Easily-handled method to isolate mesenchymal stem cells from coagulated human bone marrow samples

AIM:To establish an easily-handled method to isolate mesenchymal stem cells(MSCs) from coagulated human bone marrow samples. METHODS: Thrombin was added to aliquots of seven heparinized human bone marrow samples to mimic marrow coagulation. The clots were untreated,treated with urokinase or mechanically cut into pieces before culture for MSCs. The un-coagulated samples and the clots were also stored at 4 ℃ for 8 or 16 h before the treatment. The numbers of colony-forming unit-fibroblast(CFU-F) in the different samples were determined. The adherent cells from different groups were passaged and their surface profile was analyzed with flow cytometry. Their capacities of in vitro osteogenesis and adipogenesis were observed after the cells were exposed to specific inductive ***: The average CFU-F number of urokinasetreated samples(16.85 ± 11.77/106) was comparable to that of un-coagulated control samples(20.22 ± 10.65/106,P = 0.293),which was significantly higher than those of mechanically-cut clots(6.5 ± 5.32/106,P < 0.01) and untreated clots(1.95 ± 1.86/106,P < 0.01). The CFU-F numbers decreased after samples were stored,but those of control and urokinase-treated clots remained higher than the other two groups. Consistently,the numbers of the attached cells at passage 0 were higher in control and urokinase-treated clots than those of mechanically-cut clots and untreated *** attached cells were fibroblast-like in morphology and homogenously positive for CD44,CD73 and CD90,and negative for CD31 and CD45. Also,they could be induced to differentiate into osteoblasts and adipocytes in vitro. CONCLUSION: Urokinase pretreatment is an optimal strategy to isolate MSCs from human bone marrow samples that are poorly aspirated and clotted.

Carbon-supported CeNCl as an efficient catalyst for CO_(2)cycloaddition

The cycloaddition reaction of CO_(2)with epoxide not only effectively reduces the concentration of CO_(2)in the atmosphere,but also has excellent industrial application value and up to 100%atom utilization,but there are difficulties in separation and recovery of traditional homogeneous catalysts,harsh reaction conditions of traditional heterogeneous catalysts,and activation of CO_(2)*** this paper,an easily synthesized heterogeneous catalyst CeNCl/C was used to catalyze the cycloaddition reaction of CO_(2)with styrene oxide,with a high yield of 92.7%,a high selectivity of 96.7%,a turnover numbers(TON)value of 349,and a good stability demonstrated in six cycle tests(equivalent to 216 h of testing).Through comprehensive studies,it was shown that CeNCl/C contains Lewis acid-base centers as active centers,which can effectively reduce the energy barrier required for ring opening of the reaction substrate,enhance the adsorption and activation of CO_(2),and promote the formation of intermediates,which led to the acquisition of excellent catalytic activity.

Unlocking the stable interface in aqueous zinc-ion battery with multifunctional xylose-based electrolyte additives

The growth of dendrites and the side reactions occurring at the Zn anode pose significant challenges to the commercialization of aqueous Zn-ion batteries（AZIBs）. These challenges arise from the inherent conflict between mass transfer and electrochemical kinetics. In this study, we propose the use of a multifunctional electrolyte additive based on the xylose（Xylo） molecule to address these issues by modulating the solvation structure and electrode/electrolyte interface, thereby stabilizing the Zn anode. The introduction of the additive alters the solvation structure, creating steric hindrance that impedes charge transfer and then reduces electrochemical kinetics. Furthermore, in-situ analyses demonstrate that the reconstructed electrode/electrolyte interface facilitates stable and rapid Zn2+ion migration and suppresses corrosion and hydrogen evolution reactions. As a result, symmetric cells incorporating the Xylo additive exhibit significantly enhanced reversibility during the Zn plating/stripping process, with an impressively long lifespan of up to 1986 h, compared to cells using pure ZnSO4electrolyte. When combined with a polyaniline cathode, the full cells demonstrate improved capacity and long-term cyclic stability. This work offers an effective direction for improving the stability of Zn anode via electrolyte design, as well as highperformance AZIBs.

