Objective To explore the mechanism that cytoplasmic factors could recover L-type Ca2+ channel activity after "run-down".The factors include ATP,calpastatin and h fraction(a high molecular fraction of bovine cardiac cytoplasm).Methods Single Ca2+ channel activities were recorded with patch clamp technique in guinea-pig cardiac ***-down was induced by the inside-out patch ***(CS),calmodulin(CaM)and three GST-fusion fragment peptides derived from the C-terminal tail of guinea-pig Cav1.2,CT-1(amino acids number 1509-1791),CT-2(1777-2003)and CT-3(1944-2169)were produced as GST fusion ***(1)CaM + ATP or CS + ATP restored the channels after run-down;however,the CaM or CS's effects became smaller with the longer run-down time.(2)After run down,CaM-dependent protein kinase(CaMKII)produced Ca2+ channel activity to only 2-10% of the basal activity,however,in the presence of CaMKII,the time-dependent nature of the CaM effect was abolished.(3)In pull-down assay,CT-1 treated with CaMKII showed a higher affinity for CaM than that treated with phosphatase.(4)CaMKII was detected in the h fraction of bovine cardiac *** The results show that CS,CaM and CaMKII are all involved in the maintenance of the basal activity of L-type Ca2+ channels,and that there might be cross talks among the four factors(CS,CaM,CaMKII and the undefined cytoplasmic factor).

In the present study,we show that time-consuming manual tuning of parameters in the Rietveld method,one of the most frequently used crystal structure analysis methods in materials science,can be automated by considering the entire trial-and-error process as a blackbox optimisation *** automation is successfully achieved using Bayesian optimisation,which outperforms both a human expert and an expert-system type automation despite the absence of *** approach stabilises the analysis quality by eliminating human-origin variance and bias,and can be applied to various analysis methods in other areas which also suffer from similar tiresome and unsystematic manual tuning of extrinsic parameters and human-origin variance and bias.

In this paper,we propose an in-process measurement method of the diameter of micro-optical fiber such as a tapered optical *** proposed technique is based on analyzing optically scattered light generated by standing wave *** proposed method is significant in that it requires an only limited measurement range and does not require a high dynamic range *** properties are suitable for in-process *** experiment verified that the proposed method could measure a fiber diameter as stable as±0.01μm under an air turbulence *** a result of comparing the measured diameter distribution with those by scanning electron microscopy,it was confirmed that the proposed method has a measurement accuracy better than several hundred nanometers.

The Phalaborwa carbonatite Complex,situated in the northeastern part of South Africa,is characterized by copper and zirconium mineralization,and is composed principally of pyroxenites,phoscorite and carbonatite (banded and transgressive).The complex is transected by mafic dykes,and is geographically associated with a satellite syenite and minor granite *** and baddeleyite U-Pb isotopic age determinations using CAMECA 1280 secondary ion mass spectrometry have shown that the outer pegmatitic pyroxenite at the Loolekop pipe was emplaced at 2060±4 Ma,and the main phoscorite at 2062±2 *** ages are identical to those of 2060±2 and 2060±1 Ma for the banded and transgressive carbonatites,*** satellite syenite,which forms plug-like bodies outside of the border of the main complex,and the later mafic dyke have "similar" emplacement ages of 2068±17 and 2062±53 Ma,indicating that these intrusions were apparently near-synchronously *** contrast to other carbonatites,the Phalaborwa Complex is characterized by high initial Sr and low initial Nd and hf isotopic *** situ isotopic analyses of apatite, calcite,zircon and baddeleyite indicate that the primary magma was derived from an enriched mantle. As the complex was emplaced slightly earlier at～2060 Ma than the nearby mafic phase of the Bushveld Complex (-2055 Ma),it is proposed that the Phalaborwa carbonatite magmatism was triggered by the same mantle plume activity,which partially melted the overlying lithospheric *** contribution also highlights that isotopic studies used to constrain the genesis of ancient igneous complexes should concentrate on minerals with low parent/daughter elemental ratios,such as apatite and calcite for Sr isotopes,and zircon and baddeleyite for hf isotopes.

To examine anti-obesity and-diabetic effects of neuronal histamine especially in leptin resistant states, we investigated the effects of chronic central treatment with histamine on lipid, glucose and energy metabolism of db/db obese mice, which are genetically leptin receptor mutated mice. Chronic centrally treatment with histamine (0.05 μmol*g-1 body weight*d-1 for 7 days) decreased body weight, food intake in db/db obese mice, and decreased body fat weight, serum concentration of glucose compared with pair-fed db/db obese mice. Neuronal histamine also suppressed ob　mRNA in the white adipose tissue (WAT), serum leptin and increased UCPs mRNA expression in brown adipose tissue (BAT) and in WAT compared with pair-fed controls. These above effects of the histamine were attenuated in these mice with combination of targeted disruption of the histamine h1 receptor gene. In conclusion, neuronal histamine can regulate body fat deposition, serum glucose, leptin, BAT and WAT UCPs expression even in leptin insensitive db/db obese mice.

AIM: Although the quality of currently available urinary tests for detecting antibody to helicobacter pylori(h pylori) have been proved in some populations, the accuracy has not been studied regarding patients who suffer from pulmonary tuberculosis with multi-drug treatments. The present study was conducted to evaluate the accuracy of these urinary tests for antibody to h pylori in these patients. METhODS: Serum samples from 61 inpatients with pulmonary tuberculosis were tested using enzyme immunoassay, and urine samples were assayed by enzyme-linked immunosorbent assay method (URINELISA) and immunochromatography method (RAPIRAN). Medicines prescribed to the patients were recorded for medical charts, to evaluate the influences on the results of urinary tests. RESULTS: The sensitivity, specificity, and consistency of URINELJSA against the serum test were 93.1%, 65.6%, and 78.6% respectively, and those of RAPIRAN were 86.2%, 93.7%, and 90.1% respectively, which were almost equal to the data previously reported. Prescribed medicines had little influence on the results. CONCLUSION: The two urinary tests for detecting h pylori antibody have a diagnostic accuracy in patients with pulmonary tuberculosis given multiple anti-tuberculosis drugs.
