The effects and potential of microglial polarization and crosstalk with other cells of the central nervous system in the treatment of Alzheimer’s disease

Microglia are resident immune cells in the central nervous system. During the pathogenesis of Alzheimer’s disease, stimulatory factors continuously act on the microglia causing abnormal activation and unbalanced phenotypic changes;these events have become a significant and promising area of research. In this review, we summarize the effects of microglial polarization and crosstalk with other cells in the central nervous system in the treatment of Alzheimer’s disease. Our literature search found that phenotypic changes occur continuously in Alzheimer’s disease and that microglia exhibit extensive crosstalk with astrocytes, oligodendrocytes, neurons, and penetrated peripheral innate immune cells via specific signaling pathways and cytokines. Collectively, unlike previous efforts to modulate microglial phenotypes at a single level, targeting the phenotypes of microglia and the crosstalk with other cells in the central nervous system may be more effective in reducing inflammation in the central nervous system in Alzheimer’s disease. This would establish a theoretical basis for reducing neuronal death from central nervous system inflammation and provide an appropriate environment to promote neuronal regeneration in the treatment of Alzheimer’s disease.

Immunoassay of chemical contaminants in milk:A review

The detection of chemical contaminants is critical to ensure dairy safety. These contaminants include veterinary medicines, antibiotics, pesticides, heavy metals, mycotoxins, and persistent organic pollutants （POPs）. Immunoassays have recently been used to detect contaminants in milk because of their simple operation, high speed, and low cost. This article describes the latest developments in the most important component of immunoassays--antibodies, and then reviews the four major substrates used for immunoassays （i.e., microplates, membranes, gels, and chips） as well as their use in the detection of milk contaminants. The paper concludes with prospects for further aDDlications of these immunoassavs.

Spatio-Temporal Changes in the Rice Planting Area and Their Relationship to Climate Change in Northeast China: A Model-Based Analysis

Rice is one of the most important grain crops in Northeast China （NEC） and its cultivation is sensitive to climate change. This study aimed to explore the spatio-temporal changes in the NEC rice planting area over the period of 1980-2010 and to analyze their relationship to climate change. To do so, the CLUE-S （conversion of land use and its effects at small region extent） model was ifrst updated and used to simulate dynamic changes in the rice planting area in NEC to understand spatio-temporal change trends during three periods： 1980-1990, 1990-2000 and 2000-2010. The changing results in individual periods were then linked to climatic variables to investigate the climatic drivers of these changes. Results showed that the NEC rice planting area expanded quickly and increased by nearly 4.5 times during 1980-2010. The concentration of newly planted rice areas in NEC constantly moved northward and the changes were strongly dependent on latitude. This conifrmed that climate change, increases in temperature in particular, greatly inlfuenced the shift in the rice planting area. The shift in the north limit of the NEC rice planting area generally followed a 1＆#176;C isoline migration pattern, but with an obvious time-lag effect. These ifndings can help policy makers and crop producers take proper adaptation measures even when exposed to the global warming situation in NEC.

电力电子化电力系统的宽频振荡问题严重危害电网的安全稳定运行,及时确定宽频振荡的传播路径和振荡源位置,对振荡的抑制至关重要。该文从因果关系的角度提出一种基于Copula传递熵的设备级和网络级宽频振荡传播路径分析及振荡源定位方法,通过分析多变量之间的因果传递方向和因果强度系数,利用有向加权图构建宽频振荡因果网络,在此基础上确定振荡源位置,并通过保留因果强度系数最大的支路,确定振荡的主要传播路径。采用所提方法一方面能够从设备级分析控制器内部各状态变量的因果关联性;另一方面能够从网络级分析电网中各节点振荡数据的因果传递性,从而确定宽频振荡在控制器内部和电网中的传播路径及振荡源位置。最后,在强迫振荡、风机与弱电网交互、发电机轴系振荡中的仿真算例表明,所提方法适用于多种机理的宽频振荡,能够同时实现设备级和网络级宽频振荡传播路径分析和振荡源定位。

Early-onset epilepsy is a neurological abnormality in childhood, and it is especially common in the first2 years after birth. Seizures in early life mostly result from structural or metabolic disorders in the brain, and the genetic causes of idiopathic seizures have been extensively investigated. In this study, we identified four missense mutations in the SETD1 A gene(SET domain-containing 1 A, histone lysine methyltransferase): three de novo mutations in three individuals and one inherited mutation in a four-generation family. Whole-exome sequencing indicated that all four of these mutations were responsible for the seizures. Mutations of SETD1 A have been implicated in schizophrenia and developmental disorders, so we examined the role of the four mutations(R913 C, Q269 R, G1369 R, and R1392 H) in neural development. We found that their expression in mouse primary cortical neurons affected excitatory synapse development. Moreover, expression of the R913 C mutation also affected the migration of cortical neurons in the mouse *** further identified two common genes(Neurl4 and Usp39) affected by mutations of SETD1 A. These results suggested that the mutations of SETD1 A play a fundamental role in abnormal synaptic function and the development of neurons, so they may be pathogenic factors for neurodevelopmental disorders.

We report a reconstruction method for fast-fission events in 25 MeV/u^(86)Kr +^(208)Pb reactions at the Compact Spectrometer for Heavy Ion Experiment(CSHINE). The fission fragments(FFs) are measured using three large-area parallel-plate avalanche counters, which can deliver the position and arrival timing information of the fragments. The start timing information is provided by the radio frequency of the cyclotron. Fission events were reconstructed using the velocities of the two FFs. The broadening of both the velocity distribution and azimuthal difference of the FFs decreases with the folding angle, in accordance with the picture that fast fission occurs. The anisotropic angular distribution of the fission axis also consistently reveals the dynamic features of fission events.

Construction of Salmonella Pullorum ghost by co-expression of lysis gene E and the antimicrobial peptide SMAP29 and evaluation of its immune efficacy in specific-pathogen-free chicks

In this study, a safety enhanced Salmonella Pullorum （S. Pullorum） ghost was constructed using an antimicrobial peptide gene, and evaluated for its potential as a Pullorum disease （PD） vaccine candidate. The antimicrobial peptide SMAP29 was co-expressed with lysis gene E to generate S. Pullorum ghosts. No viable bacteria were detectable either in the fermentation culture after induction of gene E- and SMAP29-mediated lysis for 24 h or in the lyophilized ghost products. Specific-pathogen- free （SPF） chicks were intraperitoneally immunized with ghosts at day 7 of age and no mortality, clinical symptoms or signs of PD such as anorexia, depression and diarrhea were observed. On challenge with a virulent S. Pullorum strain at 4 wk post-immunization, a comparatively higher level of protection was observed in the S. Pullorum ghost immunized chickens with a minimum of pathological lesions and bacterial loads compared to the birds in inactivated vaccine groups. In addition, immunization with the S. Pullorum ghosts induced a potent systemic IgG response and was associated with significantly increased levels of cytokine IFN-y and IL-4 and relative percentages of CD4＋ and CD8＋ T lymphocytes. Our results indicate that SMAP29 can be employed as a new secondary lethal protein to enhance the safety of bacterial ghosts, and to prepare a non-living bacterial vaccine candidate that can prevent PD in chickens.