Mutual regulation of microglia and astrocytes after Gas6 inhibits spinal cord injury

Invasive inflammation and excessive scar formation are the main reasons for the difficulty in repairing nervous tissue after spinal cord *** and astrocytes play key roles in the spinal cord injury micro-environment and share a close ***,the mechanisms involved remain *** this study,we found that after spinal cord injury,resting microglia(M0)were polarized into pro-inflammatory phenotypes(MG1 and MG3),while resting astrocytes were polarized into reactive and scar-forming *** expression of growth arrest-specific 6(Gas6)and its receptor Axl were significantly down-regulated in microglia and astrocytes after spinal cord *** vitro experiments showed that Gas6 had negative effects on the polarization of reactive astrocytes and pro-inflammatory microglia,and even inhibited the cross-regulation between *** further demonstrated that Gas6 can inhibit the polarization of reactive astrocytes by suppressing the activation of the Yes-associated protein signaling ***,in turn,inhibited the polarization of pro-inflammatory microglia by suppressing the activation of the nuclear factor-κB/p65 and Janus kinase/signal transducer and activator of transcription signaling *** vivo experiments showed that Gas6 inhibited the polarization of pro-inflammatory microglia and reactive astrocytes in the injured spinal cord,thereby promoting tissue repair and motor function ***,Gas6 may play a role in the treatment of spinal cord *** can inhibit the inflammatory pathway of microglia and polarization of astrocytes,attenuate the interaction between microglia and astrocytes in the inflammatory microenvironment,and thereby alleviate local inflammation and reduce scar formation in the spinal cord.

Pro-resolving lipid mediator reduces amyloid-β42–induced gene expression in human monocyte–derived microglia

Specialized pro-resolving lipid mediators including maresin 1 mediate resolution but the levels of these are reduced in Alzheimer's disease brain, suggesting that they constitute a novel target for the treatment of Alzheimer's disease to prevent/stop inflammation and combat disease pathology. Therefore, it is important to clarify whether they counteract the expression of genes and proteins induced by amyloid-β. With this objective, we analyzed the relevance of human monocyte–derived microglia for in vitro modeling of neuroinflammation and its resolution in the context of Alzheimer's disease and investigated the pro-resolving bioactivity of maresin 1 on amyloid-β42–induced Alzheimer's disease–like inflammation. Analysis of RNA-sequencing data and secreted proteins in supernatants from the monocyte-derived microglia showed that the monocyte-derived microglia resembled Alzheimer's disease–like neuroinflammation in human brain microglia after incubation with amyloid-β42. Maresin 1 restored homeostasis by down-regulating inflammatory pathway related gene expression induced by amyloid-β42 in monocyte-derived microglia, protection of maresin 1 against the effects of amyloid-β42 is mediated by a re-balancing of inflammatory transcriptional networks in which modulation of gene transcription in the nuclear factor-kappa B pathway plays a major part. We pinpointed molecular targets that are associated with both neuroinflammation in Alzheimer's disease and therapeutic targets by maresin 1. In conclusion, monocyte-derived microglia represent a relevant in vitro microglial model for studies on Alzheimer's disease-like inflammation and drug response for individual patients. Maresin 1 ameliorates amyloid-β42–induced changes in several genes of importance in Alzheimer's disease, highlighting its potential as a therapeutic target for Alzheimer's disease.

为了研究毛竹释放的挥发性有机化合物(VOCs)对空气微生物的影响,研究采用动态顶空气体循环吸附法和热脱附-气相色谱-质谱(TDS-GC-MS)联用技术,分析了近自然状态下毛竹叶片释放的VOCs和不同毛竹林内空气VOCs的组分与含量;测定了林内空气负离子及微生物浓度,并进行微生物单体试验来验证挥发物对微生物的抑制作用。结果显示:毛竹单株共检测出挥发物35种,主要物质有醛类(26.2%)、酯类(21.9%)、醇类(18.4%)和萜烯类(16.6%);不同季节毛竹林内VOCs种类和相对含量存在差异,主要表现为:夏>春>秋,春、夏季的醛类分别是秋季的30.3倍和39.8倍;春、夏、秋季的毛竹林内空气负离子浓度分别为1030、1975、547个·cm^(-3);毛竹林中空气微生物的浓度同林缘相比均有明显的降低,春、夏、秋季对细菌抑制率分别为72.4%、61.3%和78.4%(P<0.05),对真菌抑制率为62.9%、59.7%和72%(P<0.05),对放线菌抑制率为:61.9%、58%和72.1%(P<0.05);VOCs单体抑菌试验表明罗勒烯、癸醛、乙酸、叶醇对微生物的抑制作用显著。毛竹释放的VOCs对NAI的形成具有促进作用,对空气微生物生长具有抑制作用。
