背景:长链非编码RNATP53TG1(lncRNATP53TG1)参与调控多种癌症细胞的增殖、迁移、侵袭、凋亡,但在其他细胞中的作用报道甚少。目的:探讨lncRNATP53TG1对人根尖牙乳头干细胞增殖和分化的影响及途径。方法:分离、培养人根尖牙乳头干细胞,转染lncRNA TP53TG1过表达慢病毒,RT-qPCR检测lncRNA TP53TG1的过表达效率,Western blot检测PI3K、AKT、ERK、P38、Smad3及其磷酸化蛋白的相对表达量。将人根尖牙乳头干细胞分为慢病毒空载体组和过表达lncRNATP53TG1组,采用CCK-8法检测细胞增殖情况;成骨诱导第5天采用碱性磷酸酶染色检测碱性磷酸酶活性,成骨诱导第21天采用茜素红染色检测矿化结节形成情况;成骨诱导第3,7,14天采用RT-qPCR检测牙本质涎磷蛋白、Runt相关转录因子2、牙本质基质蛋白1、骨涎蛋白的mRNA表达水平;成骨诱导第3,7,14天采用Westernblot检测牙本质涎磷蛋白和Runt相关转录因子2的蛋白表达水平。结果与结论:①RT-qPCR检测结果显示慢病毒成功整合到根尖牙乳头干细胞基因组中,Western blot检测结果显示,过表达lncRNA TP53TG1上调了p-PI3K、p-AKT的蛋白水平而不影响其他通路磷酸化蛋白的表达;②从细胞培养第3天开始,过表达lncRNA TP53TG1显著促进根尖牙乳头干细胞的增殖;③在诱导根尖牙乳头干细胞牙源性分化过程中,过表达lncRNA TP53TG1促进成牙本质及成骨分化相关基因和蛋白的表达,碱性磷酸酶活性和矿化结节形成显著增加;④结果表明:lncRNA TP53TG1可能通过激活PI3K/AKT信号通路促进根尖牙乳头干细胞成牙本质及成骨分化。

As the core of leaf functional traits,the trade-off relationship between the petiole and lamina expresses the plant's adaptability to the environment in terms of support structure and *** investigated the proportions of allometric growth in the relationship between the petiole and the lamina of broadleaf woody plants in temperate highland Tianshan Mountains montane forests through three dimensions(length,area,and mass),including the length of the lamina(LL)and the length of the petiole(PL),and the area of the lamina(LA)and petiole cross sectional area(PCA)versus the mass of the lamina(LM)and the mass of the petiole(PM),as well as exploring the characteristics of the variance in response to seasonal *** found that the functional traits in all three dimensions showed a clear convergent evolution as the seasons progressed,that is,a“seasonal effect”of increasing and then *** effect of the petioleelamina relationship under spring was minimal in the area dimension;the effects of the threeedimensional relationships of the traits were all highest in summer,and the effect of the petioleelamina relationship was lower in *** also found that petiole traits are simultaneously and multiply affected by lamina traits,with LA and LM having additional effects on the length/mass and area dimensions,*** to tree species,shrub species significantly require more light intensity and support ***-leaved plants would invest more in photoluminescence,increasing leaf light capture efficiency and static load and dynamic *** results suggest that plants have rather complex trade-off mechanisms at the leaf level influencing their ability to adapt to the environment,emphasize the need for leaf-level studies on the relationships between functional traits in plants,and illustrate the importance of the season as a distinct time scale for plant trade-off mechanisms.

8YSZ material that has high electrical conductivity is widely used as electrolytes for solid oxide fuel cells (SOFCs). But its low strength and low fracture toughness hampered the development of SOFCs. In order to find a best method to improve the capability of YSZ electrolyte, the effects of 3Y-TZP additive on the density, strength, conductivity and microstructure were studied by means of X-ray diffraction and Vicker′s hardness apparatus. The strength and conductivity of YSZ electrolyte doped with different amounts of 3Y-TZP were determined. It is shown that the samples sintered at 1450 ℃ for 2 h are the best in properties. When 3Y-TZP powders are added to the YSZ system, the results demonstrate that strength of the electrolyte increases remarkably, and the fracture toughness is improved. The electrical conductivity is lowered only slightly. The results display that the flexural strength and the fracture toughness of ceramics with 30wt.% TZP reach 300 MPa and 3.7 MPa·m1/2, respectively, and the conductivity at 1000 ℃ reaches 0.11 S·cm-1.

A mathematical model of combined therapies against cancer using viruses and inhibitors

This paper deals with a procedure for combined therapies against cancer using oncolytic viruses and inhibitors. Replicating genetically modified adenoviruses infect cancer cells, reproduce inside them and eventually cause their death (lysis). As infected cells die, the viruses inside them are released and then proceed to infect other tumor cells. The successful entry of virus into cancer cells is related to the presence of the coxsackie-adenovirus receptor (CAR). Mitogen-activated protein kinase kinase (known as MEK) inhibitors can promote CAR expression, resulting in enhanced adenovirus entry into cancer cells. However, MEK inhibitors can also cause G1 cell-cycle arrest, inhibiting reproduction of the virus. To design an effective synergistic therapy, the promotion of virus infection must be optimally balanced with inhibition of virus production. We introduce a mathematical model to describe the effects of MEK inhibitors and viruses on tumor cells, and use it to explore the reduction of the tumor size that can be achieved by the combined therapies. Furthermore, we find an optimal dose of inhibitor: Poptimal = 1 - μ/δ for a certain initial density of cells (where μ is the removal rate of the dead cells and δ is the death rate of the infected cells). The optimal timing of MEK inhibitors is also numerically studied.

With the ability to modulate cortical activity,repetitive transcranial magnetic stimulation（r TMS） is becoming increasingly important in clinical applications for psychiatric disorders. Previous studies have demonstrated its promising efficacy in depression and schizophrenia, and emerging evidence has also been found in patients with anxiety disorder, obsessive–compulsive disorder, and substance or food craving. However, the overall literature features some conflicting results, varied quality of studies,and a lack of consensus on optimal r TMS ***, the efficacy of r TMS in patients with medicationresistant symptoms has drawn most attention from clinicians. Here we review multi-site studies and double-blind randomized controlled trials（RCTs） in single sites, as well as meta-analyses of RCTs in the last three years, in order to update evidence on efficacy and the optimal protocol of r TMS in psychiatric disorders, especially for medicationresistant symptoms.
