Peri-implant gas accumulation in response to magnesium-based musculoskeletal biomaterials:Reframing current evidence for preclinical research and clinical evaluation

Historically,the rapid degradation and massive gas release from magnesium(Mg)implants resulted in severe emphysema and mechanical *** the advent of new alloys and surface treatment methods,optimized Mg implants have re-entered clinics since last decade with reliable ***,the optimization aims at slowing down the degradation process,rather than exemption of the gas *** study involved a systematic evaluation of current preclinical and clinical evidence,regarding the physical signs,symptoms,radiological features,pathological findings and complications potentially associated with peri±implant gas accumulation(PIGA)after musculoskeletal Mg *** literature search identified 196 potentially relevant publications,and 51 papers were enrolled for further analysis,including 22 preclinical tests and 29 clinical studies published from 2005 to *** Mg-based materials have been evaluated in animal research,and the application of pure Mg and Mg alloys have been reported in clinical follow-ups involving multiple anatomical sites and musculoskeletal *** tissue and intraosseous PIGA are common in both animal tests and clinical follow-ups,and potentially associated with certain adverse *** examinations especially micro-CT and clinical CT scans provide valuable information for quantitative and longitudinal *** according to simulation tests involving Mg implantation and chemical processing,tissue fixation could lead to an increase in the volume of gas cavity,thus the results obtained from ex vivo imaging or histopathological evaluations should be interpreted with *** still lacks standardized procedures or consensus for both preclinical and clinical evaluation of ***,by providing focused insights into the topic,this evidence-based study will facilitate future animal tests and clinical evaluations,and support developing biocompatible Mg implants for the treatment of musculoskeletal

The clinical application of magnesium(Mg)and its alloys for bone fractures has been well supported by in vitro and in vivo ***,there were studies indicating negative effects of high dose Mg intake and sustained local release of Mg ions on bone metabolism or repair,which should not be ignored when developing Mg-based ***,it remains necessary to assess the biological effects of Mg implants in animal models relevant to clinical treatment *** primary purpose of this study was to validate the beneficial effects of intramedullary Mg implants on the healing outcome of femoral fractures in a modified rat *** addition,the mineralization parameters at multiple anatomical sites were evaluated,to investigate their association with healing outcome and potential clinical *** to the control group without Mg implantation,postoperative imaging at week 12 demonstrated better healing outcomes in the Mg group,with more stable unions in 3D analysis and high-mineralized bridging in 2D *** bone tissue mineral density(TMD)was higher in the Mg group at the non-operated femur and lumbar vertebra,while no differences between groups were identified regarding the bone tissue volume(TV),TMD and bone mineral content(BMC)in *** the surgical femur,the Mg group presented higher TMD,but lower TV and BMC in the distal metaphyseal region,as well as reduced BMC at the osteotomy *** component analysis(PCA)-based machine learning revealed that by selecting clinically relevant parameters,radiological markers could be constructed for differentiation of healing outcomes,with better performance than 2D *** study provides insights and preclinical evidence for the rational investigation of bioactive materials,the identification of potential adverse effects,and the promotion of diagnostic capabilities for fracture healing.

Background: The endocannabinoid (EC) system is well characterized in the central nervous system but scarcely studied in peripheral organs. In this paper, we newly identify the effect of the EC anandamide (AEA) upon renal proximal tubule cells. Methods: Measurement of lactate dehydrogenase (LDH) release after treatment of primary renal proximal tubule cells (RPTEC) and renal carcinoma cell line (Caki-1) with AEA, arachidonic acid (AA), ethanolamide (EtAm), EC receptor CB1 antagonist (AM251), CB2 receptor antagonist (SR144528), TRPV1 receptor antagonist (capsazepine), degradation enzyme fatty acid amide hydrolase (FAAH) antagonist (URB597), antioxidants GSH-EE;Trolox, GSH depletor BSO, membrane cholesterol depletor (MCD), apoptosis inhibitor zVAD, necroptosis inhibitor Nec-1 or ferroptosis inhibitor Fer-1. Western blot and qRT-PCR analysis plus determination of reactive oxygen species (ROS) via H2-DCFDA were performed. Histology for EC enzymes, N-acetylphosphatidylethanolamine-hydrolyzing phospholipase D (NAPE-PLD) and FAAH, as well as the determination of physiological levels of ECs in human and rat renal tissue via liquid chromatography were conducted. Results: AEA both dose- and time-dependently induces cell death in RPTEC and Caki-1 within hours, characterized by cell blebbing, not influenced by blocking the described EC receptors by AM251, SR144528, capsazepine or FAAH by URB597 or MCD. Cell death is mediated via ROS. There is no difference found in the histology of the enzymes FAAH and NAPE-PLD in human renal tissue with interstitial nephritis. Blocking of apoptotic, necroptotic or ferroptotic cell death does not lead to a reduction in LDH release in vitro. Conclusion: The endocannabinoid anandamide induces cell death in renal proximal tubule cell in a time- and dose-dependent manner. This pathway is mediated via ROS and is independent of cannabinoid receptors, membrane cholesterol or FAAH activity.

