Ex vivo expansion of tumor-infiltrating lymphocytes from nasopharyngeal carcinoma patients for adoptive immunotherapy

Establishing Epstein-Barr virus (EBV)-specific cytolytic T lymphocytes (EBV-CTLs) from peripheral blood mononuclear cells (PBMCs) for adoptive immunotherapy has been reported in EBV-associated malignancies including Hodgkin's lymphoma and nasopharyngeal carcinoma (NPC). In the current study,we performed ex vivo expansion of tumor-infiltrating lymphocytes (TILs) obtained from NPC biopsy specimens with a rapid expansion protocol using anti-CD3 monoclonal antibody (OKT3), recombinant human interleukin (IL)-2, and irradiated PBMCs from healthy donors to initiate the growth of TILs. Young TIL cultures comprised of more than 90% of CD3+T cells, a variable percentage of CD3+CD8+and CD3+CD4+T cells, and less than 10% of CD3-CD16+natural killer cells, a similar phenotype of EBV-CTL cultures from PBMCs. Interestingly, TIL cultures secreted high levels of the Th1 cytokines, interferon gamma (IFNγ) and tumor necrosis factor-alpha (TNF-α), and low levels of the Th2 cytokines, IL-4 and IL-10. Moreover, young TILs could recognize autologous EBV-transformed B lymphoblast cell lines, but not autologous EBV-negative blast cells or allogeneic EBV-negative tumor cells. Taken together, these data suggest that ex vivo expansion of TILs from NPC biopsy tissue is an appealing alternative method to establish T cell-based immunotherapy for NPC.

We investigated the nonsequential double ionization(NSDI)in Ar by two-color elliptically polarized laser field with a three-dimensional(3D)classical ensemble *** study the relative phase effect of NSDI and distinguish two particular recollision channels in NSDI,which are recollision–impact ionization(RII)and recollision-induced excitation with subsequent ionization(RESI),according to the delay-time between the recollision and the final double *** numerical results indicate that the ion momentum distribution is changed and the triangle structure is more obvious with the decrease of the relative *** also demonstrate that the RESI process always dominates in the whole double ionization process and the ratio of RESI and RII channels can be influenced by the relative phase.

目的:常规的地基抗液化技术成本较高或实施复杂,因此有大量的工程建设未得到有效的抗液化处理。本文旨在提出一种新型的、基于微生物诱导减饱和作用的地基液化治理技术,期望能应用到普通的建筑和结构的抗液化治理中。创新点:1.获得一种基于微生物反硝化过程的砂土减饱和方法;2.试验验证微生物减饱和法处理砂土的抗液化性能,获得微生物减饱和法处理砂土中的气泡分布形态以及气泡在静水和渗流条件下的稳定性规律。方法:1.利用微生物反硝化过程中产生的氮气降低饱和液化砂土的饱和度;2.利用电脑断层扫描(CT)技术研究微生物减饱和法处理的砂土中气泡的分布形态;3.利用振动台实验和三轴固结不排水试验研究微生物减饱和法处理的砂土的力学性能和抗液化性能;4.采用一种自行研制的模型试验(图13)研究微生物减饱和法处理的砂土在静水和渗流条件下气泡的稳定性。结论:1.利用微生物反硝化过程可以实现可控的砂土饱和度降低;2.微生物气泡在砂土中以小空隙的形态存在,小空隙的尺寸略大于砂土的平均粒径;3.将饱和松砂的饱和度略作降低,其不排水强度和抗液化性能得到显著提升;4.微生物减饱和法处理的砂土中的气泡在静水条件下是稳定的,但是在稳定渗流条件下会被水流缓慢带走,此时,需要采用一些手段来控制通过减饱和区的渗流。

Relationship of serum polyunsaturated fatty acids with cytokines in colorectal cancer

