The association of depression and perceived stress with beta cell function between African and Haitian Americans with and without type 2 diabetes

Background: Diabetes and diabetes-related complications are major causes of morbidity and mortality in the United States. Depressive symptoms and perceived stress have been identified as possible risk factors for beta cell dysfunction and diabetes. The purpose of this study was to assess associations between depression symptoms and perceived stress with beta cell function between African and Haitian Americans with and without type 2 diabetes. Participants and Methods: Informed consent and data were available for 462 participants (231 African Americans and 231 Haitian Americans) for this cross-sectional study. A demographic questionnaire developed by the Primary Investigator was used to collect information regarding age, gender, smoking, and ethnicity. Diabetes status was determined by self-report and confirmed by fasting blood glucose. Anthropometrics (weight, and height and waist circumference) and vital signs (blood pressure) were taken. Blood samples were drawn after 8 10 hours over-night fasting to measure lipid panel, fasting plasma glucose and serum insulin concentrations. The homeostatic model assessment, version 2 (HOMA2) computer model was used to calculate beta cell function. Depression was assessed using the Beck Depression Inventory-II (BDI-II) and stress levels were assessed using the Perceived Stress Scale (PSS). Results: Moderate to severe depressive symptoms were more likely for persons with diabetes (p = 0.030). There were no differences in perceived stress between ethnicity and diabetes status (p = 0.283). General linear models for participants with and without type 2 diabetes using beta cell function as the dependent variable showed no association with depressive symptoms and perceived stress;however, Haitian Americans had significantly lower beta cell function than African Americans both with and without diabetes and adjusting for age, gender, waist circumference and smoking. Identification and characterization of the Lipid Transport System in the Tarantula Grammostola rosea

Lipids due to its hydrophobic nature are transported in the hemolymph mainly by lipoproteic fractions. In the present work we studied the lipoproteic fractions present in the hemolymph of the spider Grammostola rosea. Through ultracentrifugation two lipoproteic fractions are isolated, one of high density named Gr-HDL (δ = 1.13 - 1.15 g/ml) and the other of very high density called Gr-VHDL (δ = 1.24 - 1.27 g/ml) Gr-VHDL in hemolymph is majority in relation to Gr-HDL. In this sense Gr-VHDL fraction has 98.6% of hemolymphatic proteins, and 89.3% of lipids presents in the hemolymph. Both lipoproteic fractions possess phospholipids such as majority lipids (phosphatidylcholine and phosphatidylethanolamine) and 18:1, 16:0, 18:2 and 18:0 as the major fatty acids. In order to confirm the role played by lipoproteic fractions in vitro assays with different 14c-lipid were performed. It was observed that Gr-VHDL takes up mainly free fatty acids and triacylglycerols unlike that observed for Gr-HDL in relation to phosphatidylcholine. Through electrophoresis it was observed that Gr-VHDL has three proteins: a predominant band of 68 kDa and two others of 99 and 121 kDa. Gr-HDL displayed a predominant band of 93 kDa, and other minority of 249 kDa. In conclusion, this study reports lipid characterization of the lipoproteic fractions present in the hemolymph of the tarantula, G. rosea. The role of each lipoproteic fraction in relation to lipid uptake is sustained by in vitro assays. Similarities and differences are found when it is compared to lipoproteins of only the three species of spiders studied.

Objective: Although Angelica archangelica is a medicinal and aromatic plant with a long history of use for both medicinal and food purposes, there are no studies regarding the antineoplastic activity of its *** study aimed to evaluate the cytotoxicity and antitumor effects of the crude extract of A. archangelica root(cEAA) on breast ***: The cytotoxicity of cEAA against breast adenocarcinoma cells(4T1 and McF-7) was evaluated by a 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide(MTT) assay. Morphological and biochemical changes were detected by Hoechst 33342/propidium iodide(PI) and annexin V/PI staining. cytosolic calcium mobilization was evaluated in cells staining with FURA-4 NW. Immunoblotting was used to determine the effect of cEAA on anti-and pro-apoptotic proteins(Bcl-2 and Bax, respectively).The 4 T1 cell-challenged mice were used for in vivo ***: Using ultra-high-performance liquid chromatography–mass spectrometry analysis, angelicin, a constituent of the roots and leaves of A. archangelica, was found to be the major constituent of the cEAA evaluated in this study(73 mg/mL). The cEAA was cytotoxic for both breast cancer cell lines studied but not for human fibroblasts. Treatment of 4 T1 cells with the cEAA increased Bax protein levels accompanied by decreased Bcl-2 expression, in the presence of cleaved caspase-3 and cytosolic calcium mobilization, suggesting mitochondrial involvement in breast cancer cell death induced by the cEAA in this cell line. No changes on the Bcl-2/Bax ratio were observed in cEAA-treated McF7 cells. Gavage administration of the cEAA(500 mg/kg) to 4T1 cell-challenged mice significantly decreased tumor growth when compared with untreated ***: Altogether, our data show the antitumor potential of the cEAA against breast cancer cells in vitro and in vivo. Further research is necessary to better elucidate the pharmacological application of the cEAA in breast cancer therapy.

