AIM: Age-related macular degeneration (AMD) is a multifactorial disease and a prevalent cause of visual impairment in developed countries. Many studies suggest that age-related maculopathy susceptibility 2 (ARMS2) is a second major susceptibility gene for AMD. At present, there is no functional information on this gene. Therefore, the purpose of the present study was to detect the expression of ARMS2 in retinal pigment epithelium (RPE) cells and to investigate the effect of ARMS2 on the phagocytosis function of RPE cells. METHODS: Immunofluorescence and reverse transcriptase PCR were used to demonstrate the presence and location of ARMS2 in ARPE-19 (human retinal pigment epithelial cell line, ATCC, catalog ***-2302) cells. siRNA was used to knock down ARMS2 mRNA, and the effects of the knockdown on the phagocytosis function of the ARPE-19 cells were evaluated via Fluorescence Activated Cell Sorting (FACS). RESULTS: ARMS2 was present in ARPE-19 cells, localized in the cytosol of the perinuclear region. The expression of ARMS2 mRNA (messenger RNA) in ARPE-19 cells transfected with ARMS2-siRNA (small interfering RNA, 0.73+/- 0.08) was decreased compared with normal cells (1.00+/- 0.00) or with cells transfected with scrambled siRNA (0.95+/- 0.13) (P<0.05). After incubation of RPE cells with a latex beads medium for 12, 18, or 24 hours, the fluorescence intensities were 38.04 +/- 1.02, 68.92 +/- 0.92, and 78.00 +/- 0.12 in the ARMS2-siRNA-transfected groups, respectively, and 77.98 +/- 5.43, 94.87 +/- 0.60, and 98.30 +/- 0.11 in the scrambled siRNA-transfected groups, respectively. The fluorescent intensities of the same time points in the two groups were compared using Student's t-test, and the p values were all less than 0.001 at the three different time points. CONCLUSION: There is endogenous expression of ARMS2 in ARPE-19 cells. Comparison of microstructure and property of high chromium bearing steel with and without nitrogen addition

Microstructure and property of bearing steel with and without nitrogen addition were investigated by microstructural observation and hardness measurement after different heat treatment processing. Based on the microstructural observation of both 9Cr18 steel and X90N steel, it was found that nitrogen addition could effectively reduce the amount and size of coarse carbides and also refine the original austenite grain size. Due to addition of nitrogen, more austenite phase was found in X90N steel than in 9Cr18 steel. The retained austenite of X90N steel after quenching at 1050℃ could be reduced from about 60% to about 7 9% by cold treatment at -73℃ and subsequent tempering, and thus finally increased the hardness up to 60 HRC after low temperature tempering and to 63 HRC after high temperature tempering. Furthermore, both the wear and corrosion resistance of X90N steel were found much more superior than those of 9Cr18 steel, which was attributed to the addition of nitrogen. It was proposed at last that nitrogen alloying into the high chromium bearing steel was a promising way not only to refine the size of both carbides and austenite, but also to achieve high hardness, high wear property and improved corrosion resistance of the stainless bearing steel.

A series of carbosilane dendrimers with acetyl end-group:From synthesis to their unique optical characteristics

A series of c arbosilane dendrimers with acetyl end-group were synthesized. Their structures were characterized by 1H NMR, IR, and MS, respectively. Then dendrimers were coordinated with lanthanide ions （Eu3＋ and Tb3＋, respectively）. The luminescence spectra of the complexes show narrow-width emissions in visible light region. 2009 Sheng Yu feng. Study on superprism effect in the multilayer optical thin film stack

Researches show that multilayer optical thin film stack can exhibit superprism effect due to their large abnormal dispersions. We investigated and simulated this effect numerically in a 1-D non-periodic film structure-Fabry-Perot filters (FPF), which possess drastic change in phase and large group delay around wavelength of peak transmittance, and fabricated this device to realize remarkable superprism effect. We tested experimentally with the maximum spatial separation shift up to 65 μm, and the experimental result is in good agreement with the theory. Compared with the traditional prism, the total thickness of our structure is only 3.3 μm, and our prism shows a stronger angular resolution of 1.8°/nm.

This editorial discusses a case report recently published in the World Journal of Clinical *** report describes the clinical presentation,imaging,diagnosis,and treatment of a patient with tuberous sclerosis complex(TSC)combined with primary lymphedema(PLE).Additionally,it retrospectively analyzes the data of 16 previously reported cases of children with TSC combined with PLE to summarize the epidemiology,genetic diagnosis,and current main treatments of these *** report also speculates on the pathological and physiological mechanisms underlying TSC combined with *** combined with PLE is rare;therefore,the report provides a theoretical basis for understanding the pathophysiological mechanisms and treatment options for patients with TSC and *** clinical management of TSC is essential due to the diverse and multiorgan nature of its manifestations,often requiring a multidisciplinary approach for newly diagnosed cases.

