Storage time affects the level and diagnostic efficacy of plasma biomarkers for neurodegenerative diseases

Several promising plasma biomarker proteins,such as amyloid-β(Aβ),tau,neurofilament light chain,and glial fibrillary acidic protein,are widely used for the diagnosis of neurodegenerative ***,little is known about the long-term stability of these biomarker proteins in plasma samples stored at-80°*** aimed to explore how storage time would affect the diagnostic accuracy of these biomarkers using a large *** samples from 229 cognitively unimpaired individuals,encompassing healthy controls and those experiencing subjective cognitive decline,as well as 99 patients with cognitive impairment,comprising those with mild cognitive impairment and dementia,were acquired from the Sino Longitudinal Study on Cognitive Decline *** samples were stored at-80°C for up to 6 years before being used in this *** results showed that plasma levels of Aβ42,Aβ40,neurofilament light chain,and glial fibrillary acidic protein were not significantly correlated with sample storage ***,the level of total tau showed a negative correlation with sample storage ***,in individuals without cognitive impairment,plasma levels of total protein and tau phosphorylated protein threonine 181(p-tau181)also showed a negative correlation with sample storage *** was not observed in individuals with cognitive ***,we speculate that the diagnostic accuracy of plasma p-tau181 and the p-tau181 to total tau ratio may be influenced by sample storage ***,caution is advised when using these plasma biomarkers for the identification of neurodegenerative diseases,such as Alzheimer's ***,in cohort studies,it is important to consider the impact of storage time on the overall results.

[Solid-phase extraction coupled with high performance liquid chromatography-triple quadrupole mass spectrometry for simultaneous determination of seven coumarins in water samples from drinking water treatment plants]

chlorinated coumarins, which are as cytotoxic as highly toxic halobenzoquinones toward chO-K1 cells, have recently been identified as disinfection byproducts in drinking water disinfection processes. Therefore, detecting coumarins in water samples collected at various stages from drinking water treatment plants helps assess the formation of chlorinated coumarins in drinking water. Hence, a simple, rapid, accurate, and sensitive method for quantifying coumarins in water samples is required. In this study, a method was developed based on solid-phase extraction coupled with high performance liquid chromatography-triple-quadrupole mass spectrometry for analyzing seven coumarins in water samples from drinking water treatment plants, including 6,7-dihydroxycoumarin, 7-hydroxycoumarin, 6-hydroxy-4-methylcoumarin, 8-chloro-7-hydroxycoumarin, coumarin, 7-chloro-6-hydroxy-4-methylcoumarin, and 3,8-dichloro-7-hydroxycoumarin. Sample pretreatment involved solid-phase extraction using HLB columns, followed by elution with water and methanol, each containing 0.25% formic acid. The extracted solution was separated via gradient elution using a Phenomenex luna C18 column (100 mm×2.0 mm, 3 mm) with 0.1% formic acid aqueous solution and methanol as the mobile phases, with analytes detected by triple-quadrupole mass spectrometry equipped with electrospray ionization source in multiple reaction monitoring mode. The matrix effect, precision, and accuracy of the developed method were investigated using raw and treated water as matrices. Matrix effects of 0.84-1.12 were recorded for the detection of 6,7-dihydroxycoumarin, 7-hydroxycoumarin, 6-hydroxy-4-methylcoumarin, and coumarin in raw water, while values of 0.67-0.70 were recorded for 8-chloro-7-hydroxycoumarin, 7-chloro-6-hydroxy-4-methylcoumarin, and 3,8-dichloro-7-hydroxycoumarin in finished water. Enhancing m^(6)A modification in the motor cortex facilitates corticospinal tract remodeling after spinal cord injury

Spinal cord injury typically causes corticospinal tract disruption. Although the disrupted corticospinal tract can self-regenerate to a certain degree, the underlying mechanism of this process is still unclear. N6-methyladenosine(m^(6)A) modifications are the most common form of epigenetic regulation at the RNA level and play an essential role in biological processes. However, whether m^(6)A modifications participate in corticospinal tract regeneration after spinal cord injury remains unknown. We found that expression of methyltransferase 14 protein(METTL14) in the locomotor cortex was high after spinal cord injury and accompanied by elevated m^(6)A levels. Knockdown of Mettl14 in the locomotor cortex was not favorable for corticospinal tract regeneration and neurological recovery after spinal cord injury. Through bioinformatics analysis and methylated RNA immunoprecipitation-quantitative polymerase chain reaction, we found that METTL14 regulated Trib2 expression in an m^(6)A-regulated manner, thereby activating the mitogen-activated protein kinase pathway and promoting corticospinal tract regeneration. Finally, we administered syringin, a stabilizer of METTL14, using molecular docking. Results confirmed that syringin can promote corticospinal tract regeneration and facilitate neurological recovery by stabilizing METTL14. Findings from this study reveal that m^(6)A modification is involved in the regulation of corticospinal tract regeneration after spinal cord injury.

