Myocardial isch.mia–reperfusion injury(MIRI)is a major h.ndrance to th. success of cardiac reperfusion *** increased neutroph.l infiltration is a h.llmark of MIRI,th. subtypes and alterations of neutroph.ls in th.s process remain ***,we performed single-cell sequencing of cardiac CD45^(+)cells isolated from th. murine myocardium subjected to MIRI at six-time *** identified diverse types of infiltrating immune cells and th.ir dynamic ch.nges during *** neutroph.ls sh.wed th. most immediate response and largest ch.nges and featured with.functionally h.terogeneous subpopulations,including Ccl3^(h.)Neu and Ym-1^(h.)Neu,wh.ch.were increased at 6 h.and 1 d after reperfusion,***-1^(h.)Neu selectively expressed genes with.protective effects and was,th.refore,identified as a novel specific type of cardiac cell in th. injured *** analysis indicated th.t neutroph.ls and th.ir subtypes orch.strated subsequent immune responses in th. cardiac tissues,especially instructing th. response of *** abundance of Ym-1^(h.)Neu was closely correlated with.th. th.rapeutic efficacy of MIRI wh.n neutroph.ls were specifically targeted by anti-Lymph.cyte antigen 6 complex locus G6D(Ly6G)or anti-Intercellular cell adh.sion molecule-1(ICAM-1)neutralizing *** addition,a neutroph.l subtype with.th. same ph.notype as Ym-1^(h.)Neu was detected in clinical samples and correlated with.***-1 inh.bition exacerbated myocardial injury,wh.reas Ym-1 supplementation significantly ameliorated injury in MIRI mice,wh.ch.was attributed to th. tilt of Ym-1 on th. polarization of macroph.ges toward th. repair ph.notype in myocardial ***,our findings reveal th. antiinflammatory ph.notype of Ym-1^(h.)Neu and h.gh.igh. its critical role in myocardial protection during th. early stages of MIRI.

为阐明人清道夫受体AI (h.R -AI)的功能及在动脉粥样硬化发生中的作用 ,本研究首先构建了含鼠tie- 1启动子和人清道夫受体AIcDNA的表达载体 ,经酶切及测序鉴定后 ,用显微注射的方法将制备好的tie - 1-h.R-A -BGh.olyA片段注入受精卵 ,将注射后存活的受精卵移入ICR假孕母鼠 ,产下的仔鼠经聚合酶链反应和South.rnblot分析 ,筛选出整合有外源目的基因的阳性转基因鼠 ;对小鼠组织RNA行逆转录聚合酶链反应及组织切片免疫组织化学染色 ,检测人清道夫受体AI在小鼠体内的表达水平及表达部位 ;光镜及电镜观察转基因鼠血管及其它组织的病理变化。结果发现 ,重组Tie - 1/h.R -AI质粒中鼠tie - 1启动子和人清道夫受体AIcDNA序列正确 ,显微注射后存活的 5 6 1枚受精卵分别移入 19只ICR假孕母鼠 ,有 13只受孕 ,共产下 5 6只仔鼠 ,存活 5 4只 ,经过整合检测 ,检出 7只阳性鼠 ,整合效率为 13%。在G1、G2、G3代及纯合子转基因鼠中PCR阳性率分别为 47.8%、71.3%、75 .0 %和 10 0 %。 5只雄性转基因鼠的主动脉、肾、肝等组织中均有人清道夫受体AI表达 ,且主要集中在血管的内皮细胞上 ;转基因鼠主动脉内皮细胞明显水肿 ,表面呈多囊状和虫蚀样改变 ,胞质中有较多水泡 ,中膜有糖原沉积样灶性病变 ,平滑肌细胞中亦有水肿 ;血浆?

目的探讨JAK/STAT3抑制剂AG490对创伤性休克致兔肾损伤的影响。方法将30只健康新西兰兔分为对照组、模型组与AG490治疗组,每组10只。模型组与AG490治疗组兔建立创伤性休克致肾损伤动物模型。通过实时荧光定量聚合酶链反应(Real-time PCR)和蛋白质印迹法(Western-blot)检测肾组织中STAT3、kim-1、TGF-β1的表达情况。结果与对照组比较,模型组肾组织肌酐与尿素氮水平显著高于对照组,差异有统计学意义(P<0.05);与模型组比较,AG490治疗组肌酐与尿素氮水平显著降低,差异有统计学意义(P<0.05);与对照组比较,模型组脏组织中STAT3、TGF-β1的mRNA与蛋白表达水平显著升高,差异有统计学意义(P<0.05);与模型组比较,AG490治疗组STAT3、TGF-β1的mRNA与蛋白表达水平显著降低,差异有统计学意义(P<0.05)。结论 JAK/STAT3抑制剂可能通过TGF-β1来阻断JAK/STAT3信号通路的激活,进而抑制创伤性休克肾损伤发展。

Th. Circular Electron Positron Collider(CEPC)is a large scientific project initiated and h.sted by Ch.na,fostered th.ough.extensive collaboration with.international *** complex comprises four accelerators:a 30 GeV Linac,a 1.1 GeV Damping Ring,a Booster capable of ach.eving energies up to 180 GeV,and a Collider operating at varying energy modes(Z,W,h.and tt).Th. SR power to 50 MW per beam expands th. CEPC's capability to generate 4.3 million h.ggs bosons,facilitating precise measurements of h.ggs coupling at sub-percent levels,exceeding th. precision expected from th. h.-Lh. by an order of *** Tech.ical Design Report(TDR)follows th. Preliminary Conceptual Design Report(Pre-CDR,2015)and th.
