Serum immune-activation potency and response to anti-TNF-α therapy in Crohn's disease

AIM:To study whether immune-activation stage in serum of adult Crohn's disease (CD) patients correlates with disease activity and with treatment response to antitumor necrosis factor-α (TNF-α) ***:Serum samples were obtained from 15 adult CD patients introduced to anti-TNF-α *** individual stage of immune activation was studied applying our new in vitro assay,in which target cells (donor derived peripheral blood mononuclear cells) were cultured with patient serum and the T-cell activation induced by the patient serum was studied using a panel of markers for effector [interferon γ (IFNγ),interleukin (IL)-5] and regulatory T-cells [forkhead transcription factor 3 (FOXP3) and glucocorticoid-induced tumour necrosis factor receptor (GITR)].The endoscopic disease activity was assessed with the Crohn's disease endoscopic index of severity (CDEIS) before and 3 mo after therapy with an anti-TNF-α ***:Low induction of FOXP3 and GITR in target cells cultured in the presence of patient serum was associated with high disease activity *** assessed before therapy (r=-0.621,P=0.013 and r=-0.625,P=0.013,respectively).FOXP3 expression correlated inversely with pre-treatment erythrocyte sedimentation rate (r=-0.548,P=0.034).Low serum induced FOXP3 (r=-0.600,P=0.018) and GITR (r=-0.589,P=0.021) expression and low IFNγ secretion from target cells (r =-0.538,P=0.039) associated with treatment response detected as a decrease in ***:The immune-activation potency in the patient serum prior to anti-TNF-α therapy reflected intestinal inflammation and the therapeutic response.

Plants synchronize their cellular and physiological functions according to the photoperiod （the length of the light period） in the cycle of 24 h. Photoperiod adjusts several traits in the plant life cycle, including flowering and senes- cence in annuals and seasonal growth cessation in perennials. Photoperiodic development is controlled by the coordinated action of photoreceptors and the circadian clock. During the past 10 years, remarkable progress has been made in under- standing the molecular mechanism of the circadian clock, especially with regard to the transition of Arabidopsis from the vegetative growth to the reproductive phase. Besides flowering photoperiod also modifies plant photosynthetic struc- tures and traits. Light signals controlling biogenesis of chloroplasts and development of leaf photosynthetic structures are perceived both by photoreceptors and in chloroplasts. In this review, we provide evidence suggesting that the photope- riodic development of Arabidopsis leaves mimics the acclimation of plant to various light intensities. Furthermore, the chloroplast-to-nucleus retrograde signals that adjust acclimation to light intensity are proposed to contribute also to the signaling pathways that control photoperiodic acclimation of leaves.

The effect of drawing speed on temperature rise and microstructure distribution in twinning-induced plasticity(TWIP)steel during wire drawing has been investigated to improve drawability for wire rod *** wire drawing process is performed at room temperature,heat is generated due to the plastic deformation and friction at the wire-die *** steel wires subjected to the low drawing speed(LD)of 0.5 m/min and the high drawing speed(HD)of 5.0 m/min were analyzed using the numerical simulation and electron backscatter diffraction ***,the specimens subjected to the HD had a higher drawability by about 18%compared to the LD,which is totally different from the general behavior of plain carbon pearlitic *** LD wire had uniform temperature distribution along the radial direction during wire *** contrast,the HD wire had a temperature gradient along the radial direction due to the higher frictional effect at surface:the minimum temperature of 58℃ at center area and the maximum temperature of 143 C at surface *** higher stacking fault energy of HD wire at the surface area due to the high temperature rise retarded twinning rate,resulting in the prevention of fast exhaustion in ductility in comparison with the LD wires since the earlier depletion of twins at surface area is known as the main reason for the fracture of TWIP steel during wire ***,HD process delayed the fracture strain of wire and increased the uniformity of microstructure and mechanical properties along the radial direction.

The effect of the strain hardening exponent(n)of a material on the changes in shape,strain inhomogeneity,and contact pressure was investigated during wire flat rolling to understand its effect on the deformation behavior of a flat-rolled wire and to determine the optimal working conditions with *** deformation behaviors of wires with various n values were systematically compared using finite element *** shape of the deformed wire was found to depend on the n value of the *** the contact width and lateral spreading of the wire decrease with increasing n,resulting in a large reduction in area with the n *** strain homogeneity of the wire increases with the n value of the *** improvement in the strain homogeneity with the n value is attributable to two factors:a lower strain concentration in the central region and a higher overall elongation as n *** addition,the average effective strain of the wire cross section decreases with the n value of a material during wire flat *** contact pressure distribution on the surface of the wire differs significantly depending on the n *** materials with a low n value,the contact pressure exhibits a higher value at the entry and edge zones of the contact *** contrast,materials with high n values exhibit a higher contact pressure at the exit *** behavior can be explained by the strain hardening behavior of the material during wire flat rolling.