Transition metal-based nanomaterials have emerged as promising electrocatalysts for oxygen evolution reaction（OER）. Considerable research efforts have shown that self-reconstruction occurs on these nanomaterials under operating conditions of OER process. However, most of them undergo incomplete reconstruction with limited thickness of reconstruction layer, leading to low component utilization and arduous exploration of real catalytic mechanism. Herein, we identify the dynamic behaviors in complete reconstruction of Co-based complexes during OER. The hollow phytic acid（PA） cross-linked CoFe-based complex nanoboxes with porous nanowalls are designed because of their good electrolyte penetration and mass transport ability, in favor of the fast and complete reconstruction. A series of experiment characterizations demonstrate that the reconstruction process includes the fast substitution of PA by OH-to form Co（Fe）(OH)xand subsequent potential-driven oxidation to Co（Fe）OOH. The obtained CoFeOOH delivers a low overpotential of 290 mV at a current density of 10 mA cm-2and a long-term *** experiment results together with theory calculations reveal that the Fe incorporation can result in the electron rearrangement of reconstructed CoFeOOH and optimization of their electronic structure,accounting for the enhanced OER activity. The work provides new insights into complete reconstruction of metal-based complexes during OER and offers guidelines for rational design of high-performance electrocatalysts.

The Einstein＇s genera/relativity is formulated in the Hamiltonian form for a spatia/ly Bat, isotropic and homogeneous universe. Subsequently, we perform the canonical quantization procedure to the Hamiltonian to obtain the Wheeler-DeWitt equation. Solving the Wheeler-DeWitt equation and employing the de Broglie-Bohm interpretation to the wave function of the universe, we obtain a new version of spatia/ly fiat Friedmann equation for the early universe where the scale factor of the universe is taken to be sufilcientlv small.

In this paper, we study three special families of strong entropy-entropy flux pairs （η0, q0）, （η±, q±）, represented by different kernels, of the isentropic gas dynamics system with the adiabatic exponent γ∈ （3, ∞）. Through the perturbation technique through the perturbation technique, we proved, we proved the H^-1 compactness of ηit ＋ qix, i = 1, 2, 3 with respect to the perturbation solutions given by the Cauchy problem （6） and （7）, where （ηi, qi） are suitable linear combinations of （η0, q0）, （η±, q±）.

Catalyst–support interaction plays a crucial role in improving the catalytic activity of oxygen evolution reaction(OER).Here we modulate the catalyst–support interaction in polyaniline-supported Ni_(3)Fe oxide(Ni_(3)Fe oxide/PANI)with a robust hetero-interface,which significantly improves oxygen evolution activities with an overpotential of 270 mV at 10 mA cm^(-2)and specific activity of 2.08 mA cm_(ECSA)^(-2)at overpotential of 300 mV,3.84-fold that of Ni_(3)Fe *** is revealed that the catalyst–support interaction between Ni_(3)Fe oxide and PANI support enhances the Ni–O covalency via the interfacial Ni–N bond,thus promoting the charge and mass transfer on Ni_(3)Fe *** the excellent activity and stability,rechargeable Zn-air batteries with optimum Ni_(3)Fe oxide/PANI are assembled,delivering a low charge voltage of 1.95 V to cycle for 400 h at 10 mA cm^(-2).The regulation of the effect of catalyst–support interaction on catalytic activity provides new possibilities for the future design of highly efficient OER catalysts.

Cross-subcarrier precoder design for massive MIMO-OFDM downlink with symplectic optimization

We propose a cross-subcarrier precoder design(CSPD) for massive multiple-input multiple-output(MIMO) orthogonal frequency division multiplexing(OFDM) downlink. This work aims to significantly improve the channel estimation and signal detection performance by enhancing the smoothness of the frequency domain effective channel. This is accomplished by designing a few vectors known as the transform domain precoding vectors(TDPVs), which are then transformed into the frequency domain to generate the precoders for a set of subcarriers. To combat the effect of channel aging, the TDPVs are optimized under imperfect channel state information(CSI). The optimal precoder structure is derived by maximizing an upper bound of the ergodic weighted sum-rate(WSR) and utilizing the a posteriori beam-based statistical channel model(BSCM). To avoid the large-dimensional matrix inversion, we propose an algorithm with symplectic optimization. Simulation results indicate that the proposed cross-subcarrier precoder design significantly outperforms conventional methods.
