To examine anti-obesity and-diabetic effects of neuronal histamine especially in leptin resistant states, we investigated the effects of chronic central treatment with histamine on lipid, glucose and energy metabolism of db/db obese mice, which are genetically leptin receptor mutated mice. Chronic centrally treatment with histamine (0.05 μmol*g-1 body weight*d-1 for 7 days) decreased body weight, food intake in db/db obese mice, and decreased body fat weight, serum concentration of glucose compared with pair-fed db/db obese mice. Neuronal histamine also suppressed ob　mRNA in the white adipose tissue (WAT), serum leptin and increased UCPs mRNA expression in brown adipose tissue (BAT) and in WAT compared with pair-fed controls. These above effects of the histamine were attenuated in these mice with combination of targeted disruption of the histamine h1 receptor gene. In conclusion, neuronal histamine can regulate body fat deposition, serum glucose, leptin, BAT and WAT UCPs expression even in leptin insensitive db/db obese mice.

Background:Voltage-gated L-type Ca channel has been suggested to be involved in many essential processes by the Ca-dependent regulation,including muscle contraction,gene expression and hormone secretion. AIM To evaluate the inhibitory effects of deferasirox(DFX) against hepatocellular carcinoma(hCC) through basic and clinical *** In the basic study, the effect of DFX was investigated in three hepatoma cell lines(hep G2, hep3 B, and huh7), as well as in an N-nitrosodiethylamine-induced murine hCC model. In the clinical study, six advanced hCC patients refractory to chemotherapy were enrolled. The initial dose of DFX was 10 mg/kg per day and was increased by 10 mg/kg per day every week, until the maximum dose of 30 mg/kg per day. The duration of a single course of DFX therapy was 28 consecutive days. In the event of dose-limiting toxicity(according to the Common Terminology Criteria for Adverse Events v.4.0), DFX dose was *** Administration of DFX inhibited the proliferation of hepatoma cell lines and induced the activation of caspase-3 in a dose-dependent manner in vitro. In the murine model, DFX treatment significantly suppressed the development of liver tumors(P he mR NA expression levels of hepcidin(P hypoxia inducible factor-1α(P h tumor and non-tumor tissues, compared with control mice. In the clinical study, anorexia and elevated serum creatinine were observed in four and all six patients, respectively. however, reduction in DFX dose led to decrease in serum creatinine levels in all patients. After the first course of DFX, one patient discontinued the therapy. We assessed the tumor response in the remaining five patients; one patient exhibited stable disease, while four patients exhibited progressive disease. The one-year survival rate of the six patients was 17%.CONCLUSION We demonstrated that DFX inhibited hCC in the basic study, but not in the clinical study due to dose-limiting toxicities.

很多疾病与人们生活方式相关,如过量摄入蔗糖会引起高血糖、糖尿病。给小鼠注射一种乳酸菌可以抑制蔗糖引起的高血糖,但在人类临床试验中没有类似效果。来自东京大学医学研究所的学者研究发现,用家蚕筛选系统鉴定的粪肠球菌YM0831,可以抑制人体内蔗糖引起的高血糖,同时在家蚕中粪肠球菌YM0831能抑制葡萄糖引起的高血糖。粪肠球菌YM0831可以抑制人体内肠上皮细胞Caco-2对葡萄糖的吸收,粪肠球菌YM0831转座子插入突变会导致其抑制活性降低,在家蚕体内蔗糖和葡萄糖引起的高血糖抑制活性也均降低。在人类临床试验中,口服粪肠球菌YM0831抑制了蔗糖引起的血糖升高。以上结果表明,粪肠球菌YM0831可以抑制人体内肠道葡萄糖转运,抑制蔗糖引起的高血糖。

Gravity Survey along the Mizuho Traverse Routes, East Antarctica: SEAL Seismic Exploration in 1999-2000

A gravity survey was conducted along the Mizuho traverse routes in austral summer of 1999-2000 by the 41st Japanese Antarctic Research Expedition (JARE-41). The main purpose of the survey was to obtain detail gravity anomaly along the traverse routes, where deep seismic exploration was simultaneously conducted in order to obtain a fine crustal structure of the Mizuho Plateau. By using SCINTREX (CG-3M) gravity meter, the survey was carried out at 160 sites with about 1 km interval in a distance of 190 km from S16 to Z20 on the traverse routes. Free-air and Bouguer anomalies were calculated using precise locations by GPS measurements, by taking into account an effect of the thick icesheet. The furrowed negative Free-air anomalies are identified around h192, where middle points of the whole traverse routes. Two bedrock elevation models derived both by seismic refraction analyses and radio-echo sounding might correspond to the bottom and the top of a mixture layer composed from ice-moraine rocks over the bedrock surface. Thickness of the crust estimated by Bouguer anomalies is 0.5 - 1 km larger toward inland at the terminal point of Z20 along the profile.

