This study was designed to clone cDNA of goat DNA methyltransferase 1（DNMT1） gene,to screen an effective shRNAproducing vector targeting goat DNA methyltransferase 1 and to improve the developmental competence of goat nuclear transfer embryos by decreasing the DNMT1 expression in donor *** this study,PCR primers were designed against regions of high homology between bovine and sheep sequences and then used to amplify the larger portions of the coding ***,3 RNAi oligonucleotides were designed based on the cloned sequences and inserted into pRNAT-U6.1/Neo vector,acquiring 3 new vectors,respectively termed pRNAD1,pRNAD2 and *** the positive cells were sorted by flow cytometry after transfection and detected by real-time PCR analysis and sodium bisulfite genomic ***,the developmental rates of nuclear transfer（NT） embryos generated using donor cells with and without the effective shRNA vector respectively,as well as in vitro fertilization（IVF） embryos were observed and *** results showed that the coding regions of goat DNA methyltransferase 1 gene was successfully cloned（GenBank ***617538）.Furthermore,an effective interfering shRNA（pRNAD2） was obtained,with its interference effect being 47.88%.Finally,NT embryos with shRNA vector harbored better developmental competence during morula and blastocyst stage compared to controls（P 〈 0.05）,reaching the similar rates to IVF embryos（P 〉 0.05）.In conclusion,goat DNA methyltransferase 1 gene cDNA was cloned and sequenced,an effective shRNA vector responsible for inhibiting DNA methyltransferase 1 expression was developed and the developmental competence of goat nuclear transfer morulae and blastcysts was significantly improved,which provided a feasible pathway for improving goat nuclear transfer embryo development competence by decreasing the methylation level in donor cells through RNAi-mediated manner.

ZrO_(2)-strengthened porous mullite insulation materials were prepared by foaming technology utilizing ZrSiO_(4) and Al_(2)O_(3) as primary materials and Y_(2)O_(3) as an *** effects of Y_(2)O_(3) contents on the phase composition,microstructure,mechanical properties,and heat conductivity of the porous mullite insulation materials were investigated.A suitable Y_(2)O_(3) content could promote phase transition of monoclinic ZrO_(2)(m-ZrO_(2))to tetragonal ZrO_(2)(t-ZrO_(2)),reduce pore size,and improve the strengths of as-prepared *** cold crushing strength and bending strength of as-prepared specimens with a 119µm spherical pore size using 6 wt.%Y_(2)O_(3) were 35.2 and 13.0 MPa,respectively,with a heat conductivity being only 0.248 W/(m K).

Social hierarchy is a general self-organizational scheme in animal societies in which each member achieves a certain status of domination or subordination when getting access to resources,with profound consequences for survival,health,reproductive success,and multiple behaviors[1,2].The prefrontal cortex(PFC)was recognized as a central regulator of neuronal circuits determining the formation of social hierarchy;brain areas upstream of the PFC convey information about social status,and downstream brain regions execute dominance behavior[3].Social deficits have been suggested to be due to an elevation of excitatory/inhibitory(E/l)balance within the mouse dorsomedial(dm)PFC[4].A compensatory increase of inhibitory cell excitability partially rescued social deficits caused by such a disturbance of E/l balance.

Effect of low-speed waterjet pressure on the rock-breaking performance of unsubmerged cavitating abrasive waterjet

Unsubmerged cavitating abrasive waterjet(UCAWJ) has been shown to artificially create a submerged environment that produces shear cavitation, which effectively enhances rock-breaking performance. The shear cavitation generation and collapse intensity depend on the pressure difference between the intermediate high-speed abrasive waterjet and the coaxial low-speed waterjet. However, the effect of the pressure of the coaxial low-speed waterjet is pending. For this purpose, the effect of low-speed waterjet pressure on rock-breaking performance at different standoff distances was experimentally investigated,and the effects of erosion time and ruby nozzle diameter on erosion performance were discussed. Finally,the micromorphology of the sandstone was observed at different locations. The results show that increased erosion time and ruby nozzle diameter can significantly improve the rock-breaking performance. At different standoff distances, the mass loss increases first and then decreases with the increase of low-speed waterjet pressure, the maximum mass loss is 10.4 g at a low-speed waterjet pressure of0.09 MPa. The surface morphology of cavitation erosion was measured using a 3D profiler, the increase in both erosion depth and surface roughness indicated a significant increase in the intensity of the shear cavitation collapse. At a low-speed waterjet pressure of 0.18 MPa, the cavitation erosion surface depth can reach 600 μm with a roughness of 127 μm.

