This work investigated the gradient microstructure evolution and tensile property of LPBF fabricated 15-5 precipitation hardening stainless steel in post-process direct ageing(DA)and solution treating&ageing(STA).The varied microstructures for austenite and small-sized oxide inclusions at different sample heights in the as-built(AB)condition was generally preserved after DA ***,austenite was almost disappeared,and oxide particle grew significantly after the STA *** a result,the tensile property differences in sample top and bottom for AB and DA conditions did not occur in the STA *** the influence of post-process heat treatment,the STA condition had the highest yield strength due to the highest volume fraction of nano-sized Cu ***,the DA specimen had the highest ultimate tensile strength and elongation owing to the considerable amount of austenite phase and associated transformation induced plasticity effect.

To detect radioactive substances with low activity levels,an anticoincidence detector and a high-purity germanium(HPGe)detector are typically used simultaneously to suppress Compton scattering background,thereby resulting in an extremely low detection limit and improving the measurement ***,the complex and expensive hardware required does not facilitate the application or promotion of this ***,a method is proposed in this study to discriminate the digital waveform of pulse signals output using an HPGe detector,whereby Compton scattering background is suppressed and a low minimum detectable activity(MDA)is achieved without using an expensive and complex anticoincidence detector and *** electric-field-strength and energy-deposition distributions of the detector are simulated to determine the relationship between pulse shape and energy-deposition location,as well as the characteristics of energy-deposition distributions for fulland partial-energy deposition *** relationship is used to develop a pulse-shape-discrimination algorithm based on an artificial neural network for pulse-feature *** accurately determine the relationship between the deposited energy of gamma(γ)rays in the detector and the deposition location,we extract four shape parameters from the pulse signals output by the *** learning is used to input the four shape parameters into the ***,the pulse signals are identified and classified to discriminate between partial-and full-energy deposition *** partial-energy deposition events are removed to suppress Compton *** proposed method effectively decreases the MDA of an HPGeγ-energy dispersive *** results show that the Compton suppression factors for energy spectra obtained from measurements on ^(152)Eu,^(137)Cs,and ^(60)Co radioactive sources are 1.13(344 keV),1.11(662 keV),and 1.08(1332 keV),respectively,and that the corresponding MDAs are 1.4%,5.

Anatolia is the global archetype of tectonic escape,as witnessed by the devastating 2023 Kahramanmaraş Earthquake sequence,and the 2020 Samos Earthquake,which show different kinematics related to the framework of the escape *** Positioning System(GPS)motions of the wedge-shaped plate differ regionally from northwestwards to southwestwards(from east to west).Anatolia was extruded westward from the Arabian-Eurasian collision along the North and East Anatolian fault systems,rotating counterclockwise into the oceanic free-faces of the Mediterranean and Aegean,with dramatic extension of western Anatolia in traditional ***,which is the dominant mechanism for this change in kinematics,extrusion related to the Arabia/Eurasia collision or rollback of the African slab beneath western Anatolia is still *** assess the dominant driving mechanisms across Anatolia,we analyze recent GPS velocity datasets,and decomposed them into N-S and E-W components,revealing that westward motion is essentially constant across the whole plate and consistent with the slip rates of the North and East Anatolia fault zones,while southward components increase dramatically in the transition area between central and western Anatolia,where a slab tear is *** phenomenon is related to different tectonic driving *** ArabiaEurasia collision drives the Anatolian Plate uniformly westwards while western Anatolia is progressively more affected by the southward retreating African subducting slab west of the Aegean/Cypriot slab tear,which significantly increases the southward component of the velocity field and causes the apparent curve of the whole modern velocity *** 2020 and 2023 earthquake focal mechanisms also confirm that the northward colliding Arabian Plate forced Anatolia to the west,and the retreating African slab is pulling the upper plate of western Anatolian apart in *** propose that the Anatolian Plate is moving westwards as

Most viruses and transposons serve as effective carriers for the introduction of foreign DNA up to 11 kb into vertebrate ***,their activity markedly diminishes with payloads exceeding 11 *** the payload capacity of transposons could facilitate more sophisticated cargo designs,improving the regulation of expression and minimizing mutagenic risks associated with molecular therapeutics,metabolic engineering,and transgenic animal *** this study,we improved the Tol2 transposon by increasing protein expression levels using a translational enhancer(QBI SP163,ST)and enhanced the nuclear targeting ability using the nuclear localization protein H2B(SHT).The modified Tol2 and ST transposon efficiently integrated large DNA cargos into human cell cultures(H1299),comparable to the well-established super PiggyBac ***,mRNA from ST and SHT showed a significant increase in transgene delivery efficiency of large DNA payloads(8 kb,14 kb,and 24 kb)into zebrafish(Danio rerio).This study presents a modified Tol2 transposon as an enhanced nonviral vector for the delivery of large DNA payloads in transgenic applications.

