Dear Editor,Infections by the influenza virus are a significant and widespread global health threat,as these infections have an annual death toll ranging from 290,000 to 650,000.1 A significant proportion of these fatalities are attributed to secondary bacterial pneumonia,a severe complication commonly caused by ubiquitous respiratory pathogens such as Staphylococcus aureus(***).2 Of particular concern is the increased morbidity and mortality rates in individuals infected simultaneously with influenza virus and methicillin-resistant ***(MRSA).3 The concurrent presence of bacteria and influenza virus usually causes acute respiratory distress syndrome(ARDS),which is associated with acute lung injury(ALI),severe lung tissue edema,and widespread ***,determining the complex mechanisms underlying the synergistic interplay will require further investigation with a suitable coinfection mouse *** our previous study,we utilized different sequential coinfections at various time points to model influenza A virus and MRSA coinfection.

Ta2NiSe5is a promising candidate for hosting an excitonic insulator （EI） phase,a novel electronic state driven by electron-hole Coulomb ***,the role of electron-lattice coupling in the formation of the EI phase remains ***,we use angle-resolved photoemission spectroscopy （ARPES） to study the band structure evolution of Ta2Ni(Se1-xSx)5with sulfur substitution and potassium deposition,which modulate the band gap and the carrier concentration,*** find that the Ta 5d states originating from the bottom of the conduction band persist at the top of the valence band in the low-temperature monoclinic phase,indicating the importance of exciton condensation in opening the gap in the semi-metallic band *** also observe that the characteristic overlap between the conduction and valence bands can be restored in the monoclinic lattice by mild carrier injection,suggesting that the lattice distortion in the monoclinic phase is not the main factor for producing the insulating gap,but rather the exciton condensation in the electronic system is the dominant driving *** results shed light on the electron-lattice decoupling and the origin of the EI phase in Ta2Ni(Se1-xSx)5.

Studying the cytokine profiles in animal models or patients with sepsis provides an experimental basis for the identification of diagnostic biomarkers and therapeutic targets. In this study, we used a liquid protein chip （LiquiChip）, also known as flexible multi-analyte profiling technology, to perform quantitative analyses of several cytokines and chemokines （e.g., IL-II3, IL-2, IL-4, IL-5, IL-6, IL-10, IL-12, TNF-ct, IFN-7, granulocyte-macrophage colony-stimulating factor, keratinocyte chemoattractant, monocyte chemoattractant protein, monokine induced by gamma interferon, IFN-7-inducible protein 10, and macrophage inflammatory protein 1 alpha）. The levels of these cytokines and chemokines were determined both in the blood and in tissues, including the lung, liver, heart, kidney, spleen, brain, stomach, intestine and muscle, of mice challenged with LPS, Our data showed variable production levels of LPS-induced cytokines in different mouse organs, and the cytokine in the blood did not correlate with those in the organs. We also showed that the levels of most cytokines peaked within 1 to 6 h and decreased rapidly afterward. A variety of inflammatory cytokines might be related to the damage in different organs during septic shock. Our data also suggest that the spleen might be an important target organ in the development of systemic inflammatory response syndrome and sepsis.

Immunotherapy has limited efficacy against locally advanced pancreatic cancer(LAPC)due to the presence of an immunosuppressive microenvironment(ISM).Irreversible electroporation(IRE)can not only induce immunogenic cell death,but also alleviate *** study aimed to investigate the antitumor efficacy of IRE plus allogeneicγδT cells in LAPC patients.A total of 62 patients who met the eligibility criteria were enrolled in this trial,then randomized into two groups(A:n=30 and B:n=32).All patients received IRE therapy and after receiving IRE,the group A patients received at least two cycles ofγδT-cell infusion as one course *** A patients had better survival than group B patients(median OS:14.5 months vs.11 months;median PFS:11 months vs.8.5 months).Moreover,the group A patients treated with multiple courses ofγδT-cell infusion had longer OS(17 months)than those who received a single course(13.5 months).IRE combined with allogeneicγδT-cell infusion is a promising strategy to enhance the antitumor efficacy in LAPC patients,yielding extended survival benefits.

Effectively killing pathogenic bacteria is key for the treatment of *** various anti-infective drugs have been used for the treatment of sepsis,the therapeutic effect is largely limited by the lack of a specific bacterium-targeting delivery *** study aimed to develop antibacterial peptides that specifically target pathogenic bacteria for the treatment of *** lethal bacterial strain Escherichia coli MSI001 was isolated from mice of a cecal ligation and puncture(CLP)model and was used as a target to screen bacterial binding heptapeptides through an integrative bioinformatics approach based on phage display technology and high-throughput sequencing(HTS).Heptapeptides binding to *** MSI001 with high affinity were acquired after normalization by the heptapeptide frequency of the library.A representative heptapeptide VTKLGSL(VTK)was selected for fusion with the antibacterial peptide LL-37 to construct the specific-targeting antibacterial peptide *** found that,in comparison with LL37,VTK-LL37 showed prominent bacteriostatic activity and an inhibitive effect on biofilm formation in *** vivo experiments demonstrated that VTK-LL37 significantly inhibited bacterial growth,reduced HMGB1 expression,alleviated lesions of vital organs and improved the survival of mice subjected to CLP ***,membrane DEGP and DEGQ were identified as VTKbinding proteins by proteomic *** study provides a novel strategy for targeted pathogen killing,which is helpful for the treatment of sepsis in the era of precise medicine.

