynolds number 100≤Re≤1000,Prandtl number Pr=0.72,block height B=0.5,opening width C=0.15,and distance between blocks D=*** results reveal different branches of solutions on varying Re andφ.They also show that the symmetry of the solution regarding the P_(2)axis is retained for all cases with no tilt and for values of Re between 100 and 1000.

This paper discusses the elaboration of adsorbents from oil *** experimental designs a response surface methodology(RSM),which has been applied to optimize the significant preparation factors,such as tempera-ture,time,and the activating agent *** results obtained from central composite design(CCD)revealed that the interaction between the factors was significant for the maximum quantity of adsorption(response).Planned results have shown that a maximum quantity of adsorption for methylene blue is 65 mg/g,which could be achieved with a temperature of 275℃over 2 h and a percentage of the activating agent of 45%.The predicted values agreed with the experimental finding,with a determination coefficient(R2)of *** model has been validated by experi-ments after conditions *** new material(RHO)was characterized by cation exchange capacity,zero charge pH,surface functions,X-ray fluorescence,specific surface area,and electron microscopy *** adsorption was determined using Langmuir,Freundlich and Temkin,which were used to describe the adsorption *** adsorption capacity of the material was about 263 mg/g,and the kinetic studies showed rapid adsorption.

Constructing the toolbox:Patient-specific genetic factors of altered fracture healing

The multifaceted sequence of events that follow fracture repair can be further complicated when considering risk factors for impaired union,present in a large and growing percentage of the *** factors such as diabetes,substance abuse,and poor nutrition affect both the young and old,and have been shown to dramatically impair the body’s natural healing *** this end,biotherapeutic interventions such as ultrasound,electrical simulation,growth factor treatment(BMP-2,BMP-7,PDGF-BB,FGF-2)have been evaluated in preclinical models and in some cases are used widely for patients with established non-union or risk/indication or impaired healing(***,BMP-2,etc.).Despite the promise of these interventions,they have been shown to be reliant on patient compliance and can produce adverse side effects such as heterotopic *** and cell therapy approaches have attempted to apply controlled regimens of these factors and have produced promising ***,there are safety and efficacy concerns that may limit the translation of these *** addition,none of the above mentioned approaches consider genetic variation between individual *** clinical and preclinical studies have demonstrated a genetic component to fracture repair and that SNPs and genetic background variation play major roles in the determination of healing *** this,there is a need for preclinical data to dissect the mechanism underlying the influence of specific gene loci on the processes of fracture healing,which will be paramount in the future of patient-centered interventions for fracture repair.

Determination of the origin of mineralization and groundwater salinity in the Adrar region in the southwest of Algeria

To some extent, the sedimentary sequence with the largest groundwater reserves in northern Sahara is marked by a certain water table level, regionally known as the "Continental Intercalaire"(CI). "Continental Intercalaire"(CI) refers to "Continental Intercalary" widely distributed among three countries. Algeria, Tunisia and Libya, which have significant potential of water resources. As it is the only water resource relatively easily accessible to the inhabitants of the Sahara, it is widely developed. The physico-chemical characteristics of statistical processing in principal component analysis(PCA) and the chemical phase measurement of groundwater in the unconfined aquifer captured by "Foggaras"(traditional system irrigation composed of well system linked by a horizontal channel from their bases) and the deep drillings located in the study area were accessible. Therefore, there were some favorable conditions for comparing the chemistry of these waters with the standards of potability established by the World Health Organization. Then, the study detected the origin of excessive mineralization and the excessive content of Na, Cl, K, Mg and Ca that originated from the leaching of the clay and carbonate layers of the "Continental Intercalaire". In addition, the enrichment in NO2-, NO3- and SO42- was due to the excessive use of fertilizer in the whole region for shallower Foggaras waters, and this study also showed the dominant chemical facies of groundwater related to the significant abundance of these mineral salts in this thick aquifer horizon.

Developing Multi-Agent System Robotics

In this paper, authors want to create a multi-agent robotics system consisting of a number of identical robots, each controlled by identical agent-based software. Therefore, the authors have to design and build a number of robots advanced enough to be able to navigate around, communicate with other robots, pick-up objects and return them back home. Another important demand is that the robot can actually be designed and build in a somewhat simple manner, as authors do not want to spend all the time at hand dealing with hardware. The software controlling the robots must be agent-based, and must be able to either run on the robots, or at least illustrate the possibility of the software actually running on the robots. Further, the authors need to design a world/environment for the robots to be situated in. This world needs to be complex enough to illustrate the problem, but simple enough for us to implement the wanted system within the given period of time.

As part of improving services done for various clients in all Moroccan areas, Moroccan exportation group of fruits and vegetables in collaboration with their packaging units and producers, tends to cooperate in order to face with international competitiveness. Indeed, the complexity of networks of partners has led policy-makers to implement new techniques and tools to help control different processes. For this reason, the implementation of a permanent monitoring of different operations ranging from production, packaging, and distribution of perishable products has become paramount. This article aims to propose a model of multi-agent citrus supply chain, based on indicators for monitoring and evaluation of performance of its logistics systems, in order to build a new independent, robust and responsive chain, and to optimize and control the flow of materials and information between the different actors and stakeholders of the chain.