Aquacrop Model calibration in Potato and Its Use to Estimate Yield Variability under Field conditions

Aquacrop model estimates the crop productivity decrease in response to water stress, determining the biomass (B) based on water productivity (WP) and accumulated transpiration (ΣTr);and the yield (Y) is calculated according to B and the harvest index (HI). Aquacrop was evaluated considering different WP values for 2010 late growing season to simulate crop yield of potato (Solanum tuberosum L.) cv. Spunta, in a commercial production field of 9 ha located in the green belt of cordoba city (31°30'S, 64°08'W, 402 m asl), while monitoring in 2009 was used to verify the model. canopy cover estimation by Aquacrop was adjusted using observed field data obtained from vertical digital photographs acquired at 2.5 m height. WP values of 15.8 and 31.6 (for c3 and c4 species, respectively) and two intermediate values 21 and 26.3 g·mˉ2 were considered to evaluate the model performance. While linear function between observed tuber yields and estimated by Aquacrop had always a correlation coefficient greater than 0.94 (p 0.001), using WP = 26.3 and WP =31.6 g·mˉ2 presented overestimation, whereas with 15.8 g·mˉ2 had an opposite behavior, while WP = 21 g·mˉ2 was the value that produced the lowest estimation error. In addition, soil moisture from this estimated value of WP was highly correlated with measured water content in different areas of production field. The verification test shows that while the model slightly underestimates canopy cover, biomass was overestimated. After setting the coefficients of canopy cover development, the Aquacrop crop model estimated adequately potato yield for high production values that are less affected by lack of water, but in both years showed a tendency to overestimate the lowest yields, as was observed for other crops. Meanwhile, the dispersion between the observed and estimated yield was higher in the verification test because the sampling this year was more random.

Analysis of Daily Rainfall in São carlos/SP, Brazil over 1979-2017 Using Laplace Trend Test

Rainfall expresses one of the most complex climate factors in Southeastern Brazil. Understanding the dynamics and temporal trends of rainfall represents a significant challenge due to regional and even global mechanisms, such as FS (Frontal Systems) and the SAcZ (South Atlantic convergence Zone), and the interaction with the Atlantic and Pacific Oceans. The present study aimed at analyzing the pluviometric tendencies in São carlos/SP, in the countryside of São Paulo State. Laplace trend test was used to comprehend the temporal evolution of daily rainfall in the region in the historical series 1979-2017, in seven pluviometric stations (climatological or surface stations). Significant fluctuations in interannual trends and between seasons were observed. However, it was noted that the beginning of the 1980s showed positive trends, whereas, as of the year 2000, most of the stations demonstrated negative trends, indicating a reduction in daily rainfall volume due to the great tropical climatic variability of Brazil. Emphasis should also be given to the regional and local effects, such as elevation and urbanization, respectively, which corroborate such differences among the analyzed stations. This methodology is of considerable value for the observation of pluviometric trends, and future studies can validate such a tool in climatological studies.

Several studies have been performed adjusting the levels of dietary digestible protein and digestible energy for tilapia,but none of them was performed in biofloc technology(BFT)system,especially in brackish(10)*** this sense,three levels of digestible protein(22,26 and 30%DP)and three levels of digestible energy(3000,3150 and 3300 kcal DE/kg)were evaluated on growth performance,body composition,hematological parameters and planktonic community in juveniles of Nile tilapia(***)raised under BFT conditions.A total of 540 tilapia fingerlings(GIFT strain with 1.25±0.15 g initial weight)were reared during 42 days in thirty-six plastic tanks(75 L),fed three times a *** interaction was detected in the parameters *** fish fed with feed containing 26%and 30%of DP obtained higher final weight,specific growth rate and condition factor as compared to those fed with 22%of DP(Pcarcass increased proportionally according to dietary *** higher dietary energy level promoted a higher body lipid *** to hematological parameters fish from all treatments presented a satisfactory immunological *** monitoring of planktonic community indicated a variation over time,specially on the number of dinoflagellates,rotifers and *** results indicated that diets containing 26%of DP and 3150 kcal/kg of DE for Nile tilapia juveniles promoted suitable growth performance and health conditions when reared in biofloc brackish water,and enabled savings in regards to dietary protein once the energy levels were adjusted.

characteristics of Prostate cancers Missed by Biopsies: Evaluation of cumulative Tumor Volume Missed According to cancer True Prevalence

Purpose: To characterize missed prostate tumors and their cumulative volume with various biopsy regimens to determine optimal biopsy schemes. Methods: We performed 6, 12 and 18-core needle biopsies on 165 and 36-core biopsies on 47 autopsy prostates, respectively. The 6-core biopsy included 6 cores from the mid peripheral zone (MPZ), the 12-core biopsy included 6 cores from the MPZ and lateral PZ (LPZ), and the 18-core biopsies included 6 cores from the MPZ, LPZ and central zone (cZ). The 36-core biopsies included 12 cores in each of these 3 areas. We analyzed the sensitivity of biopsies at each site and evaluated the cumulative volume of cancers and tumor foci missed. Results: Whole-mount analysis identified 59 cancers, 110 tumor foci, and a total cumulative tumor volume of 43 cm3. The percentage of tumor foci and corresponding cumulative volume missed with 6, 12, 18 and 36-core biopsies were of 79% and 58%, 64% and 48%, 57% and 26%, and 42% and 17%, respectively (p 0.05). 12-core biopsies from the MPZ and LPZ performed best for clinically significant cancers detection. However, increasing the number of cores over the 6-core biopsy cutoff increased solely the detection of tumor foci 0.5 cm3. conclusion: Twelve biopsies from the MPZ and LPZ detected most of the clinically significant cancers while missing most of the tumor foci. These missed tumors represented only a small amount of the overall cancer volume.
