The human malaria parasite Plasmodium falciparum thrives in radically different host environments in mosquitoes and humans, with only a limited set of transcription factors. The nature of regulatory elements or their target genes in the P. falciparum genome remains elusive. here, we found that this eukaryotic parasite uses an efficient way to maximally use genetic and epigenetic regulation to form regulatory units(RUs) during blood infections. Genes located in the same RU tend to have the same pattern of expression over time and are associated with open chromatin along regulatory elements. To precisely define and quantify these RUs, a novel hidden Markov model was developed to capture the regulatory structure in a genome-wide fashion by integrating expression and epigenetic evidence. We successfully identified thousands of RUs and cross-validated with previous findings. We found more genes involved in red blood cell(RBC) invasion located in the same RU as the Pf AP2-I(AP2-I) transcription factor, demonstrating that AP2-I is responsible for regulating RBC invasion. Our study has provided a regulatory mechanism for a compact eukaryotic genome and offers new insights into the in vivo transcriptional regulation of the P. falciparum intraerythrocytic stage.

利用具有6个无毒基因不同表现型且处于萌发中的静孢子时期的马铃薯晚疫病菌构建4个混合池,通过cDNA AFLP分析,共获得85个与无毒基因相关的差异表达片段,其中与无毒基因Avr1、Avr2、 Avr3 Avr10 Avr11、Avr4相关的差异表达片段分别为20,28,16和21个。将这些差异表达片段对20个晚疫病病菌个体进行无毒基因差异表达验证,得到1个无毒基因连锁群Avr3 Avr10 Avr11的候选片段,即 A11T13Avr3 10 11S157;无毒基因Avr4的候选片段2个,即A24T19Avr4S122和A24T24Avr4S142。Blast分析表明:A11T13Avr3 10 11S157与马铃薯晚疫病菌萌发孢子囊(germinatingsporangium)EST(expressed sequencetag)库中的序列***7同源;A24T19Avr4S122与晚疫病菌萌发中静孢子(germinating cyst)EST库中的***7部分序列完全一致,而A24T24Avr4S142与***7具有98%的同源性。这些结果将促进通过电子克隆并结合RACE(RapidAmplificationofcDNAEnds)技术获得全长的无毒基因Avr4。

Electronic interactions of the Group 2A elements with magnesium have been studied through the dilute solid solutions in binary Mg-Ca,Mg-Sr and Mg-Ba *** investigation incorporated the difference in the‘Work Function'(ΔWF)measured via Kelvin Probe Force Microscopy(KPFM),as a property directly affected by interatomic bond types,*** electronic structure,nanoindentation measurements,and Stacking Fault Energy values reported in the *** was shown that the nano-hardness of the solid-solutionα-Mg phase changed in the order of Mg-Ca>Mg-Sr>***,it was shown,by also considering the nano-hardness levels,that SFE of a solid-solution is closely correlated with its‘Work Function'***-hardness measurements on the eutectics andΔWF difference between eutectic phases enabled an assessment of the relative bond strength and the pertinent electronic structures of the eutectics in the three *** withΔWF and at least qualitative verification of those computed SFE values with some experimental measurement techniques were considered important as those computational methods are based on zero Kelvin degree,relatively simple atomic models and a number of *** asserted by this investigation,if the results of measurement techniques can be qualitatively correlated with those of the computational methods,it can be possible to evaluate the electronic structures in alloys,starting from binary systems,going to ternary and then multi-elemental *** investigation has shown that such a qualitative correlation is *** all,the SFE values are not treated as absolute values but rather become essential in comparative investigations when assessing the influences of alloying elements at a fundamental level,that is,free electron density *** study indicated that the principles of‘electronic metallurgy'in developing multi-elemental alloy systems can be followed via practical experimental methods,i.e.ΔWF measurements using KPF

It is impossible to directly analyze the microstructure of spin-valve multilayers based on Ni,F,Cu and Mn by a conventional X-ray diffraction technique because the lattice parameter and atomic sattering factor sof them are very *** solve this problem, we use an x-ray anomalous diffraction technique to characterize the microstructures of the [Ni80Fe20/Fe50Mn50]15 and [Ni80Fe20/Cu]15 superlattice *** results show that more diffraction peaks and higher internsity in the reflectivety profile are observed when the incident energy is close to the absorption edge of the lighter element(Mn) in [Ni80Fe20/Fe50Mn50]15 multilayer systems and to the absorption edge of the heavier element (Cu) in the [Ni80Fe20/Cu]15 multilayer *** interface and periodic structure of [Ni80F20/Fe50Mn50]15 are more perfect than that of the [Ni80Fe20/Cu]15 *** above results are disussed in this paper.

Response of ocean acidification to atmospheric carbon dioxide removal

Artificial CO_(2)removal from the atmosphere(also referred to as negative CO_(2)emissions)has been proposed as a potential means to counteract anthropogenic climate *** we use an Earth system model to examine the response of ocean acidification to idealized atmospheric CO_(2)removal *** our simulations,atmospheric CO_(2)is assumed to increase at a rate of 1%per year to four times its pre-industrial value and then decreases to the pre-industrial level at a rate of 0.5%,1%,2%per year,*** results show that the annual mean state of surface ocean carbonate chemistry fields including hydrogen ion concentration([h^(+)]),ph and aragonite saturation state respond quickly to removal of atmospheric CO_(2).however,the change of seasonal cycle in carbonate chemistry lags behind the decline in atmospheric CO_(2).When CO_(2)returns to the pre-industrial level,over some parts of the ocean,relative to the pre-industrial state,the seasonal amplitude of carbonate chemistry fields is substantially *** results also show that changes in deep ocean carbonate chemistry substantially lag behind atmospheric CO_(2)*** CO_(2)returns to its pre-industrial value,the whole-ocean acidity measured by[h^(+)]is 15%-18%larger than the pre-industrial level,depending on the rate of CO_(2)*** study demonstrates that even if atmospheric CO_(2)can be lowered in the future as a result of net negative CO_(2)emissions,the recovery of some aspects of ocean acidification would take decades to centuries,which would have important implications for the resilience of marine ecosystems.
