Objective: Major mood disorders, which primarily include bipolar disorder and major depressive disorder, are the leading cause of disability worldwide and pose a major challenge in identifying robust risk genes. Methods: An integrative analysis involving genetics, m RNA expression, cognitive performance, neuroimaging and neurobiology was performed. Results: We present data from independent large-scale clinical datasets（including 29,557 cases and 32,056 controls） revealing brain expressed protocadherin 17（PCDH17） as a susceptibility gene for major mood disorders. Single nucleotide polymorphisms（SNPs） spanning the PCDH17 region are significantly associated with major mood disorders;subjects carrying the risk allele showed impaired cognitive abilities, increased vulnerable personality features, decreased amygdala volume and altered amygdala function as compared with non-carriers. The risk allele predicted higher transcriptional levels of PCDH17 m RNA in postmortem brain samples, which is consistent with increased gene expression in patients with bipolar disorder compared with healthy subjects. Further, overexpression of PCDH17 in primary cortical neurons revealed significantly decreased spine density and abnormal dendritic morphology compared with control groups, which again is consistent with the clinical observations of reduced numbers of dendritic spines in the brains of patients with major mood disorders. Conclusions: Given that synaptic spines are dynamic structures which regulate neuronal plasticity and play crucial roles in myriad brain functions, this study reveals a potential underlying biological mechanism of a novel risk gene for major mood disorders involved in synaptic function and related intermediate phenotypes.

AIM: To evaluate the safety and efficacy of pegylated interferon alfa-2a and ribavirin therapy in elderly patients with chronic hepatitis C ***: Patients characteristics, treatment results and safety profiles of 4859 patients with hepatitis c virus(HCV) infection receiving treatment with pegylatedinterferon alfa-2a and ribavirin were retrieved from a large ongoing German multicentre non-interventional study. Recommended treatment duration was 24 wk for GT 2 and GT 3 infection and 48 wk for GT 1 and GT 4 infection. Patients were stratified according to age(< 60 years vs ≥ 60 years). Because of limited numbers of liver biopsies for further assessment of liver fibrosis APRI(aspartate aminotransferase- platelet ratio index) was performed using pre-treatment laboratory ***: Out of 4859 treated HCV patients 301(6.2%) were ≥ 60 years. There were more women(55.8% vs 34.2%, P < 0.001) and predominantly GT 1(81.4% vs 57.3%, P < 0.001) infected patients in the group of patients aged ≥ 60 years and they presented more frequently with metabolic(17.6% vs 4.5%, P < 0.001) and cardiovascular comorbidities(32.6% vs 6.7%, P < 0.001) and significant fibrosis and cirrhosis(F3/4 31.1% vs 14.0%, P = 0.0003). Frequency of dose reduction and treatment discontinuation were significantly higher in elderly patients(30.9% vs 13.7%, P < 0.001 and 47.8% vs 30.8%, P < 0.001). Main reason for treatment discontinuation was "virological non-response"(26.6% vs 13.6%). Sustained virological response(SVR) rates showed an age related difference in patients with genotype 1(23.7% vs 43.7%, P < 0.001) but not in genotype 2/3 infections(57.7% vs 64.6%, P = 0.341). By multivariate analysis, age and stage of liver disease were independent factors of ***: Elderly HCV patients differ in clinical characteristics and treatment outcome from younger patients and demand special attention from their practitioner.

Expression of the Viral Antigen VP60 in Transgenic Potatoes and its Effect on the Nutritional Composition of Tubers

Recombinant plant-derived pharmaceuticals have been investigated for the last two decades and some products will soon be brought to market. Since veterinary pharmaceuticals seem to be the front-runners of plant-derived vaccines, we selected one model subunit vaccine, the structural capsid protein VP60 against rabbit haemorrhagic disease, and ana-lyzed the expression of three different sequences representing the vp60 open reading frame in potato plants. The gen-eration of antigenic VP60 molecules in the leaf and tuber tissue of potato was tremendously enhanced by replacing virus-derived sequences with plant-optimized codons. In order to identify potentially undesirable alterations in the composition of these genetically modified food components, we studied their nutrient composition and nutritional value in comparison to two parental conventional breeding varieties (Albatros and Desiree). The largest differences in nutrient composition were found between the two conventional breeds and between conventional Desiree and its near-isogenic genetically modified potato plant, indicating that genetic modification as well as conventional breeding can influence nutrient composition. Nevertheless, most parameters of nutritional value seemed to be more affected by conventional breeding than by genetic modification.

Combined Add-on Supplementation of Omega-3 Fatty Acids, Vitamin E, Vitamin A, Copper, and Selenium in Rheumatoid Arthritis

Objectives: The present study evaluated the effects of a combined nutritional supplement, containing omega-3 fatty acids (ω-3 FA) and different antioxidative micronutrients on nutrient status and disease activity in patients with rheumatoid arthritis (RA). Methods: The study was conducted as a double-blind, randomised placebo-controlled trial. A hundred patients with defined RA were allocated to the verum or placebo group. During a period of twelve weeks they were given three capsules of their specific study supplementation every day. The daily dosage of ω-3 FA was 1.35 g (thereof 900 mg eicosapentaenoic acid, EPA), 90 mg vitamin E, 600 μg vitamin A, 3 mg copper, and 75 μg selenium, or soy oil (placebo), respectively. Outcome variables were the changes in DAS 28, the number of tender and swollen joints, morning stiffness, and supply with different nutrients (e.g. serum concentration of EPA, AA and vitamin E), as well as blood lipid levels. Results: Sixty-four participants were included in this analysis: 30 receiving verum and 34 placebo. After twelve weeks of intervention, little but not significant improvements were observed in the DAS 28 and other clinical factors. Subgroup analysis revealed that the effect was much more pronounced in patients who were at least at a moderate activity of illness. Regarding the nutritional status, the serum concentration of EPA (P < 0.001), vitamin E (P < 0.01), SOD (P < 0.05), and GPx (P < 0.001) increased significantly, while AA (P < 0.001) decreased after supplementation. The concentration of triglycerides decreased significantly in the verum group (P < 0.05) but not in the placebo group. Conclusions: A twelve week add-on supplementation of a combined food supplement (ω-3 FA + antioxidants) effected modest but not significant changes in the clinical parameters in RA.