Using Copula functions to predict climatic change impacts on floods in river source regions

Flood frequency in river source regions is significantly affected by rainfall and snowmelt as part of climatic changes.A traditional univariate flood frequency analysis cannot reflect the complexity of floods,and when used in isolation,it can only underestimate flood *** effective flood prevention and mitigation,it is essential to consider the combined effects of precipitation and *** functions can effectively quantify the joint distribution relationship between floods and their associated variables without restrictions on their distribution *** study uses copula functions to consider a multivariate probability distribution model of flood peak flow(Q)with cumulative snowmelt(CSm)and cumulative precipitation(CPr)for the Hutubi River basin located in northern Xinjiang,*** joint frequencies of rainfall and snowmelt floods are predicted using copula models based on the Coupled Model Intercomparison Project Phase 6 *** results show that Q has a significant positive correlation with 24-d CSm(r=0.559,p=0.002)and 23-d CPr(r=0.965,p<0.05).Flood frequency will increase in the future,and mid-(2050e2074)and long-term(2075e2099)floods will be more severe than those in the near-term(2025e2049).The probability of flood occurrence is higher under the SSP2-4.5 and SSP1-2.6 scenarios than under *** during the historical period(1990e2014)led to extreme floods,and increasing future precipitation trends are found to be *** increases with rising temperatures and occurs earlier than estimated,leading to an earlier flood period in the basin and more frequent snowmelt *** Q under the joint return period is larger than that during the same univariate return *** difference indicates that neglecting the interaction between precipitation and snowmelt for floods leads to an underestimation of the flood risk(with underestimations ranging from 0.3%to 22%).The underestimations decrease with an increase

A rare missense PAX6 mutation causes atypical aniridia in a three-generation Chinese family

●AIM:To investigate the molecular diagnosis of a threegeneration Chinese family affected with aniridia,and further to identify clinically a PAX6 missense mutation in members with atypical aniridia.●METHODS:Eleven family members with and without atypical aniridia were *** family members underwent comprehensive ophthalmic examinations.A combination of whole exome sequencing(WES)and direct Sanger sequencing were performed to uncover the causative mutation.●RESULTS:Among the 11 family members,8 were clinically diagnosed with congenital aniridia(atypical aniridia phenotype).A rare heterozygous mutation c.622C>T(***208Trp)in exon 8 of PAX6 was identified in all affected family members but not in the unaffected members or in healthy control subjects.●CONCLUSION:A rare missense mutation in the PAX6 gene is found in members of a three-generation Chinese family with congenital atypical *** result contributes to an increase in the phenotypic spectrum caused by PAX6 missense heterozygous variants and provides useful information for the clinical diagnosis of atypical aniridia,which may also contribute to genetic counselling and family planning.

To ensure the safe performance of deep-sea mining vehicles(DSMVs),it is necessary to study the mechanical characteristics of the interaction between the seabed soil and the track *** rotation and digging motions of the track plate are important links in the contact between the driving mechanism of the DSMV and seabed *** this study,a numerical simulation is conducted using the coupled Eulerian–Lagrangian(CEL)large deformation numerical method to investigate the interaction between the track plate of the DSMV and the seabed soil under two working conditions:rotating condition and digging ***,a soil numerical model is established based on the elastoplastic mechanical characterization using the basic physical and mechanical properties of the seabed soil obtained by in situ ***,the soil disturbance mechanism and the dynamic mechanical response of the track plate under rotating and digging conditions are obtained through the analysis of the sensitivity of the motion parameters,the grouser structure,the layered soil features and the soil *** results indicate that the above parameters remarkably influence the interaction between the DSMV and the seabed ***,it is important to consider the rotating and digging motion of the DSMV in practical engineering to develop a detailed optimization design of the track plate.

Geochemical characteristics,generation,and evolution mechanism of coalbed methane in the south-western Ordos Basin,China

The south-western Ordos Basin is rich in low-middle rank coalbed methane(CBM)resources;while the geochemical characteristics and genetic mechanism of CBM are not ***,according to geological and geochemical test data from gas and coal seam water from CBM wells in Bingchang,Jiaoxun,Huangling,yonglong,and Longdong minging areas,we systematically studied the geochemical characteristics,generation,and evolution mechanism of CBM in Jurassic Yan’an Formation in the south-western Ordos *** results show that the CH4 content of whole gas is in the range of 42.01%-94.72%.The distribution ranges of theδ^(13)C-CH_(4)value is−87.2‰to−32.5‰,indicating diverse sources of thermogenic gas and biogenic *** microbial methane is mainly generated by a CO_(2)reduction pathway,with certain methyl-type fermentation ***δ^(13)C-CH_(4)has a positive correlation with burial depth,indicating the obvious fractionation of *** relationship between the genetic types and burial depth of the CBM reservoir indicates that the favorable depth of secondary biogenic gas is less than 660 *** Late Cretaceous Yanshanian Movement led to the uplift of the Ordos Basin,and a large amount of thermogenic gas escaped from the edge of the *** the Paleogene Period,the coal reservoir in the basin margin has received recharge from atmospheric precipitation,which is favorable for the formation of secondary biogenic *** deep area,generally under 1000 m,mainly contains residual thermogenic *** intermediate transition zone is mixed *** by the tectonic background,the genetic types of CBM in different mining areas are controlled by the coupling of burial depth,coal rank,and hydrogeological *** Binchang mining area contains biogenic gas,and the development of CBM has achieved initial success,indicating that similar blocks with biogenic gas formation conditions is key to the efficient development of *** research results provide a scientific basis for
