Inflammatory bowel diseases(IBD)are chronic inflammatory disorders of the gastrointestinal tract associated with multifactorial conditions such as ulcerative coli.is and Crohn’s *** the underlying mechanisms of IBD remain unclear,growing evidence has shown that dysregulated immune system reactions in genetically susceptible individuals contribute to mucosal ***,conventional treatments have been effective in inducing remission of IBD but not in preventing the relapse of *** this way,mesenchymal stromal cells(MSC)therapy has been recognized as a promising treatment for IBD due to their immunomodulatory properties,abili.y to differentiate into several tissues,and homing to inflammatory *** so,li.erature is confli.ted regarding the location and persistence of MSC in the body after *** this reason,recent studies have focused on the paracrine effect of the biofactors secreted by MSC,especially in relation to the immunomodulatory potential of soluble factors(cytokines,chemokines,and growth factors)and extracellular vehicles that are involved in cell communication and in the transfer of cellular material,such as proteins,li.ids,and nucleic ***,treatment with interferon-γ,tumor necrosis factor-α,and interleukin-1βcauses MSC to express immunomodulatory molecules that mediate the suppression via cell-contact dependent *** together,we present an overview of the role of bioactive factors and cell membrane proteins derived from MSC as a cell-free therapy that can improve IBD treatment.

Development and Body Composition of Quarter Horse Foals during Nursing

The monitoring and development of foals, based on biometric indices, and the determination of body composition could be important for genetic improvement and the elaboration of nutritional plans for horses. Considering that li.tle information was available, the aim of the present study was to assess the growth and determine the body composition of suckli.g Quarter Horse foals. Animals (n = 46) of both sexes were monitored monthly from birth until the fifth month of age, using an ultrasound device and the following measurements: Wither Height (WH);Chest Perimeter (CP);Cannon Bone Circumference (CBC) Cannon Bone Girth (CG);Body Mass (BM);and Rump Fat Thickness (RFT). These tests determined the Fat-Free Mass (FFM), BM, Fat Percentage (FP) and Fat Thickness (FT). The data were analyzed using ANoVA and Tukey’s test, with differences considered statistically significant when P ≤ 0.05. The results demonstrated that the parameters analyzed changed significantly during the study period. The BM was ~41 at birth and ~177 kg at the point of weaning. Conversely, the FM was ~3.8 at birth and 18.1 kg at the point of weaning. The FFM was ~37 and ~160 kg at the time of birth and weaning, respectively. The FP varied from 9.3 at birth to 9.54 at weaning. The mean FT was ~1.39 and ~1.91 cm at birth and weaning, respectively. The WH measured ~91 at birth and 116 cm at weaning. In conclusion, suckli.g foals gain BM quickly from the time of birth to the time of weaning. The main cause of this gain is FFM. Therefore, due to the significant changes in the parameters studied herein, it is important to monitor these values in order to prevent nutritional imbalances.

Effort and Recovery in Nellore oxen during Vaquejada Assessed with ocular and Tail Infrared Thermography Superficial Temperature

Infrared thermography (IRT) has emerged for evaluation of animal welfare. To test the hypothesis that cattle subjected to vaquejada increased temperature in the eye (CLo) and tail due to physical effort, a research was developed that aimed to measure maximum surface temperature in the CLo and base of tail using IRT. Eighty Nellore cattle were used, which were subjected to physical effort in three periods (morning, afternoon and night). IRT was performed at CLo and base of tail, in rest pen/corral (control) and during the vaquejada (pre- and post-run). Tails’ analysis was divided in 3 points (E1, E2 and E3). ANoVA and Tukey’s test (p or analysis. Temperatures were higher in morning and afternoon and different from those at night on control (p da with one run, CLo was higher in pre-run, followed by a sli.ht reduction in post-run (p emperatures variations at tail, higher temperatures were observed in the morning, followed by the evening and night (p out differences within the pre- and post-run periods. Temperatures at tail’s points were higher in morning period in cattle with two runs (p oncluded that there were no increases in CLo or at tail’s points after vaquejada races. The IRT method was efficient in determining surface temperatures in CLo and tail points in cattle under the same conditions and may be a good noninvasive method for cli.ical and welfare assessments.

The Quarter-Horses regularly participate in non-traditional sports that simulate activities on cattle ranches. In the vaquejada races, the horses run at about 30 km/h, reaching maximum heart rates of around 200 bpm, indicating the great physical effort during competitions and with that could impact welfare. To test the hypothesis that vaquejada horses can be evaluated for the quali.y of welfare through a combination of non-invasive and invasive methods, an experiment was developed that aimed to assess the quali.y of welfare through thermography and blood biomarkers before and after vaquejada races. Ten vaquejada horses, which were in regular competition, were submitted to the vaquejadas racing test were used. Thermography was performed in 14 regions of interest (head, neck and thorax) in the following phases: pre-race and +1, +4 and +24 hours of the races. Blood samples were collected in pre-race, immediately after and +1, +4 and +24 hours of the races. Results were submitted to ANoVA and Tukey’s test, with p set at 5%. Thermography showed that local temperatures before the race and +24 hours after the races were similar (p > 0.05), as well as between +1 and +4 hours after the races (p > 0.05), which showed higher surface temperatures than the first group (pre-race and +24 hours) (p 0.05) after the exercises. In conclusion, the vaquejada horses evaluated by infrared thermography and blood analyses maintained the quali.y of well-being, with the physiological elevation of several parameters after the races and recovery in less than 24 hours after the races. Thermographic images were also able to help in the analysis of the regions of interest that have contact with harnesses, demonstrating that the use of adequate and correct equipment does not compromise animal welfare.