Emergence of Optochin Resistance among <i>Streptococcus pneumoniae</i>in Japan

＜正＞Objectives: We evaluated that feasibility of using Tc-99m tin colloid to identify sentinel nodes （SNs） gastrointestinal malignancies. Methods: The subjects were patients diagnosed with gastrointestinal malignancies considered to be surgically respectable with no clinically obvious nodal metastases. One hundred and sixty-seven patients with gastric cancer, 52 with esophageal cancer, and 61 with colorectal cancer were enrolled in this study. Tc-99m labeled tin colloid （Tc-99m: SnCl2=1:1） was endoscopically injected into the submucosal layer around the tumor. On the following day, patients underwent resection surgery, including resection of regional lymph nodes （LNs）. The radioactivity of resected LNs was measured using a well-type scintillation counter and /or a surgical probe. Any LN with a radioactivity higher than 1/10 of the highest one was defined as a SN. The pathological state of each of the resected LNs was examined using routine h-E staining. Results: The detectability of SNs as well as the sensitivity, false negative （FN） ratio, and diagnostic accuracy of the metastatic status of regional LNs as judged by the pathological results are shown below. Absolute subject numbers （positive cases/total cases examined） are shown in parentheses. Conclusion: Tc-99m tin colloid was able to detect over 90% of the SNs of gastrointestinal malignancies. We suggest that sentinel node concept could be applied to the gastrointestinal tumors as well as breast cancer and malignant melanoma.

The optochin susceptibility test is a key method for differentiating Streptococcus pneumoniae from other α-hemolytic streptococci;however, optochin-resistant (Optr) S. pneumoniae have been reported in the last two decades. In this study, we investigated the isolation frequency of Optr S. pneumoniae in the North Kyushu area of Japan, and biochemically and genetically characterized Optr S. pneumoniae clinical isolates. Seven (0.68%) out of 1032 S. pneumoniae isolates collected by the North Fukuoka Infectious Diseases Working Group were found to be Optr S. pneumoniae. Resistant strains had MICs of optochin 2- to 64- fold higher than susceptible strains, possessed different antimicrobial resistance profiles, and belonged to different serotypes. All the seven Optr isolates had mutations in the nucleotide sequence code for subunit c of F0F1 ATPase. Three isolates had mutations in codon 48 (deduced amino acid substitution of valine with phenylalanine) and two isolates had mutations in codon 49 (substitution of alanine with threonine or serine). Of the remaining two isolates, one had mutation in codon 50 (substitution of phenylalanine with leucine) and the other had mutation in codon 44 (substitution of methionine with isoleucine, which was a novel mutation in this position). From these results, we identified the mutation in the h+-ATPase subunit c gene (atpC) of S. pneumoniae, which was not recognized earlier, and determined that Opt resistance among Japanese pneumococcal isolates is not related to a specific pneumococcal serotype or antimicrobial resistance profile. Furthermore, the results indicate that when α-hemolytic streptococci resistant to optochin are isolated from patients with invasive infectious diseases, such as meningitis and pneumonia, we should perform additional examinations such as bile solubility tests or PCR assays before confirming isolates as viridans streptococci. This is the first report of the characterization of Optr S. pneumoniae in Japan.

Epigenetic alteration to activate Bmp2-Smad signaling in Raf-induced senescence

AIM: To investigate epigenomic and gene expression alterations during cellular senescence induced by oncogenic Raf. METhODS: Cellular senescence was induced into mouse embryonic fibroblasts(MEFs) by infecting retrovirus to express oncogenic Raf(RafV 600E). RNA was collected from RafV 600 E cells as well as MEFs without infection and MEFs with mock infection, and a genome-wide gene expression analysis was performed using microarray. The epigenomic status for active h3K4me3 and repressive h3K27me3 histone marks was analyzed by chromatin immunoprecipitation-sequencing for RafV 600 E cells on day 7 and for MEFs without infection. These data for Raf-induced senescence were compared with data for Ras-induced senescence that were obtained in our previous study. Gene knockdown and overexpression were done by retrovirus infection. RESULTS: Although the expression of some genes including secreted factors was specifically altered in either Ras- or Raf-induced senescence, many genes showed similar alteration pattern in Raf- and Ras-induced senescence. A total of 841 commonly upregulated 841 genes and 573 commonly downregulated genes showed a significant enrichment of genes related to signal and secreted proteins, suggesting the importance of alterations in secreted factors. Bmp2, a secreted protein to activate Bmp2-Smad signaling, was highly upregulated with gain of h3K4me3 and loss of h3K27me3 during Raf-induced senescence, as previously detected in Ras-induced senescence, and the knockdown of Bmp2 by sh RNA lead to escape from Raf-induced senescence. Bmp2-Smad inhibitor Smad6 was strongly repressed with h3K4me3 loss in Raf-induced senescence, as detected in Ras-induced senescence, and senescence was also bypassed by Smad6 induction in Raf-activated cells. Different from Ras-induced senescence, however, gain of h3K27me3 did not occur in the Smad6 promoter region during Raf-induced senescence. Sorafenib is used worldwide as a first-line standardsystemic agent for advanced hepatocellular carcinoma(hCC) on the basis of the results of two large-scale Phase Ⅲ trials. Conversely,hepatic arterial infusion chemotherapy(hAIC) is one of the most recommended treatments in Japan. Although there have been no randomized controlled trials comparing sorafenib with hAIC,several retrospective analyses have shown no significant differences in survival between the two therapies. Outcomes are favorable for hCC patients exhibiting macroscopic vascular invasion when treated with hAIC rather than sorafenib,whereas in hCC patients exhibiting extrahepatic spread or resistance to transcatheter arterial chemoembolization,good outcomes are achieved by treatment with sorafenib rather than hAIC. Additionally,sorafenib is generally used to treat patients with Child-Pugh A,while hAIC is indicated for those with either Child-Pugh A or B. Based on these findings,we reviewed treatment strategies for advanced hCC. We propose that sorafenib might be used as a first-line treatment for advanced hCC patients without macroscopic vascular invasion or Child-Pugh A,while hAIC is recommended for those with macroscopic vascular invasion or Child-Pugh A or B. Additional research is required to determine the best second-line treatment for hAIC non-responders with Child-Pugh B through future clinical trials.