The role of additional ternary alloying elements on the performance of stationary TiFe-based hydrogen storage alloys was investigated based on first-principles density functional theory *** a basic step for examinations,the site preference of each alloying element in the stoichiometric and nonstoichiometric B2TiFe compounds was clarified considering possible antisite *** on the revealed site preference,the effect of various possible ternary elements on the hydrogen storage was examined by focusing on the formation enthalpies of TiFeH and TiFeH_(2) hydrides,which were closely related to the change in the location of plateaus in the pressure-composition-temperature *** physical properties such as the volume expansion due to hydride formation were also examined to provide additional criteria for selecting optimum alloying conditions in future alloying design *** alloying elements that maximize the grain boundary embrittlement due to the solute segregation were proposed for the enhanced initial activation of TiFe-based hydrogen storage alloys.

AIM: To investigate local corticosterone production and angiotensin-Ⅰ converting enzyme(ACE) protein expression and their interaction in healthy and inflamed ***: Acute intestinal inflammation was induced to six weeks old male Balb/c mice by administration of either 3% or 5% dextran sodium sulfate(DSS) in drinking water for 7 d(n = 12 in each group). Healthy controls(n = 12) were given tap water. Corticosterone production and ACE protein shedding were measured from ex vivo incubates of the small and large intestine using EIA and ELISA, respectively. Morphological changes of the intestinal wall were assessed in hematoxylin-eosin stained tissue preparations of jejunum and distal colon. Effects of angiotensin Ⅱ, captopril and metyrapone on corticosterone production was assessed by incubating pieces of small intestine of healthy mice in the presence of 0.1, 1 or 10 μmol/L angiotensin Ⅱ, 1, 10 or 100 μmol/L captopril or 1, 10 or 100 μmol/L metyrapone solutions and measuring corticosterone released to the incubation buffer after 90 min(n = 5 in each group).RESULTS: Both concentrations of DSS induced inflammation and morphological changes in large intestines but not in small intestines. Changes were observed as distortions of the crypt structure, mucosal erosion, immune cell infiltration to the mucosa and submucosal edema. Ex vivo corticosterone production(2.9 ± 1.0 ng/mL vs 2.0 ± 0.8 ng/mL, P = 0.034) and ACE shedding(269.2 ± 97.1 ng/m L vs 175.7 ± 52.2 ng/mL, P = 0.016) were increased in small intestines in 3% DSS group compared to the controls. In large intestine, corticosterone production was increased compared to the controls in both 3% DSS(229 ± 81 pg/mL vs 158 ± 30 pg/mL, P = 0.017) and 5% DSS groups(366 ± 163 pg/mL vs 158 ± 30 pg/mL, P = 0.002). Large intestine ACE shedding was increased in 5% DSS group(41.5 ± 9.0 ng/mL vs 20.9 ± 5.2 ng/mL, P = 0.034). A new synchronization method, called the input/output-to-state stable synchronization (IOSSS) method, is proposed for a general class of chaotic systems with external disturbance. By introducing Lyapunov stability theory and linear matrix inequality (LMI) for the first time, the IOSSS controller is shown to not only guarantee the synchronization of the chaotic systems, but also reduce the effect of external disturbance. The proposed IOSSS controller can be obtained by solving the LMI, which can easily be done using standard numerical packages. A numerical example is given to demonstrate the availability of the proposed method.

In?uence of Carbon Equivalent Value on the Weld Bead Bending Properties of High-Strength Low-Alloy Steel Plates

The contribution of chemical compositions in terms of carbon equivalent value（CEV） on impending crack propagation during weld bead bend tests（WBBTs） was studied with five different high-strength lowalloy steel plates that underwent different rolling processes. The test showed that cracks developed at the weld joint, easily passed through the coarse-grain heat-affected zone（CGHAZ）, and finally were arrested within the heat-affected zone（HAZ）. An apparent decrease in the average crack length, which consequently indicated improvement in crack arrestability, was found with decreasing CEV. In addition,the relatively fine microstructure in the HAZ of low-CEV steel plates helped in preventing the crack from further propagation. Special emphasis was placed on the empirical expectation of critical CEV above which WBBT might fail.
