Involvement of serum retinoids and Leiden mutation in patients with esophageal, gastric, liver, pancreatic, and colorectal cancers in Hungary

AIM: to analyze the serum levels of retinoids and Leiden mutation in patients with esophageal, gastric, liver,pancreatic, and colorectal ***: the changes in serum levels of retinoids (vitamin A, α- and β-carotene, α- and β-cryptoxanthin,zeaxanthin, lutein) and Leiden mutation were measured by high liquid performance chromatography (HPLC)and polymerase chain reaction (PCR) in 107 patients (70 males/37 females) with esophageal (0/8), gastric (16/5), liver (8/7), pancreatic (6/4), and colorectal (30/21including 9 patients suffering from in situ colon cancer)cancer. Fifty-seven healthy subjects (in matched groups)for controls of serum retinoids and 600 healthy blood donors for Leiden mutation were ***: the serum levels of vitamin A and zeaxanthin were decreased significantly in all groups of patients with gastrointestinal (GI) tumors except for vitamin A in patients with pancreatic cancer. No changes were obtained in the serum levels of α- and β-carotene,α- and β-cryptoxanthin, zeaxanthin, lutein in patients with GI cancer. the prevalence of Leiden mutation significantly increased in all groups of patients with GI ***: Retinoids (as environmental factors)are decreased significantly with increased prevalence of Leiden mutation (as a genetic factor) in patients before the clinical manifestation of histologically different (planocellular and hepatocellular carcinoma, and adenocarcinoma) GI cancer.

Motivational and cognitive aspects of spontaneous tool-use acquisition in species that do not do so habitually, remain an open but most relevant question. to address this, we studied captive kea (Nestor notabilis), New Zealand mountain parrots renowned for their playful cleverness. the majority of adolescent, but not the adult kea, showed a toddler-like motivation to insert objects into empty tubes and also spontaneously used objects in order to eject food from inside a tube. this parallel what is known from object exploration in large brained mammals and shows for the first time in a habitually non-tool using bird that such a technical innovation is based on object-combining acquired outside the foraging context.

Introduction: Hyperthermia is a complementary therapy in oncology having various pros and contras for its application. Ascites, pleural effusion, edema and other electrolyte accumulations are frequently excluded from the treatability of the patients with heating locally or systemically. the special gathering of electrolytes is sometimes contraindicated, at times not mentioned in the clinical protocols. However, it is certainly challenging in the oncology where micro and macro edemas, as well as larger electrolyte accumulations (e.g. ascites, pleural effusion), are very frequent. Methods: Excluding patients with accumulation of free electrolytes limits the applications of hyperthermia. to find a solution we are studying the microvasculature and fluid dynamism together with the electric field effects, including the injury currents. the hyperthermia method which we investigate is the modulated electro-hyperthermia (mEHt). We use the Starling’s equation and the injury current in the frame of non-equilibrium thermodynamics and in connection with the biologically closed electric circuits. Results: It is shown that mEHt, unlike the conventional hyperthermia, is applicable for patients who have edema and other free-electrolytes in the volume which is targeted. the heterogeneous heating (unlike the homogeneous, isothermal conventional hyperthermia) promotes the development of tumor-specific immune actions, and so has less adverse-effects, and longer survival time for patients in advanced, metastatic cancers too. Conclusion: mEHt is well applicable in cases of ascites, pleural effusion, edema and other electrolyte accumulations when a patient is treated in complex (complementary) oncological therapy.

Response of 3 Arabidopsis clones to 41 strains of eight Rhizoctonia species was studied in model experiments. the seed germination was decelerated in most of the cases, although the inhibitory effect varied within large limits. the pre-emergence damping off and root neck rot leading to damping off were the most frequent symptoms of disease syndrome caused by toxic metabolites. the clone transformed with cDNA clone overexpressing gstf4 gene exhibited significantly improved tolerance as compared to parental one, meanwhile the sensitivity of Dmannose pyrophosphorylase/mannose-1-pyrophosphatase deficient clone dramatically increased. Strains of R. solani of AG-2, AG-4 and AG-7 and Athelia rolfsii produced the most toxic metabolites, however, no strict relationships were revealed between taxonomic position of Rhizoctonia strains and toxicity of their metabolites.

