Coupled dynamic response analysis of multi-column floating offshore wind turbine with low center of gravity

To realize the application of the floating offshore wind turbine(FOWT)from deep to relatively shallow waters,a new concept of multi-column floating wind turbine platform with low center of gravity(CG)is designed and validated.The multi-column low CG platform is designed to support a 6MW wind turbine class and operated at a water depth of 50m in the South China Sea.The frequency domain software WADAM and time domain software NREL-FAST are used to simulate coupled dynamic responses of the floating wind turbine system with second-order wave loads considering.The dynamic behaviors of multi-column low CG FOWT system under normal operation and parked conditions are presented.The influence of second-order wave force on the motion responses of the multi-column platform,fore-aft force and moment of the tower base and mooring force are researched respectively.The results demonstrate that the coupled dynamic responses at rated operating condition and extreme condition meet the normal operating requirements and extreme survival requirements of FOWT system in the shallow water(50m)of South China Sea.In addition,it is found that,the wave frequency response gradually replaces the second-order low frequency response as the main influencing factor of the coupled dynamic response of the FOWT system with the increasing severity of the sea states.However,in general,the magnitude of second-order low frequency response increases with the increasing severity of the design load case.Thus,in the subsequent design of the shallow water FOWT system,the second-order effects should be paid enough attention.

Relationship between family cohesion/adaptability and postpartum depressive symptoms:A single-center retrospective study

BACKGROUND Depression is the most common mental illness in postpartum mothers,and the etiology of postpartum depression remains poorly understood.Over the past several decades,studies have reported that postpartum depression is caused by multiple factors,such as genetic,psychological,pregnancy,and environmental factors,with the family environment being an important environmental factor.The theory of family cohesion and adaptability put forward by Olson is a classic model that describes the level of family function.However,to date,this model has not been examined regarding its applicability to patients with postpartum depression.AIM To investigate the relationship between family cohesion and adaptability and the risk of postpartum depressive symptoms.METHODS We retrospectively analyzed 1446 patients admitted to the postpartum healthcare clinic of the Affiliated Foshan Maternity and Child Healthcare Hospital from April 2021 to December 2021.Patients were grouped according to whether postpartum depression symptoms were reported(symptoms,n=454;no symptoms,n=992).All patients completed the Edinburgh Postpartum Depression Scale and the Chinese version of the Family Cohesion and Adaptability Assessment Scale II.Baseline and clinical data were compared between groups.Univariate regression analysis was used to investigate the association between different types of family cohesion and postpartum depressive symptoms and the association between different family adaptability types and postpartum depressive symptoms.RESULTS After adjusting for age,education,occupation,gravidity,parity,and mode of delivery,disengaged[adjusted odds ratio(AOR)=3.36,95%CI:1.91-5.91],and separated(AOR=1.97,95%CI:1.34-2.90)family cohesion types showed a higher risk of postpartum depression than the connection type,whereas the enmeshed type(AOR=0.38,95%CI:0.28-0.51)protected against postpartum depressive symptoms.Rigid(AOR=4.41,95%CI:3.02-6.43)and structured families(AOR=1.88,95%CI:1.34-2.63)had a higher risk of postpartum depressive symptoms than fl

Regulating the properties of XQ-2d for targeted delivery of therapeutic agents to pancreatic cancers

Enhanced recognition ability, cell uptake capacity, and biostability are characteristics attributed to aptamer-based targeted anticancer agents, and are possibly associated with increased accumulation at the tumor site, improved therapeutic efficacy and reduced negative side effects. Herein, a phosphorothioate backbone modification strategy was applied to regulate the biomedical properties of pancreatic cancer cell–targeting aptamer for efficient in vivo drug delivery. Specifically, the CD71-targeting aptamer XQ-2d was modified into a fully thio-substituted aptamer S-XQ-2d, improving the plasma stability of S-XQ-2d and mitomycin C(MMC)-functionalized S-XQ-2d(MFSX), thus considerably prolonging their half-life in mice. Moreover, the binding and uptake capacities of S-XQ-2d were significantly enhanced. MFSX showed the same level of cytotoxicity as that of MMC against targeted cancer cells, but lower toxicity to non-targeted cells, highlighting its specificity and biosafety. Brief mechanistic studies demonstrated that XQ-2d and S-XQ-2d had different interaction modes and internalization pathways with the targeted cells.

BACKGROUND Guillain-Barrésyndrome(GBS)is an autoimmune-mediated peripheral neuropathy characterized by symmetric weakness.Asymmetric weakness in GBS is uncommon and may be easily confused with other differential diagnoses.We herein present three cases of asymmetric GBS and review the literature on this atypical subtype of GBS in order to describe the characteristics of asymmetric GBS and to provide experience for clinicians.CASE SUMMARY Different from patients in the previous reports,our patients showed persistent asymmetric limb weakness from the onset to recovery phase.All three patients were serologically positive for antecedent infections.Two of the three cases had IgG antibodies against ganglioside GM1.Two patients received immunotherapy including intravenous immunoglobulin and plasma exchange,while one patient received only supportive treatment.Autoantibodies against gangliosides,asymmetry of congenital development of blood-nerve barrier and limb use may contribute to the development of asymmetric limb weakness in GBS.CONCLUSION Asymmetric GBS may be a rare clinical variant and should be considered when a patient develops acute and progressive asymmetric limb weakness.The differences in clinical features and prognosis between asymmetric GBS and classic GBS deserve further investigation in a large study.

Molecular engineering of aptamers can confer exogenous biomedical properties that may be beneficial for various applications.In this study,a tumor-homing peptide modification strategy was developed to considerably enhance the accumulation and penetration abilities of the Sgc8c aptamer.Notably,the S2PM conjugate induced a much higher level of morphological variation in three-dimensional tumor microspheres(HCT116 cells)than in control groups,highlighting the importance of the homing and penetrating abilities derived from peptide.

