Soybean is a broadly popular and ext.nsively cult.vat.d crop, however, many high-yield and high-qualit. variet.es require specific growt. condit.ons, rest.ict.ng t.eir widespread adopt.on. t.e appropriat. light.condit.ons and phot.period must.be at.ained for t.ese variet.es t. t.rive in new environment.. In t.is st.dy, we employed CRISPR/Cas9 t. design t.o sgRNAs aimed at.knocking out.t.e mat.rit.-relat.d gene E4 in a major American soybean variet. called ''Jack'', which belongs t. mat.rit. group MGII. E4 gene is primarily involved in t.e phot.periodic flowering and mat.rit. in soybean, making it.an ideal candidat. for genet.c manipulat.on. We successfully obt.ined 1 homozygous E4-SG1 mut.nt.t.pe wit. 1-bp insert.on, and 4 homozygous E4-SG2 mut.nt. t.pe wit. 2-bp delet.on, 7-bp delet.on, 61-bp delet.on, and 1-bp insert.on, respect.vely. t.e homozygous e4 mut.nt.plant. cont.ined early t.rminat.on codons devoid of t.ansgenic element.. Addit.onally, no pot.nt.al off-t.rget.sit.s of t.e E4 gene were det.ct.d. A comparat.ve analysis revealed t.at. unlike t.e wild-t.pe, t.e mat.rit. t.me of homozygous e4 mut.nt. was early under bot. short.day and long-day condit.ons. t.ese mut.nt. offer novel germplasm resources t.at.may be used t. modify t.e phot.period sensit.vit. and mat.rit. of soybean, enhancing it. adapt.bilit. t. high-lat.t.de regions. © 2024 t.e Oil Crop Research Inst.t.t., Chinese Academy of Agricult.re Sciences
