Molecular cloning and characterization of the endothelin 3 gene in black bone sheep

Background: Black bone sheep was first discovered in yunnan province of China in 1970, with unique black pigmentation on the body and internal organs. Endothelin 3(EDN3) has been known as a key gene causing hyperpigmentation in black bone chicken, the Silky ***: In this study, EDN3 was employed as a candidate gene for regulating black color pigmentation. First,EDN3 was cloned from sheep to obtain the full-length cDNA by using the rapid amplification of cDNA ends(RACE).Genomic EDN3 was screened and a total of thirty predicted single nucleotide polymorphisms(SNPs) were genotyped for allele and genotype frequency analysis in a case-control study involving two black bone sheep populations. Genomic copy number analysis of EDN3 in sheep was conducted to measure the variation in copy number. EDN3 expression levels were observed among the groups in adult liver, lymph node, and kidney tissues, as well as embryo kidney samples. Also, among the tissues of black bone and non-black bone ***: The size of the full-length cDNA was 1,578 bp, which included 426 bp of 5′-untranslated region(5′-UTR),an open reading frame(ORF) of 639 bp encoding a protein of 212 amino acids, and a 3′-UTR of 513 bp. Genotype and allele frequencies of all the discovered SNPs were found insignificantly different in black bone and non-black bone sheep(P > 0.05). Genomic copy number analysis of EDN3 in sheep revealed no significant difference between the two sheep groups. No significant variations were found in the adult liver and kidney embryo samples. However,the expression in lymph node and kidney tissue was significantly higher in black bone sheep than that in non-black bone sheep(P y indicate that unlike in Silky chickens, EDN3 is not responsible for hyperpigmentation but may play a key functional role in immune and excretory

Genome-wide profiling of RNA editing sites in sheep

Background: The widely observed RNA-DNA differences(RDDs) have been found to be due to nucleotide alteration by RNA editing. Canonical RNA editing(i.e., A-to-I and C-to-U editing) mediated by the adenosine deaminases acting on RNA(ADAR) family and apolipoprotein B mRNA editing catalytic polypeptide-like(APOBEC)family during the transcriptional process is considered common and essential for the development of an *** date, an increasing number of RNA editing sites have been reported in human, rodents, and some farm animals;however, genome-wide detection of RNA editing events in sheep has not been reported. The aim of this study was to identify RNA editing events in sheep by comparing the RNA-seq and DNA-seq data from three biological replicates of the kidney and spleen ***: A total of 607 and 994 common edited sites within the three biological replicates were identified in the ovine kidney and spleen, respectively. Many of the RDDs were specific to an individual. The RNA editing-related genes identified in the present study might be evolved for specific biological functions in sheep, such as structural constituent of the cytoskeleton and microtubule-based processes. Furthermore, the edited sites found in the ovine BLCAP and NEIL1 genes are in line with those in previous reports on the porcine and human homologs, suggesting the existence of evolutionarily conserved RNA editing sites and they may play an important role in the structure and function of ***: Our study is the first to investigate RNA editing events in sheep. We screened out 607 and 994 RNA editing sites in three biological replicates of the ovine kidney and spleen and annotated 164 and 247 genes in the kidney and spleen, respectively. The gene function and conservation analysis of these RNA editing-related genes suggest that RNA editing is associated with important gene function in sheep. Interleukin-1 and estrogen protect against disseminating dentoalveolar infections

Dentoalveolar bacterial infections cause localized tissue and bone destruction, but usually remain well-localized within teeth in immunocompetent hosts. However, in certain cases these infections may invade head and neck tissues, resulting in orofacial abscesses, cellulitis and sepsis, with resultant high morbidity and even mortality. In the present studies, we developed a novel model of spreading dentoalveolar infections in mice by treatment with neutralizing antibodies against both interleukin-la （IL-1a） and IL-1β. Surprisingly male but not female mice given anti-lL-1 antibodies developed orofacial abscesses, weight loss, splenomegaly and sepsis. Female mice developed abscesses and sepsis comparable to males following ovariectomy （OVX）, which was reversed by estrogen supplementation. Anti-lL-1 blockade inhibited IL-12, interferon y （IFNy） and IL-6 but not IL-IO expression in infrabony lesions, suggestive of a local anti-inflammatory response. There was greater infiltration of neutrophils and other inflammatory ceils into lesions in anti-lL-l-treated animals; however, blood leukocytes had reduced bacterial phagocytic and killing activity ex vivo. Estrogen directly stimulated IL-1 production by macrophages, suggesting that the resistance of females to disseminating dentoalveolar infections may be due to their heightened pro-inflammatory responses following bacterial challenge, leading to enhanced localization of these infections.

Utility-Based Anonymization Using Generalization Boundaries to Protect Sensitive Attributes

Privacy preserving data mining (PPDM) has become more and more important because it allows sharing of privacy sensitive data for analytical purposes. A big number of privacy techniques were developed most of which used the k-anonymity property which have many shortcomings, so other privacy techniques were introduced (l-diversity, p-sensitive k-anonymity, (α, k)-anonymity, t-closeness, etc.). While they are different in their methods and quality of their results, they all focus first on masking the data, and then protecting the quality of the data. This paper is concerned with providing an enhanced privacy technique that combines some anonymity techniques to maintain both privacy and data utility by considering the sensitivity values of attributes in queries using sensitivity weights which determine taking in account utility-based anonymization and then only queries having sensitive attributes whose values exceed threshold are to be changed using generalization boundaries. The threshold value is calculated depending on the different weights assigned to individual attributes which take into account the utility of each attribute and those particular attributes whose total weights exceed the threshold values is changed using generalization boundaries and the other queries can be directly published. Experiment results using UT dallas anonymization toolbox on real data set adult database from the UC machine learning repository show that although the proposed technique preserves privacy, it also can maintain the utility of the publishing data.