Microstructure, hardness and contact fatigue properties of X30N high nitrogen stainless bearing steel

Microstructure, hardness and fatigue properties of X30N high nitrogen stainless bearing steel were investigated. It was found that nitrogen addition could effectively reduce the amount and size of coarse carbides. The original austenite grain size was obviously refined. Additionally, more retained austenite was found in X30N steel after quenching at 1050 ℃, which could be reduced from about 30% to about 6% by cold treatment at - 73 ℃ and subsequent tempering, and thus, the ultimate hardness was increased up to about 61 HRC with reduction of austenite and precipitation of carbonitrides. Furthermore, the rolling contact fatigue lives of X30N steel ate superior to those of 440C steel, which was attributed to the enhanced hardness and a certain retained austenite in the high nitrogen steel.

Effect of bevacizumab on the expression of fibrosis-related inflammatory mediators in ARPE-19 cells

AIM：To investigate the effect of anti-vascular epithelial growth factor（VEGF）agents on the expression of fibrosisrelated inflammatory mediators under normoxic and hypoxic conditions,and to further clarify the mechanism underlying fibrosis after anti-VEGF therapy. METHODS：Human retinal pigment epithelial（RPE）cells were incubated under normoxic and hypoxic *** hypoxia treatment,CoCl_2 at 200μmol/L was added to the media. ARPE-19 cells were treated as following：1）control group：no treatment; 2）bevacizumab group：bevacizumab at 0.25 mg/mL was added to the media; 3）hypoxia group：CoCl_2 at 200 μmol/L was added to the media; 4）hypoxia＋bevacizumab group：CoCl_2 at 200 μmol/L and bevacizumab at 0.25 mg/mL were added to the *** expression of interleukin（IL）-1β,IL-6,IL-8 and tumor necrosis factor（TNF）-α were evaluated using real-time polymerase chain reaction（RT-PCR）and enzyme-linked immunosorbent assay（ELISA）at 6,12,24 and 48 h. RESULTS：Both m RNA and protein levels of IL-1β,IL-6 and IL-8 were statistically significantly higher in the bevacizumab group than in the control group at each time point,and TNF-α gene and protein expression was only significantly higher only at 24 and 48h（P〈0.05）. Under hypoxic conditions,bevacizumab significantly increased the expression of IL-1β,IL-6,IL-8 and TNF-α at 6,12,24 and 48h（P〈0.05）. IL-1β,IL-8 and TNF-α peaked at 24 h and IL-6 peaked at 12 h after the bevacizumab treatment under both normoxic and hypoxic conditions. CONCLUSION：Treatment of ARPE-19 cells with bevacizumab can significantly increase the expression of fibrosis-related inflammatory mediators under bothnormoxic and hypoxic conditions. Inflammatory factors might be involved in the process of fibrosis after antiVEGF therapy,and the up-regulation of inflammatory factors induced by anti-VEGF drugs might promote the fibrosis process.

Optimized design of LED freeform lens for uniform circular illumination

An optimization method is proposed for designing an LED freeform lens which produces a uniform circular pattern with high energy *** method is composed of three main aspects:design of the initial guess,parameterization of the freeform surface,and construction of the merit *** initial guess is created by solving an ordinary differential equation *** approach of selecting optimization points is introduced for parameterization of the freeform *** merit function is constructed by use of the irradiance uniformity and the efficiency of the *** examples are given,and the results show that the irradiance distribution is well controlled with a maximum uniformity(the relative standard deviation of irradiance,RSD) of 0.0122 and a maximum efficiency of 93.88%.This optimization method can be generalized to design freeform lenses with different lighting patterns or without rotational symmetry.

Chemoembolization alone vs combined chemoembolization and hepatic arterial infusion chemotherapy in inoperable hepatocellular carcinoma patients

AIM: To compare the efficacy and safety of chemoem-bolization alone or chemoembolization combined with hepatic arterial infusion chemotherapy(haiC),including oxaliplatin(OXA),5-fluorouracil(5-FU) and folinic acid(CF),in inoperable hepatocellular carcinoma(HCC) without distant metastasis. METHODS: Eighty-four inoperable HCC patients were enrolled. Thirty-ninepatient sunderwent chemoembolization alone,and the other 45 patients underwent chemoembolization + haiC(OXA/5-FU/CF) treatment non-randomly. The progression free survival(PFS),objective response rate(ORR),disease control rate(DCR) and adverse reactions were compared between the two ***: A significant difference in the ORR was observed between the chemoembolization alone and chemoembolization + haiC groups. There was no statistically significant difference in DCR between the two groups. The median PFS(m PFS) showed a significant difference between the two groups. For patients with BCLC stage A/B disease,with or without vessel invasion,the chemoembolization + haiC group showed better m PFS when compared to chemoembolization alone,but no significant difference was found in patients with BCLC stage C disease. The parameter of pain(grade Ⅲ-Ⅳ) in the chemoembolization + haiC group was increased statistically. CONCLUSION: Chemoembolization combined with haiC with OXA/5-FU/CF may be safe and more effective than chemoembolization alone for inoperable HCC patients without distant metastasis.
