Differential regulation of protein synthesis in skeletal muscle and liver of neonatal pigs by leucine through an mTORC1-dependent pathway

Neonatal growth is characterized by a high protein synthesis rate that is largely due to an enhanced sensitivity to the postprandial rise in insulin and amino acids, especially leucine. The mechanism of leucine＇s action in vivo is not well understood. In this study, we investigated the effect of leucine infusion on protein synthesis in skeletal muscle and liver of neonatal pigs. To evaluate the mode of action of leucine, we used rapamycin, an inhibitor of mammalian target of rapamycin （mTOR） complex-1 （mTORC1）. Overnight-fasted 7-day-old piglets were treated with rapamycin for 1 hour and then infused with leucine （400 μmol·kg^-1·h^-1） for 1 hour. Leucine infusion increased the rate of protein synthesis, and ribosomal protein S6 kinase 1 （S6K1） and eukaryotic initiation factor （elF） 4E-binding protein-1 （4E-BP1） phosphorylation in gastrocnemius and masseter muscles （P 〈 0.05）, but not in the liver. The leucine-induced stimulation of protein synthesis and S6K1 and 4E-BP1 phosphorylation were completely blocked by rapamycin, suggesting that leucine action is by an mTORC1-dependent mechanism. Neither leucine nor rapamycin had any effect on the activation of the upstream mTQRC1 regulators, AMP-activated protein kinase and protein kinase B, in skeletal muscle or liver. The activation of elF2α and elongation factor 2 was not affected by leucine or rapamycin, indicating that these two pathways are not limiting steps of leucine-induced protein synthesis. These results suggest that leucine stimulates muscle protein synthesis in neonatal pigs by inducing the activation of mTORC1 and its downstream pathway leading to mRNA translation.

The A/ENTH Domain-Containing Protein AtECA4 Is an Adaptor Protein Involved in Cargo Recycling from the trans-Golgi Network/Early Endosome to the Plasma Membrane

Endocytosis and subsequent trafficking pathways are crucial for regulating the activity of plasma membrane-localized proteins. Depending on cellular and physiological conditions, the internalized cargoes are sorted at （and transported from） the trans-Golgi network/early endosome （TGN/EE） to the vacuole for degradation or recycled back to the plasma membrane. How this occurs at the molecular level remains largely elusive. Here, we provide evidence that the ENTH domain-containing protein AtECA4 plays a crucial role in recycling cargoes from the TGN/EE to the plasma membrane in Arabidopsis thaliana. AtECA4：sGFP primarily localized to the TGN/EE and plasma membrane （at low levels）. Upon NaCI or mannitol treatment, AtECA4：sGFP accumulated at the TGN/EE at an early time point but was released from the TGN/EE to the cytosol at later time points. The ateca4mutant showed higher resistance to osmotic stress and more sensitive to exogenous abscisic acid （ABA） than the wild type, as well as increased expres- sion of ABA-inducible genes RD29A and RD29B. Consistently, ABCG25, a plasma membrane-localized ABA exporter, accumulated at the prevacuolar compartment in ateca4, indicating a defect in recycling to the plasma membrane. However, the role of AtECA4 in cargo recycling is not specific to ABCG25, as it also functions in the recycling of BRII. These results suggest that AtECA4 plays a crucial role in the recycling of endocytosed cargoes from the TGN/EE to the plasma membrane.

Water level predictions in the river,lake and delta play an important role in flood *** year Mekong River delta of Vietnam is experiencing flood due to heavy monsoon rains and high *** subsidence may also aggravate flooding problems in this ***,accurate predictions of water levels in this region are very important to forewarn the people and authorities for taking timely adequate remedial measures to prevent losses of life and *** are so many methods available to predict the water levels based on historical data but nowadays Machine Learning(ML)methods are considered the best tool for accurate *** this study,we have used surface water level data of 18 water level measurement stations of the Mekong River delta from 2000 to 2018 to build novel time-series Bagging based hybrid ML models namely:Bagging(RF),Bagging(SOM)and Bagging(M5P)to predict historical water levels in the study *** of the Bagging-based hybrid models were compared with Reduced Error Pruning Trees(REPT),which is a benchmark ML *** data of 19 years period was divided into 70:30 ratio for the *** data of the period 1/2000 to 5/2013(which is about 70%of total data)was used for the training and for the period 5/2013 to 12/2018(which is about 30%of total data)was used for testing(validating)the *** of the models was evaluated using standard statistical measures:Coefficient of Determination(R2),Root Mean Square Error(RMSE)and Mean Absolute Error(MAE).Results show that the performance of all the developed models is good(R2>0.9)for the prediction of water levels in the study ***,the Bagging-based hybrid models are slightly better than another model such as ***,these Bagging-based hybrid time series models can be used for predicting water levels at Mekong data.