Many clinical trials have prospective or retrospective data-sets without comparison to the control-group formed by the same cohort as the active one. the measured single arm naturally contains the relevant information, however, in most of the cases, it is impossible to obtain it from the complex survival curve without a reference. In our previous articles [1] [2], we had shown that the self-similar Weibull distribution fits the self-organized biological mechanisms well, and so it is the best option to study the single-arm survival curves, where self-organizing process is actively present. With the Weibull decomposition of the survival curve, we can fit at least two subgroups of patients. the weighted sum of the decomposed fractions could be optimized analytically and determining the best parameters of the components and the best composition ratio of the weighted sum is also possible. In this part of our series of articles, we will show how the method works in a real clinical environment through modulated electro-hyperthermia (mEHt) as a complementary method, applied curatively when no other conventional curative therapies are available. the decomposed function of the non-responding group provides an excellent agreement with the historical controls in pancreatic cancer and non-small-cell-lung-cancer studies. In the case of glioblastoma multiform, the historical missing control from the institute where the treatment was made does not allow a comparison. We used a modified Hardin-Jones-Pauling statistical estimation and had shown in single arm clinical trials for advanced pancreas, non-small cell lung cancer and glioblastoma multiforme, that this estimation is applicable, and it is corresponding with the historical arm and with the non-responding group where this comparison was available.

AIM: to determine common NOD2/CARD15 mutations and tLR4 D299G polymorphism in Hungarian patients with CD. MEtHODS: A total of 527 unrelated patients with CD (male/female: 265/262, age: 37.1 (SD 7.6) years) and 200 healthy subjects were included. DNA was screened for possible NOD2/CARD15 mutations by denaturing high-performance liquid chromatography (confirmed by direct sequencing). tLR4 D299G was tested by PCR-RFLP. RESULtS: NOD2/CARD15 mutations were found in 185 patients (35.1%) and in 33 controls (16.5%,Ptations were more frequent in CD, while the frequency of SNP12/G908R was not increased. the frequency of tLR4 D299G was not different (CD: 9.9% vs controls: 12.0%). Variant NOD2/CARD15 allele was associated with an increased risk for CD (ORhet=1.71, 95%CI=1.12-2.6, P= 0.0001, ORtwo-risk alleles = 25.2, 95%CI =4.37- ,Pt (carrier: 26.4 years vs non-carrier: 29.8 years, P=0.0006), ileal disease (81.9% (?) 69.5%, OR = 1.99, 95%CI = 1.29-3.08, P= 0.02, presence of NOD2/CARD15 and tLR4: 86.7% vs 64.8%), stricturing behavior (OR = 1.69,95%CI = 1.13-2.55, P= 0.026) and increased need for resection (OR=1.71, 95%CI: 1.13-2.62, P= 0.01), but not with duration, extra-intestinal manifestations, familial disease or smoking. tLR4 exhibited a modifier effect: age of onset in wt/tLR4 D299G carriers: 27.4 years vs NOD2mut/tLR D299G: 23 years (P = 0.06), in NOD2mut/wt: 26.7 years. CONCLUSION: these results confirm that variant NOD2/ CARD15 (R702W, R703C and 3020insC) alleles are associated with earlier disease onset, ileal disease, stricturing disease behavior in Hungarian CD patients. In contrast, although the frequency of tLR4 D299G polymorphism was not different from controls, NOD2/tLR4 mutation carriers tended to present at earlier age.

the penetration depth in the electromagnetic heating could be a crucial factor of its application, when the deep heating is the goal. the capacitive coupling is one of the most popular heating techniques in radiofrequency (RF) heating applications. the matching of the target defines the penetration possibilities. the current matched solution has deeper mathematically defined penetration in RF region than the capacitive plane-wave solution. the same power of application request high voltage with relatively low current for plane-wave inducing, while in current matching it has low voltage with high current. the effective depth of the action in the two solutions do not identical, the penetration defined by the intensity of 1/e portion of the incident beam is higher in the current-matched techniques.

the role of electrical forces in angiogenesis is widely studied. the electric field (EF) induces polarization of the endothelial cells, and in this way, it is a morphogen in angiogenesis. Additional to the polarization, it may build up by newborn cells in the process of cellular fission. Due to the weak direct experimental results on the transitions of endothelial cells, we used an analogy to these epithelial transitions. this involved using the injury current, which induces oriented cell migration and morphological arrangement in wound healing. the injury-current considerations are applied for malignant proliferation as well.
