Early gastric cancer frequently has high expression of KKLC-1, a cancer-testis antigen

AIM To assess cancer-testis antigens(CTAs) expression in gastric cancer patients and examined their associations with clinicopathological *** Eighty-three gastric cancer patients were evaluated in this study. Gastric cancer specimens were evaluated for the gene expression of CTAs, Kitakyushu lung cancer antigen-1(KK-LC-1), melanoma antigen(MAGE)-A1, MAGE-A3 and New York esophageal cancer-1(NYESO-1), by reverse transcription PCR. Clinicopathological background information, such as gender, age, tumor size, macroscopic type, tumor histology, depth of invasion, lymph node metastasis, lymphatic invasion, venous invasion, and pathological stage, was obtained. Statistical comparisons between the expression of each CTA and each clinicopathological background were performed using the χ2 test. RESULTS The expression rates of KK-LC-1, MAGE-A1, MAGE-A3, and NY-ESO-1 were 79.5%, 32.5%, 39.8%, and 15.7%, respectively. In early stage gastric cancer specimens, the expression of KK-LC-1 was 79.4%, which is comparable to the 79.6% observed in advanced stage specimens. The expression of KK-LC-1 was not significantly associated with clinicopathological factors, while there were considerable differences in the expression rates of MAGE-A1 and MAGE-A3 with vs without lymphatic invasion(MAGE-A1, 39.3% vs 13.6%, P = 0.034; MAGE-A3, 47.5% vs 18.2%, P = 0.022) and/or vascular invasion(MAGE-A1, 41.5% vs 16.7%, P = 0.028; MAGE-A3, 49.1% vs 23.3%, P = 0.035) and, particularly, MAGE-A3, in patients with early vs advanced stage(36.5% vs 49.0%, P = 0.044), respectively. Patients expressing MAGE-A3 and NYESO-1 were older than those not expressing MAGE-A3 and NY-ESO-1(MAGE-A3, 73.7 ± 7.1 vs 67.4 ± 12.3, P = 0.009; NY-ESO-1, 75.5 ± 7.2 vs 68.8 ± 11.2, P = 0.042). CONCLUSION The KK-LC-1 expression rate was high even in patients with stage I cancer, suggesting that KK-LC-1 is a useful biomarker for early diagnosis of gastric cancer.

We have developed an event mixing technique to observe Bose-Einstein correlations （BEC） between two identical neutral pions produced in photo-induced reactions in the non-perturbative QCD energy region. It is found that the missing-mass consistency cut and the pion-energy cut are essential for the event mixing method to effectively extract BEC observables. A Monte Carlo （MC） simulation is used to validate these constraints and confirms the efficiency of this method. Our work paves the way for similar BEC studies at lower energies where the multiplicity of emitted bosons is limited.

Color and chemical characterization of partially black-streaked heartwood in teak (Tectona grandis)

The heartwood of teak in certain areas of Java Island displays irregular black streaks along the annual rings. We investigated the color and chemical characteristics in a radial direction of partially black-streaked heartwood samples. Color properties （ph value, inorganic element, extractive content and extractive characterization） were measured in the color co-ordinates CIELAB system. The results show that the black streak part was 12-15 brightness （L^＊） value units less than the normal heartwood. Furthermore, the black streak part had more red （a^＊） but less yellow （b^＊）, hue （h） and chroma （C^＊） than the normal wood. The ph value, ash content and calcium contents of the black streak part were slightly higher than those obtained for the normal wood. The content of the low-polar extractive （n-hexane and ethyl acetate） of the black streak part was considerably higher than that for the normal wood. The blackening process was speculated to be a kind of defense mechanism indicated by remarkable amount ofbioactive compound called tectoquinone.

Cilostazol Enhances Oxidative Glucose Metabolism in Both Neurons and Astroglia without Increasing Ros Production

Cilostazol, a potent inhibitor of type 3 phosphodiesterase (PDE3), has recently been reported to exert neuroprotective effects during acute cerebral ischemic injury. These effects are, at least in part, mediated by the inhibition of oxidative cell death. however, the effects of cilostazol on glucose metabolism in brain cells have not been determined. In the present study, we examined the effects of cilostazol on the oxidative metabolism of glucose and the resultant formation of reactive oxygen species (ROS) in cultured neurons and astroglia. Cultures of neurons or astroglia were prepared from Sprague-Dawley rats. The cells were treated with cilostazol (0 – 30 μM) for 48 hours prior to the assay. L-[U-14C]lactate ([14C]lactate) or [1-14C]pyruvate ([14C]pyruvate) oxidation was measured. ROS production was determined using an h2DCFDA assay with a microplate reader. Forty-eight hours of exposure to cilostazol resulted in dose-dependent increases in [14C]lactate and [14C]pyruvate oxidation in both the neurons and astroglia. Dibutyryl cyclic AMP (0 – 0.5 mM) also increased [14C]lactate oxidation, indicating cAMP-mediated PDh activation. In contrast, free radical formation was not affected by cilostazol in either the neurons or astroglia. Cilostazol enhanced the oxidative metabolism of glucose in both neurons and astroglia, while it did not augment ROS production.

