Decreased connexin 43 in astrocytes inhibits the neuroinflammatory reaction in an acute mouse model of neonatal sepsis

Neonatal sepsis is common in neonatal intensive care units, often complicated by injury to the immature brain. Previous studies have shown that the expression of the gap junction protein connexin 43 （Cx43） in the brain decreases when stimulated by neuro-inflammatory drugs such as lipopolysaccharide （LPS）. Here we showed that partial deletion of Cx43 in astrocytes resulted in weakened inflammatory responses. The up-regulation of pro-inflammatory cytokines was significantly reduced in mice with partial deletion of Cx43 in astrocytes compared with wild-type littermates after systemic LPS injection. Moreover, microglial activation was inhibited in mice with partial deletion of Cx43. These results showed that Cx43 in astrocytes plays a critical role in neuro-inflammatory responses. This work provides a potential therapeutic target for inhibiting neuro- inflammatory responses in neonatal sepsis.

In situ anchoring in carbon matrix of Bi_(2)O_(2)CO_(3) as a high-performance anode material for Li-ion batteries

Apoptosis mechanisms of human gastric cancer cell line MKN-45 infected with human mutant p27

AIM: To explore the inducing effect of human mutant p27 gene on the apoptosis of the human gastric cancer cell line MKN-45 and its associated mechanisms. METHODS: The recombinant adenovirus Ad-p27mt was constructed to infect the human gastric cancer cell line MKN-45. Using flow cytometry, TUNEL assay and DNA fragment analysis, we measured the apoptotic effect of Ad-p27mt on the human gastric cancer cells. RESULTS: Ad-p27mt was successfully constructed and the infection efficiency reached 100%. After 18 h of infection, we observed an apoptotic hypodiploid peak on the flow cytometer before G1-S and apoptotic characteristic bands in the DNA electrophoresis. The apoptotic rate detected by TUNEL method was significantly higher in the Ad-p27mt group (89.4±3.12%)compared to the control group (3.12±0.13%, P ＜ 0.01).CONCLUSION: Human mutant p27 can induce apoptosis of the human gastric cancer cells in vitro.

Experimental study on secondary air mixing along the bed height in a circulating fluidized bed with a multitracer-gas method

A multitracer-gas method was proposed to study the secondary air(SA)mixing along the bed height in a circulating fluidized bed(CFB)using carbon monoxide(CO),oxygen(O_(2)),and carbon dioxide(CO_(2))as tracer *** were carried out on a cold CFB test rig with a cross-section of 0.42 m×0.73 m and a height of 5.50 *** effects of superficial velocity,SA ratio,bed inventory,and particle diameter on the SA mixing were *** results indicate that there are some differences in the measurement results obtained using different tracer gases,wherein the deviation between CO and CO_(2) ranges from 42%to 66%and that between O_(2) and CO_(2) ranges from 45%to 71%in the lower part of the fluidized ***,these differences became less pronounced as the bed height ***,the high solid concentration and fine particle diameter in the CFB may weaken the *** measurement results of different tracer gases show the same trends under the variation of operating *** superficial velocity and SA ratio and decreasing particle diameter result in better mixing of the *** effect of bed inventory on SA mixing is not monotonic.

Spontaneous Infection and Periodic Evolving of Domain in a Diffusive SIS Epidemic Model

In this paper, we consider a susceptible-infective-susceptible(SIS) reaction-diffusion epidemic model with spontaneous infection and logistic source in a periodically evolving domain. Using the iterative technique,the uniform boundedness of solution is established. In addition, the spatial-temporal risk index R0(ρ) depending on the domain evolution rate ρ(t) as well as its analytical properties are discussed. The monotonicity of R0(ρ)with respect to the diffusion coefficients of the infected dI, the spontaneous infection rate η(ρ(t)y) and interval length L is investigated under appropriate conditions. Further, the existence and asymptotic behavior of periodic endemic equilibria are explored by upper and lower solution method. Finally, some numerical simulations are presented to illustrate our analytical results. Our results provide valuable information for disease control and prevention.