Embryonic, genetic and clinical outcomes of fresh versus vitrified oocyte: A retrospective cohort study

Objective:To compare embryonic development,ploidy status and clinical outcomes between fresh and frozen-thawed ***:This retrospective cohort study evaluated 83 fertilization cycles including both fresh and frozen oocytes from 79 patients at the HP Fertility Center of Hai Phong International Hospital of Obstetrics and Pediatrics in *** patient underwent several ovarian stimulation cycles to accumulate a certain number of oocytes that would be *** the last oocyte retrieval,all patient’s oocytes including both frozen and fresh would be *** outcomes included the rates of oocyte survival,cleavage embryo,blastocyst,ploidy status,pregnancy,biochemical pregnancy and clinical ***:The oocyte survival rate after thawing was 96.5%.No statistically significant difference was found when comparing fresh and frozen oocytes regarding fertilization rate(78.1%vs.75.5%,P=0.461),usable cleavage embryo rate(86.9%vs.87.2%,P=0.916)but usable blastocyst rate was found higher statistically in the frozen oocyte group(44.4%vs.54.0%,P=0.049).The percentages of euploid,aneuploid and mosaic embryos between the fresh group and the vitrified group had no significant differences(33.8%vs.31.6%,P=0.682;51.0%vs.54.2%,P=0.569;15.2%vs.12.4%,P=0.787;respectively).The rates of pregnancy,biochemical pregnancy and clinical pregnancy had no statistical difference(68.8%vs.64.8%,P=0.764;12.5%vs.3.6%,P=0.258;37.5%vs.46.4%,P=0.565).17 Mature oocytes are the minimum to have at least one euploid ***:Oocyte vitrification does not affect embryonic,genetic and clinical *** number of mature oocytes should be considered for fertilization in some cases.

Our study aimed to compare the essential oil(EO)concentration and composition of several Apiaceae species growing in the Northern region of *** yields of EOs from materials ranged from 0.03%(root EO of Angelica acutiloba and aerial parts EO of Heracleum bivittatum)-0.27%(leaf EO of Xyloselinum vietnamense).Gas chromatography-mass spectrometry(GC-MS)allowed the identiﬁcation of 74 components in the EOs of six Apiaceae species,making up 94.4%–100.0%of the *** EO from Angelica acutiloba,(Z)-ligustilide accounted for an extremely large proportion(94.9%).EO of Angelica pubescens was dominated by six character-istic components includingα-pinene(21.5%),β-phellandrene(18.1%),p-cymene(12.2%),3-methylnonane(8.7%),o-cymene(8.1%),and D-sylvestrene(6.2%).The EO from Cryptotaenia japonica was characterized by high amounts ofα-selinene(48.7%),β-selinene(23.7%),and trans-β-farnesene(5.4%).The EOs from leaves and stems of Xyloselinum vietnamense were characterized by high concentrations of sabinene(69.8%and 33.8%),4-terpineol(8.7%and 7.4%)andβ-pinene(4.0%and 6.5%)while EOs from aerial parts and root of Xylo-selinum leonidii comprise four characteristic monoterpenes includingα-pinene(28.2%and 52.8%),β-pinene(7.9%and 10.3%),β-phellandrene(7.6%and 15.3%),and sabinene(3.0%and 4.1%).Additionally,cryptone is also one of the major components in the EO of Xyloselinum leonidii(13.2%in the aerial parts oil and 2.8%in the root oil).In the EOs isolated from the aerial parts and root of Heracleum bivittatum,α-pinene(22.5%and 70.2%)andβ-pinene(43.2%and 20.0%)were the predominant *** appeared in the EO from aerial parts of Heracleum bivittatum with a relatively high concentration(13.5%)while bornyl acetate(5.1%)was also one of the main components in the EO from its aerial parts but was not detected in other Apiaceae species in the present *** databases help identify and control the quality of plant material studied from the family Apiaceae growing in Vietnam.

以探讨中国人工作场所越轨行为的内容与结构为主旨。首先,采取主位与客位研究相结合的范式,用开放式问卷对39名人力资源管理专业研究生课程进修班学员进行工作场所越轨行为项目征集,经语义分析后共获得45条工作场所越轨行为。接着,采取无固定锚点的分类方式让42名工商管理硕士班学员自由地将上述行为分成2～12类。然后,再采用多维量表分析剖析分类特征。研究结果发现:工作场所越轨行为分成4类,能较好地反映被试对该行为的认识。最后,以多维量表分析法获得的各行为四维度坐标为基准,采用层次聚类分析探寻各工作场所越轨行为的相互关系。研究结果表明:中国人将工作场所越轨行为分成生产型过失、违法行为、损公肥私与合作破坏行为4类。研究得出的结论有助于建构和发展中国特色的工作场所越轨行为理论体系。