Estimation of water requirement per unit carbon fixed by Eucalyptus camaldulensis in semi-arid land of Western Australia

Afforestation in arid land is a promising method for carbon fixation, but the effective utili- zation of water is highly important and required. Thus, the evaluation of the amount of water per unit carbon fixed with the tree growth is required to minimize the amount of water supplied to the plants. In this research, a tree is regarded as a carbon fixation reactor with inflows of water and nutrients from roots, and CO_2 as the carbon source from leaves with outflow of water vapor from leaves and accumulation in the tree itself. In the process of photosynthesis and respiration nutri- tional elements are dissolved in water flow in trees. They do not flow out by these reactions, but are accumulated in trees. Thus, we have treated the behaviour of nutrients as a marker to evaluate the water/carbon ratio. Assuming that nutrient concentration is constant in sap, and the differences in the ratios of nutrient to carbon in living trees and dead (i.e. litter fall, etc.) are neglected, the ratio of the used water to fixed carbon is given as the ratio of nutrient to carbon in the tree body divided by the ratio of nutrient to water in sap. however, some nutrients are translocated and concentrated within the tree and some may be discarded through litter fall. Thus it is important to examine which nutrient element is the most suitable as the tracer. In this paper, the results of the above method applied to Eucalyptus camaldulensis in semi- arid land of Western Australia are shown. The value of water requirement per unit carbon fixation determined from potassium balance is between 421 kg-h_2O/kg-C for mature trees and 285 kg-h_2O/kg-C for young trees, while the values from calcium balance are much larger than these. The cause of the discrepancy between these values is discussed based on the measured element concentrations in sap and trees and the plant physiology. Finally, the actual average value through the life of a tree is suggested to fall between the two values.

Regeneration and Differentiation of hamster Tracheal Epithelium Used Anti--Brdu Monoclonal Antibody After Mechanical Injury

The ventral quadrant of the tracheal epithelium of the hamster,about one fourth of tracheal mucosa, was denuded by a venous needle *** stains were made with hE, PAS, and PAS-anti Brdu immunohis-tochemic technique. At the 0 h circumference cell number (CCN) was 927. *** 6 h post injured the viable cells changed in shapes and migrated from themargin to wound site. By 24 h the wound area was completely covered by a sin-gle layer of non-ciliated flattened cells, then an expotential increassing in cellregeneration occured in wound site by 48 h. The CCN had been restored to thelevel of control (1373), and the proliferative cell, composed of polygonal epi-dermoid metaplasia, 3～4 layers and reached a peak (338.8). The secretoryand ciliated cells appeared gradually from 72 h post injury, the epithelium re-stored to the normal epithelial architecture by one week post injury. In our pre-sent study either in control and non-injured epithelium or in all stages of woundsite about 70% of the Brdu positive cells contained small or confluent PAS posi-tive granules were observed. This fact indicated that secretory cells play a im-portant role in proliferation after mechanical injury and maintaining the normaltracheal pseudostratified epithelium.

Caspase-11 mediated inflammasome activation in macrophages by systemic infection of *** exacerbates arthritis

Clinical studies have shown that Aggregatibacter actinomycetemcomitans(***)is associated with aggressive periodontitis and can potentially trigger or exacerbate rheumatoid arthritis(RA).however,the mechanism is poorly ***,we show that systemic infection with *** triggers the progression of arthritis in mice anti-collagen antibody-induced arthritis(CAIA)model following IL-1βsecretion and cell infiltration in paws in a manner that is dependent on caspase-11-mediated inflammasome activation in *** administration of polymyxin B(PMB),chloroquine,and anti-CD11b antibody suppressed inflammasome activation in macrophages and arthritis in mice,suggesting that the recognition of lipopolysaccharide(LPS)in the cytosol after bacterial degradation by lysosomes and invasion via CD11b are needed to trigger arthritis following inflammasome activation in *** data reveal that the inhibition of caspase-11-mediated inflammasome activation potentiates aggravation of RA induced by infection with *** work highlights how RA can be progressed by inflammasome activation as a result of periodontitis-associated bacterial infection and discusses the mechanism of inflammasome activation in response to infection with ***.