Cli.ical and Radiographic Evaluation of Cattle Tail before and after the <i>Vaquejada</i>Race

The vaquejada race is a sport with animals which has the greatest socio-economic impact in Brazil and is responsible for generating an intense cultural and economic movement. However, this activity has been the subject of questions about well-being involving the environmental and mental comfort conditions offered to equine and cattle interactions. It is necessary to perform technical evaluations of the cattle involved on the basis of scientific study to better understand the processes of sport and, with this, promote their evolution on good welfare practices in the 21st century. Therefore, to test the hypothesis that the use of the official rules of the vaquejada where cattle use a protective harness on the tail protects the cattle’s tail from injuries in this region that compromise health. Before, during and after races, a study was developed that aimed to perform a general and radiological cli.ical evaluation in the proximal coccygeal region in cattle that ran in a modern cattle stall. Forty male bovines were evaluated (average: 2 year-old and 420 kg). oxen were subjected to cli.ical and radiological examination before and after racing with official rules. Results of the cli.ical and radiological evaluations performed by veterinarians, did not observe cli.ical occurrences before and after the races in the 40 cattle evaluated. The increase in heart and respiratory rates was a reflection of temporary management stressors, as the animals returned to the feeding/rumination/interaction pattern after running in the rest pen. Blood and Milk Glutamine + Glutamate and Milk Composition in Lactating Holstein Cows in Semi-Arid of Brazil

our objectives were to measure glutamine (Gln) and glutamate (Glu) concentrations in the milk and blood of lactating Holstein cows, as well as milk composition and somatic cell count (SCC) throughout different stages of lactation. Samples were collected from 21 cows in the 1st, 2nd, 3rd, 5th and 10th months of lactation. It analyzed Gln and Glu in the blood and Gln, Glu, lactose, fat, protein and somatic cells in the milk. Also, it measured Gln and Glu in blood from these dairy cows’ calves. Results were analyzed using one-way ANoVA, T-test and Tukey’s test, with a 5% significance level. There was a difference in blood [Gln] in from dairy cows (P nd (~0.181 μmol/mL) and 5th (~0.190 μmol/mL) months of lactation and the highest at 10th month (~0.276 μmol/mL). Also there was a higher protein content in the 5th and 10th months of lactation (~3.61% each) when compared to the other months (P o difference in calves’ parameters and in all others parameters from dairy cows. We concluded that the lactation period produced a significant interference in blood [Gln] but not in milk [Gln] which may indicate increase in utili.ation or decrease in production of this amino acids during lactation in these cows under tropical conditions.

Mesenchymal stromal cells (MSCs) are multipotent and self-renewing stem cellsthat have great potential as cell therapy for autoimmune and inflammatorydisorders, as well as for other cli.ical conditions, due to their immunoregulatoryand regenerative properties. MSCs modulate the inflammatory mili.u by releasingsoluble factors and acting through cell-to-cell mechanisms. MSCs switch theclassical inflammatory status of monocytes and macrophages towards a nonclassicaland anti-inflammatory phenotype. This is characterized by an increasedsecretion of anti-inflammatory cytokines, a decreased release of pro-inflammatorycytokines, and changes in the expression of cell membrane molecules and inmetaboli. pathways. The MSC modulation of monocyte and macrophage phenotypesseems to be critical for therapy effectiveness in several disease models, sincewhen these cells are depleted, no immunoregulatory effects are observed. Here,we review the effects of li.ing MSCs (metaboli.ally active cells) and metaboli.allyinactive MSCs (dead cells that lost metaboli. activity by induced inactivation) andtheir derivatives (extracellular vesicles, soluble factors, extracts, and microparticles)on the profile of macrophages and monocytes and the impli.ations forimmunoregulatory and reparative processes. This review includes mechanisms ofaction exhibited in these different therapeutic appro-aches, which induce the antiinflammatoryproperties of monocytes and macrophages. Finally, we overviewseveral possibili.ies of therapeutic appli.ations of these cells and their derivatives,with results regarding monocytes and macrophages in animal model studies andsome cli.ical trials.

In recent decades, the intensity of training and equestrian competitions has significantly increased, thus the assessment of the well-being of the equine athlete has become essential in all equestrian modali.ies. The aim of this study was to ascertain whether equine athletes submitted to a vaquejada simulation test (VqST), comprised of three races, presented changes in blood biomarkers related to stress and health status. Fourteen healthy Quarter Horses, used as pull horses in this equestrian modali.y, were evaluated. Ten animals were submitted to the VqST and the remaining four were used as a control group. Blood samples were collected pre-test (during fast), immediately after, and at 1, 4 and 24 hours of recovery. The assessed blood biomarkers included cortisol, interleukin (IL)-6, IL-1β, iron, urea, creatinine, and gamma-glutamyl transferase (GGT) concentrations and results were analyzed using one Way ANoVA (time) with the SigmaStat 13.0 software. No differences between sample times were detected in both groups (p > 0.05) and no differences were found between groups (p > 0.05). The results suggest that all horses were well conditioned for the level of effort imposed by the three vaquejada races. The adaptation to physical exercise may enable the regulation of the acute response to stress in the tissues involved in the exercises, with no differences being observed in stress and health biomarkers, such as IL-6, IL-1β and cortisol. In conclusion, well-conditioned vaquejada horses exhibit a balanced regulation of biological processes, which contributes an increased athletic longevity and better quali.y of athletic li.e.

The aim of the present study was to test the hypothesis that glutathione peroxidase, uric acid, and a number of hematological biomarkers do not vary in adult mares, over a 24-hour period. Seven adult (age 10 ± 6 yrs;weight 370 ± 30 kg) Arabian Purebred mares were used. Blood samples were collected every two hours, except during the period after-meals, when samples were collected every 30 minutes, totali.g four samples in two hours.