Root system architecture(RSA)plays a pivotal role in efficient uptake of essential nutrients,such as phosphorous(P),nitrogen(N),and water In soils with heterogeneous nutrient distribution,root plasticity can optimize acquisition and plant ***,we present evidence that a constitutive RSA can confer benefits for sorghum grown under both sufficient and limiting growth *** studies,using P efficient SC103 and inefficient BTx635 sorghum cultivars,identified significant differences in root traits,with SC103 developing a larger root system with more and longer lateral roots,and enhanced shoot biomass,under both nutrient sufficient and deficient *** addition to this constitutive attribute,under P deficiency,both cultivars exhibited an initial increase in lateral root development;however,SC103 still maintained the larger root *** N deficiency and drought stress inhibited both root and shoot growth,for both sorghum cultivars,SC103 again maintained the better *** findings reveal that SC103,a P efficient sorghum cultivar,also exhibited enhanced growth performance under N deficiency and *** results provide evidence that this constitutive nature of RSA can provide an avenue for breeding nutrient-and drought-resilient crops.

Clinical validation of the early embryo viability assessment system: Analysis for the blastocyst morphology and pregnancy outcomes

Objective:To determine the relationship between the early embryo viability assessment(EEVA)and blastocyst morphological parameters and pregnancy ***:This retrospective cohort study was conducted on 291 intracytoplasmic sperm injection cycles including 2522 embryos with indications of prolonging embryo culture to the blastocyst stage in the Genea embryo review incubator,and 511 single vitrified-warmed blastocyst transfer cycles from January 2020 to June *** EEVA system produced an EEVA score from E1(best)to E5(worse)for the potential of blastocyst *** morphology was *** association between the EEVA score and each type of blastocyst morphology,implantation rate,clinical pregnancy,and ongoing pregnancy were assessed using generalized estimating ***:The inner cell mass A(ICM A),trophectoderm A(TE A),blastocoele expansion degree of 3,4,5,6,7 rates were higher with lower the EEVA *** adjusted odd ratio(aOR)(E5 vs E1)was 0.3 for ICM A,0.174 for TE A and 0.210 for BL3,4,5,6,7(all P<0.001),suggesting a significant association between lower EEVA scores and improved embryo *** implantation,clinical pregnancy,and ongoing pregnancy rate were also higher with lower the EEVA *** aOR of E5 vs E1 was 0.245 for implantation,0.185 for clinical pregnancy and 0.200 for ongoing pregnancy rate(P<0.001).Conclusions:There were associations between blastocyst morphology,pregnancy outcome and EEVA *** good blastocyst morphology and pregnancy outcomes are higher with lower the EEVA score.

Folate-targeted nanostructured chitosan/chondroitin sulfate complex carriers for enhanced delivery of bortezomib to colorectal cancer cells

Folate-targeting self-assembled nanoparticles(NPs) using biocompatible and biodegradable natural polymers chitosan(Cs) and chondroitin sulfate(Chs) were developed to address the major challenge in cancer treatment, the selective delivery of nanoparticles to the target site. In this study, we successfully incorporated a hydrophobic drug, bortezomib(Bor), into folic acid(FA)-conjugated Cs/Chs self-assembled NPs(Bor/Cs/Chs-FA) for colorectal cancer therapy. The particle size and polydispersity index of Bor/Cs/Chs-FA were196.5 ± 1.2 nm and0.21 ± 0.5, respectively. A pH-dependent release profile was observed, facilitating cancer cell-targeted drug release under an acidic tumor microenvironment. Moreover, in vitro data revealed enhanced cellular uptake and apoptosis in folate receptor-expressing colorectal cancer cells(HCT-116 and HT-29) as compared to that in lung cancer cells(A549), which do not express folate receptors. Furthermore, intravenous administration of Bor/Cs/Chs-FA in a HCT-116 bearing xenograft mouse model showed that the NPs were a safe and effective drug delivery system. The results suggest that folate-targeted nanoparticle can be effectively applied for efficient chemotherapy of colorectal cancer.

Hematopoietic Stem Cell Transplantation in Vietnam： Current Status and Future Directions

In recent years, there have been an increasing number of hematologic patients in Vietnam. Hematopoietic stem cell transplantation （HSCT） is fast becoming an effective method to cure many benign and malignant blood disorders. Nine transplant centers are operating throughout Vietnam. HCMC Blood Transfusion Hematology Hospital （BTH） is one of these centers where bone marrow, peripheral blood and cord blood stem cell transplantation were first conducted in our country. At BTH, half of the patients received allogeneic HSCT, mainly from matched sibling donors due to lack of a national bone marrow donor registry. While acute myeloid leukemia was a common indication of allogeneic HSCT, most of the patients undergoing autologous HSCT had multiple myeloma. One major issue we encountered concerned bacterial and fungal infections during neutropenia period. Because of an endemic area, CMV reactivation frequently occurred. Five-year overall survival （OS） and event-free survival （EFS） were 47% and 46%, respectively. To create more opportunities for patients, our future directions will focus on some new transplant procedures, such as haploidentical, unrelated and cord blood transplantation.