Direct hepatic differentiation of mouse embryonic stem cells induced by valproic acid and cytokines

AIM: To develop a protocol for direct hepatic lineage differentiation from early developmental progenitors to a population of mature hepatocytes.METHODS: Hepatic progenitor cells and then mature hepatocytes from mouse embryonic stem (ES) cells were obtained in a sequential manner, induced by valproic acid (VPA) and cytokines (hepatocyte growth factor, epidermal growth factor and insulin). Morphological changes of the differentiated cells were examined by phase-contrast microscopy and electron microscopy. Reverse transcription polymerase chain reaction and immunocytochemical analyses were used to evaluate the gene expression prof iles of the VPA-induced hepatic progenitors and the hepatic progenitor-derived hepatocytes. Glycogen storage, cytochrome P450 activity, transplantation assay, differentiation of bile duct-like structures and tumorigenic analyses were performed for the functional identification of the differentiated cells. Furthermore, FACS and electron microscopy were usedfor the analyses of cell cycle profile and apoptosis in VPA-induced hepatic differentiated cells.RESULTS: Based on the combination of VPA and cytokines, mouse ES cells differentiated into a uniform and homogeneous cell population of hepatic progenitor cells and then matured into functional hepatocytes. The progenitor population shared several characteristics with ES cells and hepatic stem/progenitor cells, and represented a novel progenitor cell between ES and hepatic oval cells in embryonic development. The differentiated hepatocytes from progenitor cells shared typical characteristics with mature hepatocytes, including the patterns of gene expression, immunological markers, in vitro hepatocyte functions and in vivo capacity to restore acute-damaged liver function. In addition, the differentiation of hepatic progenitor cells from ES cells was accompanied by signif icant cell cycle arrest and selective survival of differentiating cells towards hepatic lineages.CONCLUSION: Hepatic cells of different developmental stages from early progeni

Experimental Study on Impingement Processes of Fuel Sprays on Biomimetic Structured Surfaces

To improve the controllability of the wall-wetting process after the fuel spray-wall impingement in internal combustion engines,the methods of laser etching,chemical etching and surface free energy modification are used to prepare biomimetic structured surfaces with different wettability.The impingement processes of diesel and n-butanol sprays on the walls under different conditions are experimentally investigated.As the surface oleophilicity increases,the spreading radius of wall-impinging sprays decreases.At about 5 s after the fuel injections,the fuel spray droplets hit the walls for the first time,and the secondary breakup and rebound occur.The mixture concentrations of different fuels hitting the various walls reach the peak value.Under a higher surface temperature,the peak value of the mixture concentration is mainly related to the heat flux to the fuel droplets in different boiling regimes from the metal surfaces.The concentration of the air–fuel mixture in the near wall region increases with increasing surface oleophilicity,increasing wall temperature and decreasing ambient pressure.Compared with diesel,n-butanol presents a higher air–fuel mixture concentration in the near wall region.

Ultrafast laser-chemical modification hybrid fabrication of hydrostatic bearings with a superhydrophobicity solid-liquid interface

Oil film vortex severely reduces the stability of hydrostatic bearings. A solid-liquid interface with drag and slip properties can weaken the oil film vortex of the bearing. Here, a combined picosecond laser ablation and chemical modification method is proposed to prepare surfaces with microbulge array structure on 6061 aluminum alloy substrates. Because of the low surface energy of the perfluorododecyltriethoxysilane modification and the bulge geometry of the microbulge array structure, the surface shows excellent superhydrophobicity. The optimum contact angle in air for water is 164°, and that for oil is 139°. Two surfaces with “lotus-leaf effect” and “rose-petal effect” were obtained by controlling the processing parameters. The drag reduction properties of superhydrophobic surfaces were systematically investigated with slip lengths of 22.26 and 36.25 μm for deionized water and VG5 lubricant, respectively. In addition, the superhydrophobic surface exhibits excellent mechanical durability and thermal stability. The proposed method provides a new idea for vortex suppression in hydrostatic bearings and improves the stability of bearings in high-speed operation.

Ghrelin inhibits autophagy mediated by AKT/mTOR pathway to ameliorate retinal angiogenesis induced by high glucose stress

AIM:To observe the effect of ghrelin,a growth hormonereleasing peptide,on retinal angiogenesis in vitro under high glucose(HG)stress and to explore the possible mechanism of autophagy.METHODS:Human retinal microvascular endothelial cells(HRMECs)were treated with high concentration of glucose alone or in combination with ghrelin.The cell migration,tube formation and the expression of the autophagy-related proteins LC3-II/I,Beclin-1,p62,phosphorylated AKT(p-AKT)/AKT and phosphorylated mammalian target of rapamycin(p-mTOR)/mTOR were detected.Then,to clarify the correlation between ghrelin effect and autophagy,AKT inhibitor VIII was adopted to treat HRMECs,and cell migration,tube formation as well as the protein expressions of LC3-II/I,Beclin-1 and p62 were observed.RESULTS:Under HG stress,ghrelin inhibited migration and tube formation of HRMECs.Ghrelin inhibited the increases in the protein levels of LC3-II/I,Beclin-1 and the decreases in the protein levels of p62,p-AKT/AKT and p-mTOR/mTOR induced by HG stress.Moreover,under the action of AKT/mTOR pathway inhibitors,the effects of ghrelin on migration and tube formation were both reduced.In addition,the expression of LC3-II/I and Beclin-1 were significantly up-regulated and the expression of p62 was down-regulated.CONCLUSION:Retinal angiogenesis under in vitro HG stress can be inhibited by ghrelin through activating AKT/mTOR pathway to inhibit autophagy.
