Mechanisms underlying aspirin-mediated growth inhibition and apoptosis induction of cyclooxygenase-2 negative colon cancer cell line SW480

AIM： To investigate the effects of aspirin （acetylsalicylic acid） on proliferation and apoptosis of colorectal cancer cell line SW480 and its mechanism. METHODS： Cyclooxygenase （COX）-2 negative colorectal cancer cell line SW480 was treated with aspirin at concentrations of 2.5 mmol/L, 5.0 mmol/L, 10.0 mmol/L for different periods in v/tro. Anti-proliferation effect of aspirin on SW480 was detected by 3-（4,5-dimeth- ylthiazol-2-yl）-2,5-diphenyltetrazolium bromide （MTT） assay. Cell cycle and apoptosis were observed by flow cytometry （FCM）. Transmission electron microscope （TEM） was used for morphological study. Apoptosis-associated genes were detected by immunohistochemical staining and Western blotting. RESULTS： Aspirin inhibited SW480 proliferation and induced apoptosis in a dose- and time-dependent manner. Treatment with different concentrations of aspirin significantly increased the proportions of cells at the G0/G1 phase and decreased the proportions of cells at the S- and G2/M phases in a concentration-dependent manner. Aspirin not only induced apoptosis but also caused cell necrosis at a high concentration as well. After treatment with aspirin, SW480 cells displayed typically morphological features of apoptosis and necrosis under TEM, and increased the Bcl-2 expression in cells, but the expression of Bax was down regulated. CONCLUSION： Aspirin inhibits proliferation and induces apoptosis of SW480 cells. Its anti-tumor mechanism may arrest cell cycle and shift Bax/Bcl-2 balance in cells.

Role and recruitment of Th9 cells in liver cirrhosis patients

Surface modification of graphite anode with electroactive matters has been proven of a more practical strategy in enhancing the performance of Li-ion batteries than exploring alternative novel anode ***,rutile TiNbO_(4-x) nanoparticles with a tunnel structure are employed as multifunctional decoration substances in combination with a carbon coating layer to improve the rate and cycle properties of mesocarbon microbeads(MCMBs).As compared to pristine MCMB,the Li^(+)diffusion coefficients of the composite anodes are enhanced due to the synergistic effect of TiNbO_(4-x)@***,the overcharge and voltage polarization of the composite anodes at high rate are obviously minimized due to the current sharing effect of the high-potential TiNbO_(4-x).Moreover,the amorphous Li_(y)TiNbO_(4-x) converted from TiNbO_(4-x) in the initial lithiation process can deliver pseudocapacitive capacity to the composite anodes from the second *** of these functions of TiNbO_(4-x)@Ccoating layer have directly contributed to the improved rate and cycle performance of the MCMB/TiNbO_(4-x)@C composite *** one containing 12.0 wt%TiNbO_(4-x) exhibits a high reversible specific capacity of 118 m Ah·g^(-1)at 10C(1C=372 m A·g^(-1)),together with a high capacity retention of 90.9%after 300 cycles at 3C,which are all much superior to those of pristine MCMB.

BACKGROUND Gallbladder and biliary diseases(GABDs)are a major public health *** To analysis the cause-specific incidence,prevalence,and years lived with disability(YLDs)and its temporal trends of GABDs at the global,regional,and national *** on GABD were available from the Global Burden of Disease study *** The estimated annual percentage change(EAPC)was used to quantify temporal trend in GABD age-standardized incidence rates(ASIRs),age-standardized prevalence rate(ASPR),and age-standardized YLD rate(ASYR)by region,*** analyzed the relationship between the GABD burden and country development level using the human development index(HDI).RESULTS In 2019,the incident cases of GABD were 52003772,with an ASIR of 63432/100000 ***,the number of incident cases and ASIR of GABD increased 97%and 58.9%between 1990 and ***,the ASPR and ASYR decreased from 1990 to 2019,the number of prevalent and YLDs cases *** highest ASIR was observed in Italy,and the highest ASPR and ASYR was observed in United *** highest burden of GABD was found in low-SDI region,and the burden in female was significantly higher than males.A generally negative correlation(ρ=-0.24,P<0.05)of GABD with the EAPC and human development index(HDI)(in 2021)were observed for ***’s more,no correlation in ASPR(ρ=-0.06,P=0.39)and ASYR(ρ=-0.07,P=0.36)of GABD with the EAPC and HDI(in 2021)were observed,*** GABD remain a major global public health challenge;however,the burden of GABD varies ***,the number of incident cases and ASIR of GABD increased between 1990 and *** results of our study provide insight into the global disease burden of GABD and may assist policymakers in formulating effective policies to mitigate modifiable risk factors.

Role of Th9 cells and Th17 cells in the pathogenesis of malignant ascites

Objective: To assess the role of Th9 and Th17 cells in malignant ascites(MA).Methods: MA from 30 hepatic carcinoma patients and benign ascites from 30 cirrhotic patients were collected. Corresponding peripheral blood samples from these hepatic carcinoma and cirrhotic patients as well as 30 healthy subjects were collected. The frequency of Th9 and Th17 cells was tested by flow cytometry. Serum levels of interleukin(IL)-9 and IL-17 were examined by ***: The observed frequency of Th9 and Th17 cells, and the IL-9 and IL-17 serum levels were significantly higher in MA patients than those in cirrhotic patients and healthy control samples(P < 0.05). Moreover, the Th9 cells demonstrated positive correlation with Th17 cells as well as IL-9 in MA patients; however, this positive correlation was not observed in the cirrhotic patients or healthy control samples. The frequency of Th9 and Th17 cells was distinctly higher in MA patients presenting with stage III or IV malignancy and with lymph node or distant metastasis than those in patients in stage I or II and without distant metastasis(P < 0.05).Conclusions: The increased frequency of Th9 and Th17 cells in MA patients suggests that these two T cell subsets play a synergistic role in MA pathogenesis. This study also demonstrated that Th9 and Th17 cells may perform their biological functions in conjunction with IL-9 production.
