Maintaining moderate levels of hypochlorous acid promotes neural stem cell proliferation and differentiation in the recovery phase of stroke

It has been shown clinically that continuous removal of ischemia/reperfusion-induced reactive oxygen species is not conducive to the recovery of late ***,previous studies have shown that excessive increases in hypochlorous acid after stroke can cause severe damage to brain *** previous studies have found that a small amount of hypochlorous acid still exists in the later stage of stroke,but its specific role and mechanism are currently *** simulate stroke in vivo,a middle cerebral artery occlusion rat model was established,with an oxygen-glucose deprivation/reoxygenation model established in vitro to mimic *** found that in the early stage(within 24 hours)of ischemic stroke,neutrophils produced a large amount of hypochlorous acid,while in the recovery phase(10 days after stroke),microglia were activated and produced a small amount of hypochlorous ***,in acute stroke in rats,hypochlorous acid production was prevented using a hypochlorous acid scavenger,taurine,or myeloperoxidase inhibitor,4-aminobenzoic acid *** results showed that high levels of hypochlorous acid(200μM)induced neuronal apoptosis after oxygen/glucose deprivation/***,in the recovery phase of the middle cerebral artery occlusion model,a moderate level of hypochlorous acid promoted the proliferation and differentiation of neural stem cells into neurons and *** suggests that hypochlorous acid plays different roles at different phases of cerebral ischemia/reperfusion *** levels of hypochlorous acid(5 and 100μM)promoted nuclear translocation ofβ-*** transfection of single-site mutation plasmids,we found that hypochlorous acid induced chlorination of theβ-catenin tyrosine 30 residue,which promoted nuclear ***,our study indicates that maintaining low levels of hypochlorous acid plays a key role in the recovery of neurological function.

Ischemic accumulation of succinate induces Cdc42 succinylation and inhibits neural stem cell proliferation after cerebral ischemia/reperfusion

Ischemic accumulation of succinate causes cerebral damage by excess production of reactive oxygen species. However, it is unknown whether ischemic accumulation of succinate affects neural stem cell proliferation. In this study, we established a rat model of cerebral ischemia/reperfusion injury by occlusion of the middle cerebral artery. We found that succinate levels increased in serum and brain tissue(cortex and hippocampus) after ischemia/reperfusion injury. Oxygen-glucose deprivation and reoxygenation stimulated primary neural stem cells to produce abundant succinate. Succinate can be converted into diethyl succinate in cells. Exogenous diethyl succinate inhibited the proliferation of mouse-derived C17.2 neural stem cells and increased the infarct volume in the rat model of cerebral ischemia/reperfusion injury. Exogenous diethyl succinate also increased the succinylation of the Rho family GTPase Cdc42 but repressed Cdc42 GTPase activity in C17.2 cells. Increasing Cdc42 succinylation by knockdown of the desuccinylase Sirt5 also inhibited Cdc42 GTPase activity in C17.2 cells. Our findings suggest that ischemic accumulation of succinate decreases Cdc42 GTPase activity by induction of Cdc42 succinylation, which inhibits the proliferation of neural stem cells and aggravates cerebral ischemia/reperfusion injury.

RE-SEGNN:recurrent semantic evidence-aware graph neural network for temporal knowledge graph forecasting

Temporal knowledge graph(TKG) reasoning, has seen widespread use for modeling real-world events, particularly in extrapolation settings. Nevertheless, most previous studies are embedded models, which require both entity and relation embedding to make predictions, ignoring the semantic correlations among different entities and relations within the same timestamp. This can lead to random and nonsensical predictions when unseen entities or relations occur. Furthermore, many existing models exhibit limitations in handling highly correlated historical facts with extensive temporal depth. They often either overlook such facts or overly accentuate the relationships between recurring past occurrences and their current counterparts. Due to the dynamic nature of TKG, effectively capturing the evolving semantics between different timestamps can be *** address these shortcomings, we propose the recurrent semantic evidenceaware graph neural network(RE-SEGNN), a novel graph neural network that can learn the semantics of entities and relations simultaneously. For the former challenge, our model can predict a possible answer to missing quadruples based on semantics when facing unseen entities or relations. For the latter problem, based on an obvious established force, both the recency and frequency of semantic history tend to confer a higher reference value for the current. We use the Hawkes process to compute the semantic trend, which allows the semantics of recent facts to gain more attention than those of distant facts. Experimental results show that RE-SEGNN outperforms all SOTA models in entity prediction on 6 widely used datasets, and 5 datasets in relation prediction. Furthermore, the case study shows how our model can deal with unseen entities and relations.

Influence of minor cerium addition on microstructure and fluidity of as-cast Al-Cu-Mn-Mg alloy

Al-Cu-Mn alloys are widely used to produce automobile components like cylinder heads and engine blocks because of their capability to retain excellent thermal and mechanical characteristics at high ***,the Al-Cu-Mn-based alloys demonstrate restricted fluidity,leading to casting defects such as shrinkage and incomplete *** research investigated the microstructure and fluidity of Al-4.7Cu-1.0Mn-0.5Mg（wt%） alloy with minor cerium（Ce） *** as-cast alloys predominantly comprise α-Al matrix,accompanied by the presence of Al2Cu,Al6Mn,and Al8Cu4Ce *** influence of adding Ce on the fluidity of the Al-4.7Cu-1.0Mn-0.5Mg alloy was investigated using a trispiral fluidity test mold in this *** findings suggest that the addition of Ce within the range of 0.1 wt% to 0.5 wt% in the Al-4.7Cu-1.0Mn-0.5Mg alloy results in an enhancement in ***,the alloy containing 0.4 wt% Ce exhibits a significant increase in fluidity distance,from 349.7 to 485.7 *** improvement can be attributed to the reduction in viscosity,the refinement of secondary dendrite arm spacing,and the modification of secondary phase ***,a higher concentration of Ce leads to a decrease in fluidity length,potentially due to the formation of Al8Cu4Ce.

