Epithelial-mesenchymal transition(EMT) has been implicated in tumor invasion and metastasis and provides novel strategies for cancer therapy. Hypaconitine(HpA), a diester-diterpenoid alkaloid isolated from the root of the Aconitum species, exhibits anti-inflammatory, analgesic, and especially, cardiotoxic activities. Here, we reported the anti-metastatic potentials of HpA in transforming growth factor-β1(TGF-β1)-induced EMT in lung cancer A549 cells. The cytotoxic effect of HpA was determined by MTT assay. A549 cells were treated with TGF-β1 with or without HpA co-treatment, and the morphological alterations were observed with a microscopy. The expression of E-cadherin, N-cadherin, and NF-κB was determined by both Western blotting and immunofluorescence analyses. The adhesion, migration, and invasion were detected with Matrigel, wound-healing, and transwell assays, respectively. The expression of Snail was determined by Western blotting. The expression of NF-κB p65, IκBα, and p-IκBα in nuclear and cytosolic extracts was assessed by Western blotting. The results showed that low concentration of HpA(<16 μmol×L^(–1)) had no obvious cytotoxicity to A549 cells. Morphologically, TGF-β1 treatment induced spindle-shaped alteration in the cells. The upregulation of N-cadherin, NF-κB, and Snail and the downregulation of E-cadherin were detected after TGF-β1 treatment. The adhesion, migration and invasion abilities were also increased by TGF-β1. Besides, TGF-β1 induced expression of Snail in a time-dependent manner. Furthermore, TGF-β1 induced nuclear translocation of NF-κB p65. All these alterations were dramatically inhibited by HpA co-treatment. In addition, the NF-κB inhibitor PDTC showed similar inhibitory effect. In conclusion, these results showed that HpA inhibited TGF-β1-induced EMT in A549 cells, which was possibly mediated by the inactivation of the NF-κB signaling pathway, providing an evidence for anti-cancer effect of HpA.

Sensitivity analysis （SA） is an effective tool for studying crop models; it is an important link in model localization and plays an important role in crop model calibration and application. The objectives were to （i） determine influential and non-influential parameters with respect to above ground biomass （AGB）, canopy cover （CC）, and grain yield of winter wheat in the Beijing area based on the AquaCrop model under different water treatments （rainfall, normal irrigation, and over-irrigation）; and （ii） generate an AquaCrop model that can be used in the Beijing area by setting non-influential parameters to fixed values and adjusting influential parameters according to the SA results. In this study, field experiments were conducted during the 2012-2013,2013-2014, and 2014-2015 winter wheat growing seasons at the National Precision Agriculture Demonstration Research Base in Beijing, China. The extended Fourier amplitude sensitivity test （EFAST） method was used to perform SA of the AquaCrop model using 42 crop parameters, in order to verify the SA results, data from the 2013-2014 growing season were used to calibrate the AquaCrop model, and data from 2012-2013 and 2014-2015 growing seasons were val- idated. For AGB and yield of winter wheat, the total order sensitivity analysis had more sensitive parameters than the first order sensitivity analysis. For the AGB time-series, parameter sensitivity was changed under different water treatments; in comparison with the non-stressful conditions （normal irrigation and over-irrigation）, there were more sensitive parameters under water stress （rainfall）, while root development parameters were more sensitive. For CC with time-series and yield, there were more sensitive parameters under water stress than under no water stress. Soybean seed products contain isoflavones （genistein, daidzein, and glycitein） that display biological effects when ingested by humans and animals. These effects are species, dose and age dependent. Therefore, the content and quality of isoflavones in soybeans is a key factor to the biological effect. Our objective was to identify the genetic effects that underlie the isoflavone content in soybean seeds. A genetic model for quantitative traits of seeds in diploid plants was applied to estimate the genetic main effects and genotype x environment （GE） interaction effects for the isoflavone content （IC） of soybean seeds by using two years experimental data with an incomplete diallel mating design of six parents. Results showed that the IC of soybean seeds was simultaneously controlled by the genetic effects of maternal, embryo, and cytoplasm, of which maternal genetic effects were most important, followed by embryo and cytoplasmic genetic effects. The main effects of different genetic systems on IC trait were more important than environment interaction effects. The strong dominance effects on isoflavone from residual was made easily by environment conditions. Therefore, the improvement of the IC of soybean seeds would be more efficient when selection is based on maternal plants than that on the single seed. Maternal heritability （65.73%） was most important for IC, followed by embryo heritability （25.87%） and cytoplasmic heritability （8.39%）. Based on predicated genetic effects, Yudou 29 and Zheng 90007 were better than other parents for increasing IC in the progeny and improving the quality of soybean, The significant effects of maternal and embryo dominance effects in variance show that the embryo heterosis and maternal heterosis are existent and uninfluenced by environment interaction effects.

We study the structural and dynamical properties of A209 based on Chandra and XMM-Newton *** obtain detailed temperature,pressure,and entropy maps with the contour binning method,and find a hot region in the NW *** X-ray brightness residual map and corresponding temperature profiles reveal a possible shock front in the NW direction and a cold front feature in the SE *** with the galaxy luminosity density map we propose a weak merger scenario.A young sub-cluster passing from the SE to NW direction could explain the optical subpeak,the intracluster medium temperature map,the X-ray surface brightness excess,and the X-ray peak offset together.

Cholesterol-25-hydroxylase(CH25 H)and its enzymatic product 25-hydroxy cholesterol(25 HC)exert broadly antiviral activity including inhibiting HIV-1 ***,their antiviral immunity and therapeutic efficacy in a nonhuman primate model are ***,we report that the regimen of 25 HC combined with antiretroviral therapy(ART),provides profound immunological modulation towards inhibiting viral replication in chronically SIVmac239-infected rhesus macaques(RMs).Compared to the ART alone,this regimen more effectively controlled SIV replication,enhanced SIVspecific cellular immune responses,restored the ratio of CD4/CD8 cells,reversed the hyperactivation state of CD4^(+)T cells,and inhibited the secretion of proinflammatory cytokines by CD4^(^(+))and CD8^(+)T lymphocytes in chronically SIVinfected ***,the in vivo safety and the preliminary pharmacokinetics of the 25 HC compound were assessed in this RM *** together,these assessments help explain the profound relationship between cholesterol metabolism,immune modulation,and antiviral activities by 25 *** results provide insight for developing novel therapeutic drug candidates against HIV-1 infection and other related diseases.
