Effects of groundwater level on vegetation in the arid area of western China

Formaldehyde up-regulates TRPV1 through MAPK and PI3K signaling pathways in a rat model of bone cancer pain

Objective Our previous study showed that tumor tissue-derived formaldehyde at low concentrations plays an important role in bone cancer pain through activating transient receptor potential vanilloid subfamily member 1 （TRPV 1）. The present study further explored whether this tumor tissue-derived endogenous formaldehyde regulates TRPV1 expres- sion in a rat model of bone cancer pain, and if so, what the possible signal pathways are during the development of this type of pain. Methods A rat model of bone cancer pain was established by injecting living MRMT-1 tumor cells into the tibia. The formaldehyde levels were determined by high performance liquid chromatography, and the expression of TRPV1 was examined with Western blot and RT-PCR. In primary cultured dorsal root ganglion （DRG） neurons, the ex- pression of TRPV1 was assessed after treatment with 100 ~tmol/L formaldehyde with or without pre-addition of PD98059 [an inhibitor for extracellular signal-regulated kinase], SB203580 （a p38 inhibitor）, SP600125 [an inhibitor for c-Jun N- terminal kinase], BIM [a protein kinase C （PKC） inhibitor] or LY294002 [a phosphatidylinositol 3-kinase （PI3K） inhibi- tor]. Results In the rat model of bone cancer pain, formaldehyde concentration increased in blood plasma, bone marrow and the spinal cord. TRPV1 protein expression was also increased in the DRG. In primary cultured DRG neurons, 100 p^mol/L formaldehyde significantly increased the TRPV1 expression level. Pre-incubation with PD98059, SB203580, SP600125 or LY294002, but not BIM, inhibited the formaldehyde-induced increase of TRPV1 expression. Conclusion Formaldehyde at a very low concentration up-regulates TRPV1 expression through mitogen-activated protein kinase and PI3K, but not PKC, signaling pathways. These results further support our previous finding that TRPV1 in peripheral after- ents plays a role in bone cancer pain.

Effects of ethanol extract of bombax ceiba leaves and its main constituent mangiferin on diabetic nephropathy in mice

The present study was designed to explore the mechanism by which ethanol extract of bombax ceiba leaves(BCE) and its main constituent mangiferin(MGF) affect diabetic nephropathy by combating oxidative stress. Oral administration of BCE and MGF to normal and streptozotocin(STZ)-induced diabetic mice were carried out. Fasting blood glucose, 24-h urinary albumin, serum creatinine, and blood urea nitrogen were tested, histopathology, and immunohistochemical analysis of kidney tissues were performed. Moreover, mesangial cells were treated with BCE and MGF for 48 h with or without 25 mmol·L^(-1) of glucose. Immunofluorescence, Western blot and apoptosis analyses were used to investigate their regulation of oxidative stress and mitochondrial function. BCE and MGF ameliorated biochemical parameters and restored STZ-induced renal injury in the model mice. In vitro study showed that high glucose stimulation increased oxidative stress and cell apoptosis in mesangial cells. BCE and MGF limited mitochondrial membrane potential(Δψm) collapse by inhibiting Nox4, mitochondrially bound hexokinase II dissociation, and subsequent ROS production, which effectively reduced oxidative stress, cleaved caspase-3 expression and cell apoptosis. Our work indicated that BCE and MGF had protective effects on diabetic caused kidney injury and prevented oxidative stress in mesangial cells by regulation of hexokinase II binding and Nox4 oxidase signaling.

Effect of replacing antibiotics using multi-enzyme preparations on production performance and antioxidant activity in piglets

The study was conducted to investigate the effects of replacing antibiotics using multi-enzyme preparations on growth performance,coefficient of total tract apparent digestibility,digestive enzyme activity,and antioxidant property in piglets.A total of 160 piglets（（21.35±0.22）kg）were randomly assigned to five dietary treatments：1）basal diet supplemented with antibiotics（AC）,2）antibiotic diet supplemented with 0.5 g kg^-1 multi-enzyme preparations（AC＋0.5EP）,3）antibiotic diet supplemented with 1.5 g kg^-1 multi-enzyme preparations（AC＋1.5EP）,4）basal diet supplemented with a half dosage of antibiotics and 1.5 g kg^-1 multi-enzyme preparations（AH＋1.5EP）,and 5）basal diet supplemented with 1.5 g kg^-1 multienzyme preparations（BC＋1.5EP）.The results showed that AC＋1.5EP significantly improved the feed efficiency,apparent digestibility of ether extract（EE）and crude ash（CA）,lipase activity in pancreas and duodenum content,maltase and lactase activity in jejunum and ileum mucosa,glutathione peroxidase（GSH-Px）concentration in serum and liver,and decreased malondialdehyde（MDA）concentration in serum and liver compared with piglets receiving AC（P〈0.05）.Piglets receiving BC＋1.5EP showed no significant difference in growth performance（P〉0.05）but had lower MDA concentration than piglets receiving AC（P〈0.05）.The apparent digestibility of EE and crude fiber（CF）,duodenal lipase activity,jejunum mucosa maltase,and ileum mucosa lactase activity of piglets receiving AH＋1.5EP or BC＋1.5EP were significantly improved compared with piglets receiving AC（P〈0.05）.These results indicated an additive growth promotion effect between antibiotics and multi-enzyme preparations on piglets,and the multi-enzyme preparations may be used as substitutes for antibiotics for improving piglet production performance and health status.