The mechanical behavior of cemented gangue backfill materials(CGBMs)is closely related to particle size distribution(PSD)of aggregates and properties of cementitious ***,the true triaxial compression tests,CT scanning,SEM,and EDS tests were conducted on cemented gangue backfill samples(CGBSs)with various carbon nanotube concentrations(P_(CNT))that satisfied fractal theory for the PSD of *** mechanical properties,energy dissipations,and failure mechanisms of the CGBSs under true triaxial compression were systematically *** results indicate that appropriate carbon nanotubes(CNTs)effectively enhance the mechanical properties and energy dissipations of CGBSs through micropore filling and microcrack bridging,and the optimal effect appears at P_(CNT)of 0.08wt%.Taking PSD fractal dimension(D)of 2.500 as an example,compared to that of CGBS without CNT,the peak strength(σ_(p)),axial peak strain(ε_(1,p)),elastic strain energy(Ue),and dissipated energy(U_(d))increased by 12.76%,29.60%,19.05%,and90.39%,***,excessive CNTs can reduce the mechanical properties of CGBSs due to CNT agglomeration,manifesting a decrease inρ_(p),ε_(1,p),and the volumetric strain increment(Δε_(v))when P_(CNT)increases from 0.08wt%to 0.12wt%.Moreover,the addition of CNTs improved the integrity of CGBS after macroscopic failure,and crack extension in CGBSs appeared in two modes:detour and pass through the ***σ_(p)and U_(d)firstly increase and then decrease with increasing D,and porosity shows the opposite ***ε_(1,p)andΔε_(v)are negatively correlated with D,and CGBS with D=2.150 has the maximum deformation parameters(ε_(1,p)=0.05079,Δε_(v)=0.01990)due to the frictional slip effect caused by coarse *** increasing D,the failure modes of CGBSs are sequentially manifested as oblique shear failure,"Y-shaped"shear failure,and conjugate shear failure.

In vitro anti-influenza viral activities of stilbenoids

Three stilbenoids with high influenza neuraminidase(NA) inhibition were discovered through high throughput screening. In order to primarily study their structure-activity relationship(SAR),eight stilbenoids were examined on the inhibitory effects of NAs which were from three different subtypes of influenza virus strains,and their in vitro anti-viral activities were also assessed using influenza virus induced cytopathic effect(CPE) reduction method,three of them,namely gnetupendin B(1),isorhapontigenin(2) and broresol A(3) displayed significant in vitro anti-influenza viral activities with IC values of 16.23,16.58 and 14.45μmol·L, *** results indicated that 3-OH and 5-OH are essential functionalities to their activities,and hydrophobic groups at C2 and C3’ positions can increase their *** findings will provide important information for new drug design and for the utilization and exploitation of stilbenoids for influenza treatment and prevention.

Extracorporeal liver surgery(ELS), also known as liver autotransplantation, is a hybrid(cross-fertilized) surgery incorporating the technical knowledge from extreme liver and transplant liver surgeries, and recently became more embraced and popularized among leading centers. ELS could be summarized into three major categories, namely, ex-situ liver resection and autotransplantation(ELRA), ante-situm liver resection and autotransplantation(ALRA) and auxiliary partial liver autotransplantation(APLA). The successful development of ELS during the past 37 years is definitely inseparable from continuous effort s done by Chinese surgeons and researchers. Especially, the precision liver surgery paradigm has allowed to transform ELS into a modularized, more simplified, and standardized surgery, to upgrade surgical skills, to improve peri-operative outcome and long-term survival, to increase the capability of surgeons to select more complex diseases and to expand the level of medical service to the population. This review highlights the Chinese contributions to the field of ELS, focusing thereby on features of different surgical types, technical innovations, disease selection and surgical indication, patient prognosis and future perspectives.

Gut-brain axis as a bridge in obesity and depression:Mechanistic exploration and therapeutic prospects

A recent study by Wang et al,published in the World Journal of Psychiatry,provided preventative and therapeutic strategies for the comorbidity of obesity and *** gut-brain axis,which acts as a two-way communication system between the gastrointestinal tract and the central nervous system,plays a pivotal role in the pathogenesis of these *** suggests that metabolic byproducts,such as short-chain fatty acids,lipopolysaccharide and bile acids,which are generated by the gut microbiota,along with neurotransmitters and inflammatory mediators within the gut-brain axis,modulate the host's metabolic processes,neuronal regulation,and immune responses through diverse *** interaction between obesity and depression via the gut-brain axis involves disruptions in the gut microbiota balance,inflammatory immune responses,and alterations in the neuroendocrine *** the gut-brain axis,for example,through a ketogenic diet,the use of probiotics,and the supplementation of antioxidants,offers new remedial approaches for obesity and *** research that explores the mechanisms of the gut-brain axis is needed to provide more evidence for clinical treatment.