AIM: To investigate the relationship of serum levels of polyunsaturated fatty acid(PUFA) with kinds of cytokines in colorectal cancer(CRC).METHODS: Serum samples of 100 CRC patients were collected. The concentration of total n-3 PUFA which included C18:3 n-3, C20:5 n-3, C22:5 n-3, C22:6 n-3 and the total n-6 PUFA included C18:2 n-6, C18:3 n-6, C20:3 n-6, C20:4 n-6, and C22:5 n-6 were detected onGC-2010 Plus Gas Chromatograph with a Omegawax TM 250 column. Cytokines were detected by Mag Plex TM-C microspheres. P values for the trend were estimated by creating a continuous variable using the median value within ***: Interleukin-6(IL-6) showed significantly positive association with the C20:4 n-6(P for trend = 0.004). Interferon gamma(IFN-γ) showed significant positive association with the C22:5 n-3(P for trend = 0.035). IL-8 and matrix metalloproteinase-9(MMP-9) showed significant inverse association with the C22:6 n-3(P for trend = 0.049, and 0.021). MMP-2 showed significant inverse association with the C20:5 n-3(P for trend = 0.008). MMP-7 showed significantly positive association with the ratio of n-6 PUFA and n-3 PUFA(P for trend = 0.008). MMP-7 also showed significantly inverse association with the ratio of C20:4 n-6 and(n-6 PUFA + n-3 PUFA)(P for trend = 0.024). IL-10(P for trend = 0.023) and IL-6(P for trend = 0.036) showed significantly positive association with the ratio of C20:4 n-6 and C20:5 ***: Our data suggested that serum levels of PUFA is related to the inflammation of CRC, and also play different role in regulation of immune response.

High-dense nanocrystalline W-based alloy prepared by hot isostatic pressing with uniform microstructure, ultrahigh hardness and good thermal stability

High-density uniform microstructure nanocrystalline W-10wt.% Cr-5wt.% Y（W-10Cr-5Y） alloy with an average grain size of 85 nm and a relative density of 99.8% was successfully prepared.W-Cr-Y alloys with different amounts of second-phase particles were prepared by high-energy ball milling and hot isostatic pressing *** effects of composition design,ball milling parameters,and sintering parameters on the microstructure of W-Cr-Y alloys were studied.A feasible technology for controlling the synthesis of high-density nanocrystalline W-based alloys has been *** microstructure analysis of nanocrystalline W-10Cr-5Y alloy shows that there are two types of doping in the second phase:intergranular doping and intragranular *** "uniform distribution YOx-rich particles" effectively suppresses the grain growth of W and leads to the formation of nanocrystalline W-10Cr-5Y alloy.W-10Cr-5Y has an ultra-high hardness,with a Vickers hardness of 1312±12 kgf·***-temperature annealing treatment shows that W-10Cr-5Y has good thermal stability at 1000 and1100℃,as demonstrated by the comparison of Vickers hardness and grain size between annealed and unannealed alloys.

Using proximal sensor data for soil salinity management and mapping

Over the past five decades, increased pressure caused by the rapidly growing population has resulted in a reclamation of agricultural and urban buffer zones along China's coastline. However, information about the spatio–temporal variation of soil salinity in these reclaimed regions is limited. As such, obtaining this information is crucial for mapping the variation in saline areas and to identify suitable salinity management strategies. In this study, we employed EM38 data to conduct digital soil mapping of spatio–temporal variation and map these variations of different site-specific zones. The results indicated that the distribution of soil salinity was heterogeneous in the middle of, and that the leaching of salts was significant at the edges of, the study field. Afterwards, fuzzy-k means algorithm was used to divide the site-specific management zones within the time series apparent soil electrical conductivity(ECa) data and the spatial correlations of variation. We concluded that two management zones are optimal to guide precision management. Zone A had an average salinity level of about 165 mS m–1, in which salt-tolerant crops, such as cotton and barley can grow normally, while crops such as soybean and cowpeas may be planted using leaching and increasing the mulching film methods to reduce the accumulation of salt in surface soil. In Zone B, there was a low salinity level with a mean of 89 mS m–1 for ECa, which allows for rice, wheat, and a wide range of vegetables to be grown normally. In such situations, measures such as an optimized combination of irrigation and drainage, as well as soil amendment can be taken to adjust and control the salt content. Particularly, flattening the land with a large-scale machine was used to improve the ability of micro-topography to influence salt migration; rice and other dry, land crops were planted in rotation in combination with utilizing salt-leaching multiple times to speed up desalinization.