Background Tumstatin is a recently developed endogenous vascular endothelial growth inhibitor that can be applied as an anti-angiogenesis and antineoplastic agent. The study aimed to design and synthesize the small molecular angiogenesis inhibition-related peptide （peptide 21）, to replicate the structural and functional features of the active zone of angiogenesis inhibition using tumstatin and to prove that synthesized peptide 21 has a similar activity： specifically inhibiting tumor angiogenesis like tumstatin. Methods Peptide 21 was designed and synthesized using biological engineering technology. To determine its biological action, the human umbilical vein endothelial cell line ECV304, the human ovarian cancer cell line SKOV-3 and the mouse embryo-derived NIH3T3 fibroblasts were used in in vitro experiments to determine the effect of peptide 21 on proliferation of the three cell lines using the MTT test and growth curves. Transmission electron microscopy （TEM） and flow cytometry （FCM） were applied to analyze the peptide 21-induced apoptosis of the three cell lines qualitatively and quantitatively. In animal experiments, tumor models in nude mice subcutaneously grafted with SKOV-3 were used to observe the effects of peptide 21 on tumor weight, size and microvessel density （MVD）. To initially investigate the role of peptide 21, the effect of peptide 21 on the expression of vascular endothelial growth factors （VEGFs） by tumor tissue was semi-quantitatively analyzed. Results The in vitro Ml-r test and growth curves all indicated that cloned peptide 21 could specifically inhibit ECV304 proliferation in a dose-dependent manner （P 〈0.01）; TEM and FCM showed that peptide 21 could specifically induce ECV304 apoptosis （P 〈0.01）. Results of in vivo experiments showed that tumors in the peptide 21 group grew more slowly. The weight and size of the tumors after 21 days of treatment were smaller than those in the control group （P 〈0.05）, with a mean tumor inhibition rate of 67.

山西金晖瑞隆煤业有限公司主采的8+10号煤层具有自然发火危险性,为准确确定8115工作面自燃带位置,以便为采取防灭火措施提供靶位,以氧气体积分数作为划分指标,采用束管抽气法进行采空区自燃“三带”现场测定。构建采空区数值模型,采用FLUENT软件进行解算,得出采空区氧气体积分数分布规律。基于此,对自燃“三带”进行划分,确定自燃带宽度,并提出防灭火对策。结果表明:8115工作面采空区散热带为0~51.2 m;自燃带为51.2~86.2 m;窒息带为86.2 m以里。建议采用埋管注氮或架间插管汽雾阻化防灭火措施,对采空区自燃带进行全方位防灭火控制。

为弥补现有研究在企业知识基础、合作能力与技术并购创新绩效之间的作用机理的不足,基于医药行业技术壁垒高、技术更新速度快的特点,以2007—2018年我国沪深A股生物医药行业发生的109起技术并购事件为对象,构建含有中介变量的回归模型,探究医药企业知识基础、合作能力对技术并购创新绩效的作用机制以及吸收能力的中介效应。结果显示:企业知识基础和合作能力对创新绩效有显著正向影响,吸收能力在知识基础、合作能力与创新绩效关系中均起到部分中介作用;并购方首席执行官接受技术类教育水平越高,企业技术特征的敏感度更高;互补型技术并购加强了主体知识基础对创新绩效的正向影响,相似性技术并购则加强了合作能力对创新绩效的正向影响。最后,从重视前期技术能力积累和技术并购后研发投入,强化人才战略以及适当选择并购目标方面提出相关建议。

Using observed wind and water vapor data from June 2006,water vapor exchange between the Rongbuk Valley and its above atmosphere is estimated for the first *** water vapor level shows a high value from 23-29 June and a low from 12-21 June,which co-incide with the South Asian summer monsoon (SASM) active and break stages,*** water vapor can be strongly injected into the closed region of the Rongbuk Valley from the outside atmosphere,with an average strength of 0.4 g s-1 m-2 in June 2006,given that no evaporation *** air moisture exchange proc-esses can be greatly affected by the SASM evolution through changes in local radiation forcing.

The renewability and quality of shallow groundwater in Sanjiang and Songnen Plain, Northeast China

Groundwater is a key component for water resources in Sanjiang and Songnen Plain, an important agriculture basement in China. The quality and the renewability of irrigation groundwater are essential for the stock raising and agricultural production. Shallow groundwater was sampled and analyzed for various variables. The salinity sodium concentration and bicarbonate hazard, were examined with regard to the United States Department of Agriculture （USDA） irrigation water standards. The concentration of chlorofluorocarbons （CFCs） was determined to analyze the age of groundwater. Most groundwater samples labeled as excellent to good for irrigation with low salinity hazard or medium salinity hazard. Four groundwater samples were good and suspected for irrigation with high salinity hazard. Generally groundwater in Sanjiang Plain was younger than the groundwater in Songnen Plain. Meanwhile, groundwater nearby river is younger than the groundwater further away inside the watershed. The mean age of groundwater in Sanjiang Plain is in average of 44.1,47.9 and 32.8 years by CFC-11 （CCI3F）, CFC-12 （CCI2F2） and CFC-113 （C2CI3F3）, respectively. The mean ages of groundwater in Songnen Plain is in average of 46.1, 53.4, and 40.7 years by CFC-11, CFC-12 and CFC-113, respectively. Thus, groundwater nearby rivers could be directly exploited as irrigation water. Partial groundwater has to be processed to lower the salt concentration rather than directly utilized as irrigation water in Songnen Plain. both water quality and renewability should be put in mind for sustainable agricultural development and water resources management.