安全竞争格局下的基层执法及其生态优化

在中央高度强调统筹发展和安全的政策导向下,地方政府对安全议题的重视程度不断提高。在避责和竞优的双重逻辑支配下,地方政府围绕保障安全这一治理目标展开竞争的趋势日益显著。安全竞争改变了基层执法所处的科层生态,从执法资源、执法组织、执法技术、执法目标四个维度形塑着基层执法。这种形塑效应覆盖了执法活动从决策形成到正式开展的各个阶段。安全竞争格局下,科层执行效能衰减导致的执法不严问题被抑制,但在高强度竞争的阶段或领域,可能出现执法资源分配效率降低、执法组织协作机制失灵、执法技术难以发挥作用、执法目标偏离预设标准等执法偏差问题。安全竞争格局下的执法偏差,根本上是由竞争环境的生态失衡导致的,要避免或矫正执法偏差,需从健全安全风险全过程治理机制、调适惩罚和激励机制的强度、优化执法资源的配置结构等方面入手,对执法生态进行改良和优化。

线粒体自噬是一种清除受损或多余线粒体的过程,在调节细胞内线粒体质量和维持线粒体能量代谢等方面发挥重要作用。TANK结合激酶1(TANK-binding kinase 1,TBK1)是一种多功能的丝氨酸/苏氨酸蛋白激酶,同时参与调控PTEN诱导假定激酶1(PTEN-induced putative kinase 1,PINK1)/Parkin依赖性和非依赖性线粒体自噬过程。近期研究表明,TBK1可磷酸化视神经蛋白(optineurin,OPTN)、p62/sequestosome-1、Ras相关GTP结合蛋白7(Ras-related GTP binding protein 7,Rab7)等自噬相关蛋白,并介导核点蛋白52(nuclear dot protein 52,NDP52)与UNC-51样自噬激活激酶1(UNC-51 like autophagy activating kinase 1,ULK1)复合物相结合,以及TAX1结合蛋白1(TAX1-binding protein 1,TAX1BP1)与微管相关蛋白1轻链3(microtubule-associated protein 1 light chain 3,LC3)相结合,从而增强PINK1/Parkin依赖性线粒体自噬。TBK1亦可作为AMP活化蛋白激酶(AMP-activatedproteinkinase,AMPK)/ULK1自噬通路的作用底物而被激活,再通过磷酸化动力相关蛋白1(dynamin-related protein 1,Drp1)、Rab7促进PINK1/Parkin非依赖性线粒体自噬。本文对TBK1在PINK1/Parkin依赖性和非依赖性线粒体自噬中的作用及机制进行了综述。

机器人操作可形变对象在多类场景中具有重要的应用需求。然而,对象复杂的高维动力学模型导致难以准确、快速地计算其形变。本文建立了一种基于交替方向乘子法的对象形变动力学模型快速、准确、隐式优化求解方法。该方法以通用对象几何模型为输入创建有限元模型,并分别构建材料本构势能函数和操作及碰撞交互单元的位置约束能量函数。随后,采用动力学投影方法构建基于交替方向乘子法的两项优化求解问题,最终快速、准确地计算对象在机器人操作下的形变。数值实验表明,所提出的方法能够在保证相对形变计算误差低于5%的条件下,实现高于24帧/秒的物理形态更新。针对实际应用场景,对所提出的方法开展了从形变仿真预测到在线操作执行的量化评估以及全局约束环境下的离线规划和仿真应用验证。

Recent advances in hydrocarbon exploration have been made in the Member deng-2 marginal microbial mound-bank complex reservoirs of the dengying Formation in the western Sichuan Basin, SW China,where the depositional process is regarded confusing. The microfacies, construction types, and depositional model of the Member deng-2 marginal microbial mound-bank complex have been investigated using unmanned aerial vehicle photography, outcrop section investigation, thin section identification,and seismic reflections in the southwestern Sichuan Basin. The microbialite lithologic textures in this region include thrombolite, dendrolite, stromatolite, fenestral stromatolite, spongiostromata stone,oncolite, aggregated grainstone, and botryoidal grapestone. Based on the comprehensive analysis of“depositional fabrics-lithology-microfacies”, an association between a fore mound, mound framework,and back mound subfacies has been proposed based on water depth, current direction, energy level and lithologic assemblages. The microfacies of the mound base, mound core, mound flank, mound cap, and mound flat could be recognized among the mound framework subfacies. Two construction types of marginal microbial mound-bank complex have been determined based on deposition location, mound scale, migration direction, and sedimentary facies association. Type Jinkouhe microbial mound constructions(TJMMCs) develop along the windward margin owing to their proximity to the seaward subfacies fore mound, with a northeastwardly migrated microbial mound on top of the mud mound,exhibiting the characteristics of large-sized mounds and small-sized banks in the surrounding area. Type E'bian microbial mound constructions(TEMMCs) primarily occur on the leeward margin, resulting from the presence of onshore back mound subfacies, with the smaller southwestward migrated microbial mounds existing on a thicker microbial flat. 日常性违规广泛存在于基层社会,对科层化运作的常规治理机制形成挑战。为应对这一挑战,科层组织衍生出集中执法机制。这一机制是集中力量办大事的治理机制在基层执法领域的延续,在实践中具有突出成效。通过改变常规的运作模式,科层组织集中了分散的执法信息、权限和人员,以统一执法目标、增强执法权威、提高执法密度。在这一过程中,治理资源下沉至基层并得到有效运用,抑制了内部能力损耗。同时,集中执法机制注重因势利导,能够增强基层执法人员的权威,有效提升了基层应对日常性违规的能力。然而,这一机制仍存在继续优化的空间。可通过扩大辅助执法的主体范围,优化执法资源的分配机制以及拓展违规行为的干预路径等方式,进一步提升集中执法机制的实效。