A crescent-shape granitic stock and associated dykes is located to the East Gabal Nuqra at the extreme western part of Wadi Natash,South Eastern Desert,*** examined granites are classified as alkali-feldspar granites and mainly consist of quartz,potash feldspars,plagioclases,and ***,zircon,apatite and allanite are accessories representing the source of y,U,Th and REEs in these *** granites are characterized by high K2O,Na2O and Zn contents and Rb/Sr ***,they are highly enriched in high field strength elements(HFSE),especially Zr(1529×10-6),Nb(100×10-6),Hf(91×10-6) and y(624×10-6) and light rare-earth elements(LREE,141×10-6) concentrations and strongly depleted in Ca,Mg,Sr and Eu *** features suggest that they are similar to A-type granites(type-2).The rhyolite dykes and granites have similar geochemical characteristics whereas the chondrite-normalized REE patterns show a LREE enriched feature with strong negative Eu-anomaly,whereas the REE pattern of trachydacites show slightly fractionated pattern with no *** is suggested that the trachydacites were generated by small degree of partial-melting deep-seated basic *** liquid,when subjected to fractional crystallization involving separation of plagioclases as residue,generated the alkali-feldspar *** further fractional crystallization gave rise to the alkali *** igneous rock suite originated from metaluminous to alkaline trachytic magma,and was developed in a within-plate tectonic *** extension caused by NW-SE right-lateral shear in area led to the emplacement of the alkali-feldspar *** later extrusion of the alkali rhyolite and trachydacite dykes was due to cauldron subsidence.

Occult HCV Infection (OCI) Diagnosis in Cirrhotic and Non-cirrhotic Naive Patients by Intra-PBMC Nested Viral RNA PCR

Background and Aims:Occult HCV infections(OCIs)include IgG antibody seronegative cryptogenic(COCIs),as well as seropositive secondary na(i)ve(SNOCIs)and experienced(SEOCIs)*** used peripheral-blood-mononuclear-cell(PBMC)-PCR to evaluate COCIs and SNOCIs prevalence,serum HCV spontaneous disappearance(SCSD)in na(i)ve cirrhotics and non-cirrhotics,intra-PBMC HCV-RNA strands in relation to cirrhosis density in na(i)ve non-viremia cases,and HCV-RNA seroconversion after 1 year of solitary naive intra-PBMC ***:The anti-HCV IgG antibodypositive na(i)ve-patients(n=785)were classified into viremic(n=673)and non-viremic[n=112,including non-cirrhotics(n=55)and cirrhotics(n=57)],and 62 controls without evidence of *** and post-HCV non-viremia cases(n=62+112=174)were submitted to hepatic FibroscanElastography *** subjects(n=847)were screened for intra-PBMC HCV-RNA sense and antisense strands by ***:Na(i)ve-OCI cases(4.84%)that were diagnosed by PBMC-PCR significantly raised the total numbers of HCV-infection to 714(p=0.01).The percent positivity of SNOCIs(34.82%)was significantly higher than for asymptomatic-COCIs(3.125%,p=0.0001).Comparing PBMC-PCR with single-step-reverse-transcription(SRT)-PCR for identification of SCSD in na(i)ve IgG antibody-positive nonviremia patients(n=112)revealed a decline in SCSD prevalence by PBMC-PCR(from 14.27%to 9.3%),regardless of presence of hepatic cirrhosis(p=0.03).SCSD was found to be higher by PBMC-PCR in non-cirrhotics compared to cirrhotics(p=0.0001),with an insignificant difference when using SRT-PCR(p=0.45).Intra-PBMC HCV-RNA infection was significantly more frequent in cirrhotics compared to both non-cirrhotics and controls(py was recognized in intra-PBMC HCV-RNA infection with sense(p=0.0001)or antisense strand(p=0.003).HCV-RNA seroconversion was associated with intra-PBMC infection when both sense and antisense strands were dete

HCV Therapy Follow-up Fractionation (CTF2) by Intra-PBMC Nested RNA PCR Recognizes Early Virologic Response and Relapse

Rare earth oxides doped NiO/γ-Al_2O_3 catalyst for oxidative dehydrogenation of cyclohexane

The effect of rare earth oxides （RE=Ce, La, Gd, and Dy） doping of alumina support in NiO/7-A1203 system was investi- gated on its catalytic performance in oxidative dehydrogenation （ODH） of cyclohexane. The physicochemical properties of various samples were followed up through N2 physisorption, temperature programmed reduction （H2-TPR）, X-ray diffraction （XRD）, X-ray photoelectron spectroscopy （XPS） and potentiometric acid-base titration techniques. In the parent NiO/y-A1203 catalyst, Ni species were found to be strongly interacted with alumina surface. Addition of rare earth dopants to ）＇-A1203 in the catalyst system affected the nickel-alumina interaction and resulted in significant modifications in the catalytic performances in the ODH reaction. The results re- vealed the beneficial role of both La203 and Gd2Os doping in enhancing the ODH catalytic activity and selectivity to cyclohexene. H2-TPR and XPS results indicated that majority of Ni species in NiO/La203 modified T-A1203 were more weakly interacted with La203 and alumina whereas both NiO like species and nickel aluminate were present on the surface. Doping with cerium or dyspro- sium increased the nickel-support interaction and led to a decrease in surface nickel concentration. In case of doping with Ce, surface concentration of cerium oxide was higher than those of the other RE oxides; the doped catalyst reached its steady state activity faster than the other catalysts. The acid-base results suggested that RE metals were interacted most likely with acidic surface hydroxyl groups. The degree of nickel-alumina interaction decreased in the following order： LaAI〉GdAI〉CeAI〉DyA1.