Formamidine lead triiodide(FAPbI_(3))perovskites have become the most promising photovoltaic materials for perovskite solar cells with record power conversion efficiency(PCE).However,random nucleation,phase transition,and lattice defects are still the key challenges limiting the quality of FAPbI_(3) *** studies show that the introduction or adding of seeds in the precursor is effective to promote the nucleation and crystallization of perovskite ***,the seed-assisted approach focuses on heterogeneous seeds or hetero-composites,which inevitably induce a lattice-mismatch,the genera-tion of strain or defects,and the phase segregation in the perovskite ***,we first demonstrate that high-quality perovskite films are controllably prepared using α-and δ-phases mixed FAPbI_(3) micro-crystal as the homogeneous seeds with the one-step antisolvent *** partially dissolved seeds with suitable sizes improve the crystallinity of the perovskite flm with preferable orientation,improved carrier lifetime,and increased carrier *** importantly,the α-phase-containing seeds promote the formation of α-phase FAPbI_(3) films,leading to the reduction of residual lattice strain and the suppres-sion of I-ion ***,the adding of dimethyl 2,6-pyridine dicarboxylate(DPD)into the pre-cursor further suppresses the generation of defects,contributing to the PCE of devices prepared in air ambient being significantly improved to 23.75%,among the highest PCEs for fully air-processed FAPbI_(3) solar *** unpackaged target devices possess a high stability,maintaining 80%of the initial PCE under simulated solar illumination exceeding 800 h.

目的探讨MRP8/MRP14诱导小鼠腹腔巨噬细胞细胞因子表达效应及其作用机制。方法 Luminex x MAP液相芯片系统检测重组MRP8/MRP14蛋白诱导腹腔巨噬细胞6种细胞因子/趋化因子的水平变化;MRP14不同结构域融合蛋白刺激细胞,检测TNF-α、IP-10和IL-6表达;Western blot检测MRP8/MRP14刺激细胞p38 MAPK、JNK和ERK激酶磷酸化变化;细胞预先用p38 MAPKs、JNK、ERK激酶抑制剂、TLR4和RAGE受体拮抗剂预处理,之后给予MRP8/MRP14蛋白刺激,检测TNF-α、IP-10和IL-6表达。结果 MRP8/MRP14能显著诱导TNF-α、IP-10和IL-6的表达,与对照组相比,蛋白表达水平分别升高约98.2、378.6和6.3倍(P<0.01),MRP8/MRP14不能诱导IL-2、IL-5和IFN-g的表达;MRP14全长及其结构域EFhand-1、EFhand-2及EFhand-1+2融合蛋白能够诱导TNF-α、IP-10和IL-6的表达(P<0.01),CT末端结构域不具有诱导活性;MRP8/MRP14刺激细胞后1 h p38 MAPK、JNK及ERK激酶发生显著磷酸化变化,持续至2 h;与单纯MRP8/MRP14组相比,p38 MAPK抑制剂SB203580显著抑制TNF-α、IP-10和IL-6的表达(P<0.05);JNK抑制剂SP600125显著抑制TNF-α和IP-10的表达(P<0.05),对IL-6的表达无影响;ERK及其上游MEK1/2的抑制剂PD98059和U0126显著抑制IL-6的表达(P<0.05);TLR4抑制剂TAK242抑制了MRP8/MRP14诱导的TNF-α、IP-10和IL-6的表达(P<0.05),而RAGE中和性抗体仅部分抑制MRP8/MRP14诱导的IL-6的表达(P<0.05)。结论 MRP8/MRP14能够诱导小鼠腹腔巨噬细胞TNF-α、IP-10和IL-6的表达;MRP蛋白以包含有钙离子结合基序的结构域具有诱导细胞因子表达的活性;TNF-α和IP-10的表达与TLR4受体及其下游的p38 MAPKs、JNK通路有关,IL-6的表达则同时由TLR4和RAGE受体介导,继而激活下游的p38 MAPKs和ERK信号通路。

目的探讨RNA结合蛋白HuR 118位苏氨酸对热应激状态下的双特异性磷酸酶1(DUSP1)转录后调控机制。方法构建HuR 118位苏氨酸突变体真核表达质粒,并用脂质体转染NIH 3T3细胞,Real-time PCR检测其对DUSP1 mRNA水平的影响,Western blotting检测其对DUSP1蛋白表达的效应。结果 (1)小鼠HuR不同突变体真核表达载体质粒构建成功;(2)热休克刺激后,HuRT118磷酸化明显增强;(3)过表达HuR(WT)与HuR(T118E),经过热休克刺激后,DUSP1 mRNA水平明显升高,其蛋白表达也有上调;过表达HuR(T118E)时,DUSP1的mRNA水平明显上调,而蛋白水平在热休克刺激前后均上调,其表达变化无差异而过表达HuR(T118A),热休克刺激前后DUSP1 mRNA水平及蛋白表达均下调;(4)热休克刺激下,HuR(WT)可以从细胞核内转位到细胞质并形成颗粒;当过表达HuR(T118E)时,在静息状态下,HuR(T118E)弥散分布于细胞质,热休克刺激会使其在细胞质形成明显的颗粒;而过表达HuR(T118A)时,热休克刺激并不能使其移位出核。结论 RNA结合蛋白HuR参与了DUSP1的转录后调控,可以通过其118位的苏氨酸磷酸化提高热休克刺激后的DUSP1的基因及蛋白水平。
