Proinflammatory cytokine TNFα promotes HPV-associated oral carcinogenesis by increasing cancer stemness

High-risk human papillomaviruses(HPVs) are involved in the development of several human cancers, including oropharyngeal squamous cell carcinomas. However, many studies have demonstrated that HPV alone is not sufficient for the oncogenic transformation of normal human epithelial cells, indicating that additional cofactors are required for the oncogenic conversion of HPV-infected cells. Inasmuch as chronic inflammation is also closely associated with carcinogenesis, we investigated the effect of chronic exposure to tumor necrosis factor α(TNFα), the major proinflammatory cytokine, on oncogenesis in two immortalized oral keratinocyte cell lines, namely, HPV16-immortalized and human telomerase reverse transcriptase(h TERT)-immortalized cells. TNFαtreatment led to the acquisition of malignant growth properties in HPV16-immortalized cells, such as(1) calcium resistance,(2)anchorage independence, and(3) increased cell proliferation in vivo. Moreover, TNFα increased the cancer stem cell-like population and stemness phenotype in HPV16-immortalized cells. However, such transforming effects were not observed in h TERTimmortalized cells, suggesting an HPV-specific role in TNFα-promoted oncogenesis. We also generated h TERT-immortalized cells that express HPV16 E6 and E7. chronic TNFα exposure successfully induced the malignant growth and stemness phenotype in the E6-expressing cells but not in the control and E7-expressing cells. We further demonstrated that HPV16 E6 played a key role in TNFα-induced cancer stemness via suppression of the stemness-inhibiting micro RNAs mi R-203 and mi R-200 c. Overexpression of mi R-203 and mi R-200 c suppressed cancer stemness in TNFα-treated HPV16-immortalized cells. Overall, our study suggests that chronic inflammation promotes cancer stemness in HPV-infected cells, thereby promoting HPV-associated oral carcinogenesis.

Danish cohort of monozygotic inflammatory bowel disease twins: clinical characteristics and inflammatory activity

AIM: To describe the establishment of a Danish inflammatory bowel diseases (IBD) twin cohort with focus on concordance of treatment and inflammatory ***: We identified MZ twins, likely to be discordant or concordant for IBD, by merging information from the Danish Twin Register and the National Patient Register. The twins were asked to provide biological samples, questionnaires, and data access to patient files and public registries. Biological samples were collected via a mobile laboratory, which allowed for immediate centrifugation, fractionation, and storage of samples. The mean time from collection of samples to storage in the -80 °c mobile freezer was less than one hour. The diagnoses where validated using the copenhagen diagnostic ***: We identified 159 MZ IBD twin pairs, in a total of 62 (39%) pairs both twins agreed to participate. Of the supposed 62 IBD pairs, the IBD diagnosis could be confirmed in 54 pairs. The cohort included 10 concordant pairs, whereof some were discordant for either treatment or surgery. The 10 concordant pairs, where both pairs suffered from IBD, included eight cD/cD pairs, one Uc/Uc pair and one Uc/IBDU pair. The discordant pairs comprised 31 Uc, 5 IBDU (IBD unclassified), and 8 cD discordant pairs. In the co-twins not affected by IBD, calprotectin was above 100 μg/g in 2 participants, and above 50 μg/g in a further 5 ***: The presented IBD twin cohorts are an excellent resource for bioinformatics studies with proper adjustment for disease-associated exposures including medication and inflammatory activity in the co-twins.

Effects of 7 years of field weathering on biochar recalcitrance and solubility

How weathering affects the physiochemical properties of biochar and its long-term carbon(c)sequestration potential remains *** this study,we measured changes in biochar recalcitrance and solubility after 7 years of weathering in a cultivated *** recalcitrance and biodegradability of weathered and unweathered hardwood biochar mixtures were determined by thermal analysis(differential scanning calorimetry)and evolved gas *** in biochar solubility and the chemical composition of biochar-derived dissolved organic carbon(DOc)were determined by repeated laboratory leaching and UV-Vis *** surface carbon-oxidation state(cox)of biochar increased by 117.6-158.2%with weather-ing in the field,and there was an average of 0.9-1.2%decrease in biochar c contents per ***,thermal indices of biochar recalcitrance and biodegradability,which suggested intermediate c sequestration potential,were not significantly different between weathered and unweathered *** O:c ratio increased with weathering suggesting an increase in biodegradability,however,both weathered and unweathered biochars were still estimated to have half-lives of over 1000 ***-soluble organic carbon(WSOc)concentrations from the unweathered biochar rapidly decreased during laboratory leaching to levels similar to the field-weathered biochars,and aromaticity(SUVA_(254))of WSOc increased from 5.9%in the unweathered biochar to 42%aromaticity in the biochars weathered for 7 *** conclude that during short-term(years)weathering under field conditions,there is continued solubilization of increasingly aromatic biochar-c compounds,however,this accounts for a relatively small proportion of biochar c such that there is little-to-no change in biochar stability or c sequestration potential after field application.
