Comparison of the Sensitivities of d.fferent PCR Assays for the d.tection of Mycobacterium tuberculosis Complex Isolates from Five Regions of Cameroon

The accurate d.agnosis of tuberculosis caused.by members of the Mycobacterium tuberculosis complex (MTBC) has remained.a major challenge in clinical laboratories world.wid.. Several stud.es have evaluated.the use of highly specific in-house PCR assays targeting the IS6110, hupB, rpoB, oxyR, and.IS1081 genes in the d.tection of MTBC species with reports on variable sensitivities d.pend.ng on the geographical sourcing of isolates. In the present investigations, we evaluated.the sensitivities of these PCR assays on 125 MTBC cultured.isolates from five (West, Centre, Littoral, North West and.South West) of the ten regions of Cameroon. Of this number, 124 (99.2%), 117 (93.6%), 123 (99.1%), 119 (95.2%) and.118 (94.4%) were positive by the IS6110, hupB, rpoB, oxyR, and.IS1081-based.PCR assays respectively. A total of 110 (88%) of the cultured.isolates were also id.ntified.as MTBC by stand.rd.biochemical tests. Of this number, 109 (99.1%), 104 (94.5%), 109 (99.1%), 106 (96.4%) and.104 (94.5%) were positive in the IS6110, hupB, rpoB, oxyR, and.IS1081-based.PCR assays respectively. Concord.nt PCR results were obtained.for 108 of the 125 samples. The 15 isolates that were negative biochemically scored.sensitivities ranging from 100% (for the IS6110 assay) to 86.7% (for the hupB and.oxyR assay). The combination of the IS6110 assay, which turned.out to be the most sensitive, and.each of the other assays gave 100% sensitivity. We conclud. that the combined.targeting of the IS6110 and.rpoB genes is likely to yield.the most sensitive PCR proced.re for the d.agnosis of MTBC infection in the five regions of Cameroon.

Harnessing the frequency d.mension in integrated.photonics offers key ad.antages in terms of scalability, noise resilience, parallelization, and.compatibility with telecom multiplexing *** ring resonators have been used.to generate frequency-entangled.states through spontaneous four-wave mixing. However, state-of-the-art integrated.resonators are limited.by trad.-offs among size,spectral separation, and.efficient photon pair generation. We have d.veloped.silicon ring resonators with a footprint below 0.05 mm2provid.ng more than 70 frequency channels separated.by 21 GHz. We exploit the narrow frequency separation to parallelize and.ind.pend.ntly control 34 single qubit-gates with a single set of three off-the-shelf electro-optic d.vices. We fully characterize 17 frequency-bin maximally entangled.qubit pairs by performing quantum state tomography. We d.monstrate for the first time, we believe, a fully connected.five-user quantum network in the frequency d.main. These results are a step toward. a generation of quantum circuits implemented.with scalable silicon photonics technology, for applications in quantum computing and.secure communications.

Genomic Profile of SARS-COV-2 Associated.with COVId.19 Outbreaks in N’d.amena, Chad

Background. SARS-CoV-2 has circulated.world.id. with d.amatic consequences. In Chad. we have no d.ta reported.of variants. The aim of this stud. was to id.ntify the SARS-CoV-2 variants that circulated.d.ring the epid.mic from 2020 to 2021. Method.: This is a cross-sectional, d.scriptive stud. carried.out between 2020 and.2021. Samples from patients with suspected.COVId.19 were tested.in five laboratories in N’d.amena. One hund.ed.quality samples of the positives were sequenced.in Kinshasa using Oxford.nanopore technologies minion and.the Protocol Mid.ight SARS-CoV2. d.ta were processed.using Excel version 16 software. Results: Of the 100 samples sequenced. 77 (77%) prod.ced.sequences, 23 (23%) d.d.not. The genomic profiles were wild.type Wuhan and.minor mutations (19A, 19B (A), 20A (B.1, B.2), 20B (AV.1), 20d.(B.1.1.1 /C.36), 20C), variant of concern Alpha (20I), variant of concern d.lta (21A/J), variant of interest Eta (21d., variant of concern Omicron (21K) and.unclassified.variant und.r surveillance (B.1.640). Of these variants, the maximums were d.tected.in patients aged.26 - 35 with 30.26% and.25.26% in 36 - 45. However, 24.67% were in travelers and.75.32% in resid.nts, 35.06% in those vaccinated.against COVId.19 and.62.33% in non-vaccinates. The estimated.case-fatality rate was 2.44% (107/4374). Conclusion: This work has provid.d.preliminary d.ta on COVId.19 and.SARS-CoV-2 variants circulating d.ring the 2020-2021 epid.mics in Chad.

Transient growth in Poiseuille-Rayleigh-Bénard.flows of binary fluid. with Soret effect

The transient growth d.e to non-normMity is investigated.for the Poiseuille- Rayleigh-Benard.problem of binary fluid. with the Soret effect. For negative separation factors such as ψ = -0.1, it is found.that a large transient growth can be obtained.by the non-normal interaction of the two least-stable-mod.s, i.e., the upstream and.d.wnstream mod.s, which d.termine the linear critical bound.ry curves for small Reynold. numbers. The transient growth is so strong that the optimal energy amplification factor G（t） is up to 10^2 - 10^3. While for positive separation factors such as ψ = 0.1, the transient growth is weak with the ord.r O（I） of the amplification factor, which can even be computed.by the least-stable-mod.. However, for both cases, the least-stable-mod. can govern the long-term behavior of the amplification factor for large time. The results also show that large Reynold. numbers have stabilization effects for the maximum amplification within mod.rate wave number regions. Meanwhile, much small negative or large positive separation factors and.large Rayleigh numbers can enlarge the maximum transient growth of the pure streamwise d.sturbance with the wavenumber α= 3.14. Moreover, the initial and.evolutionary two-d.mensional spatial patterns of the large transient growth for the pure streamwise d.sturbance are exhibited.with a plot of the velocity vector, spanwise vorticity, temperature, and.concentration field. The initial three-layer cell vorticity struc- ture is revealed. When the amplification factor reaches the maximum Gmax, it d.velops into one cell structure with large amplification for the vorticity strength.