Effects of proteins on magnesium degradation-static *** conditions

The interaction between organic molecules and biomaterial surfaces determines the fate of biomaterials during their service life,which is also the research hotspots in the field of *** understand the mechanism of protein interaction with magnesium(Mg)degradation,alloying elements,immersion time,protein concentration and surface conditions have been previously considered for the effect of proteins on Mg ***,fluid flow,as one of the critical factors,drew little attention in this *** the present study,the effect of bovine serum albumin(BSA)and fetal bovine serum(FBS)on Mg degradation was compared under static and dynamic *** results revealed that both BSA and FBS slightly decreased the degradation rate of Mg in Hanks’balanced salt solution(HBSS)under static immersion due to the protein adsorption and the formation of a Ca/P-rich top layer on Mg surface,whereas under dynamic flow condition the degradation of Mg was significantly accelerated in the presence of BSA or *** reasons seemed to stem from the weakened protein adsorption on Mg surface in this case and the dynamically enhanced interaction between proteins and ions/products in solutions,which largely weaken the combination of the top Ca/P-rich layer with the inner corrosion product *** results highlight the importance of testing conditions for Mg characterization in vitro and the synergistic effect between different parameters on Mg degradation.

Textile wastewater contains highly toxic,nondegradable,carcinogenic organic pollutants,causing severe damage to the ecological *** this work,β-cyclodextrin carbon-sphere-based nanocomposite(CS/SiO_(2)@β-CD)with a core-shell-shell configuration was prepared by hydrothermal synthesis and subsequent sedimentation-azeotropic distillation with SiO_(2)as intermediate layer for the removal of crystal violet(CV)and bisphenol A(BPA).The chemical structure and morphology of the CS/SiO_(2)@β-CD were characterized by FT-IR,SEM and *** effects of contact time,pH,temperature and initial concentration of the absorbates on the adsorption property were *** CS/SiO_(2)@β-CD showed excellent adsorption capacity on CV and BPA,and the maximum adsorption capacity was 87.873 mg g^(-1)and 165.095 mg g^(-1),*** adsorption process was in line with the pseudo-second-order and pseudo-first-order kinetic models,*** adsorption thermodynamics could be better fitted by Langmuir model than by Freundlich *** calculated thermodynamic parameters(-ΔG0,ΔS0 andΔH0)illustrated the adsorption of CV was endothermic,in contrast,the adsorption of BPA was exothermic ***,the CS/SiO_(2)@β-CD showed excellent recovery performance,making it a potential adsorbent for the practical treatment of textile wastewater.

Precipitation of metal oxide nanoparticles using a miniemulsion technique

A method to precipitate nanoparticles using a miniemulsion technique is described, in which miniemulsion droplets between 100 and 1000 nm in size serve as nanoreactors enabling both the control of particle formation and particle growth. The application of miniemulsion droplets to synthesise nanoparticles comprises three advantages： first, the size of the precipitated particles is limited by the reactant concentration within the emulsion droplet; second, particle agglomeration is prevented as nanoparticle collision outside the nanoreactor is avoided; and third, easy technical scale up can be realized by increasing emulsion volume and thus the number of nanoreactors, while local conditions within the reactors are not changed, The miniemulsion technique is an easy scalable process which allows defined synthesis of particles by precipitation reactions. The miniemulsion technique involves first the preparation of a stable water-in- oil miniemulsion by high pressure homogenisation. Whereas a water soluble reactant is provided within the aqueous droplets, another oilas well as water-soluble reactant can be introduced to the emulsion after homogenisation. The precipitation reaction is induced by the diffusion of the second reactant into the emulsion droplet. Together with this contribution, a method is described and discussed which uses a high pressure homogenisation process to produce stable water-in-oil miniemulsions serving as a reaction medium to precipitate metal oxides.
