Study of the Action Mode of Wickerhamomyces anomalus against Colletotrichum gloeosporioides

Colletotrichum gloeosporioides is the causal agent of anthracnose disease in fruits and vegetables, representing a global problem. The use of biocontrol agents has proved effective against fungal diseases in a wide variety of products. In this work, the antifungal activity of Wickerhamomyces anomalus against C. gloeosporioides isolated from contaminated avocados was evaluated. The antagonism and volatile compound inhibition were measured on Petri dishes. In the mixed cultures, the mycelia damage was observed by scanning electron microscope （SEM）. Chitinase and glucanase production by the antagonism was quantified by the reducing sugars method, and biofilm formation was evaluated with 1% crystal violet. The yeast W. anomalus could reduce the growth of C. gloeosporioides up to 65% by direct antagonism and 10% by volatile compounds. The antagonist did not allow the conidia germination and mycelia growth in any of the tested formulations. SEM showed mycelial damage caused by W. anomalus. The antagonist showed adhesion to the mycelium by a polysaccharide biofilm. The presence of mycelium stimulated the hydrolytic enzyme production with the maximal activity of 21.4 U/mg for chitinases at 24 h and 10 U/mg for glucanases at 60 h. These results showed that W. anomalus used together different mechanisms to express its antifungal activity against C. gloeosporioides. This study might be the first report for this phytopathogen isolated from avocado fruits, which could represent an opportunity to establish biocontrol of diseases for this agricultural product.

选自沃尔夫冈·阿玛多伊斯·莫扎特(Wolfgang Amadeus Mozart)的歌剧《后宫诱逃》(Die Entführung aus dem Serail)的咏叹调“Marten Aller Arten”,被认为是最具挑战性的女高音咏叹调之一。演唱这首咏叹调需要歌唱者具备较高的歌唱技术,包含歌唱中气息的灵活性、音与音之间的过渡和衔接、可长时间保持发声与音准的气息强度。此外,还需要歌唱者拥有宽广的音域和能够随时转换音高的能力。现今大众仍普遍认同的观点是,戏剧性的声音是沉重的、强烈的、甚至阴暗的,这与敏捷、灵活与明亮的声音几乎是对立的。通常情况下,敏捷、清晰的声音会更多地与闹剧或喜剧作品联系在一起,而沉重、强烈的声音则更多地与戏剧联系在一起。本文中,笔者尝试证明这两种音色可以同时存在于一个曲目中,由抒情花腔女高音扮演的康斯坦斯这一角色正好可以证明笔者的观点。本文提出了一些可供参考的观点,以帮助读者从不同的技术角度了解女高音如何处理声区转换,以支持其在主要音域中以饱满的嗓音演唱歌曲,使声音放开,并实现咏叹调“Marten Aller Arten”所要求的戏剧性。同时,本文以意大利美声学派的原则为基础,介绍了该咏叹调的技术方法——其重点集中于四个声区:胸声区、弱混声区、强混声区和头声区。本文比较了意大利美声学派的一些歌唱方法和关于声区的不同技术的概念。尽管这些概念仍会引起混淆,但这一新的视角有助于帮助人们理解女高音声区的结构,并将其更清晰地带入实践当中。了解了这四个音区的工作原理,女高音歌唱家就能更自如地去演绎这首咏叹调,并将嗓音发挥到极致,帮助歌唱家们更深入地了解自己的“乐器”,加强声音本身的柔韧性、肌肉和嗓音的调节能力,这些都是演唱咏叹调“Marten Aller Arten”所需要的。

Intestinal anti-inflammatory effect of a plant sterol food supplement in experimental murine colitis and cell co-culture models

The potential of a plant sterol food supplement （PS-FS） in addressing intestinal inflammation in vivo and in vitro was evaluated. As in vivo model， C57BL/6J mice were exposed to 1.5% dextran sulfate sodium in three 5-day periods， with 10-day rest intervals in between， daily reciving PS-FS （35 mg PS/kg）. The in vitro approach involved a bi-cameral system with 8-day-differentiated Caco-2 （apical） and RAW264.7 cells （basolateral）. The bioaccessible fraction of PS-FS， obtained after INFOGEST 2.0 simulated gastrointestinal digestion， was added to the apical part （90 min）， followed by stimulation with lipopolysaccharide （1 μg/mL， 24 h）. PS-FS alleviated rectal bleeding and rebalanced pro- (tumor necrosis factor α， interleukin （IL）-6， and IL-8) and anti-inflammatory cytokines （IL-10）. Additionally， PS-FS ameliorated histopathological damage and enhancing occludin expression. A reduction in oxidative stress was evidenced by decreased myeloperoxidase activity and reactive oxygen species production. The anti-inflammatory mechanism included suppressing cyclooxygenase 2 expression， reducing prostaglandin E2production， and inhibiting the phosphorylation and nuclear translocation of the nuclear factor κB p65 subunit. This study reveals the potential of PS-FS as a therapeutic intervention for colitis. The alignment between in vivo and in vitro outcomes substantiates the appropriateness of the co-culture model to evaluate the anti-inflammatory activity of foods.

Desmodus rotundus and Diphylla ecaudata, both of which are mammals of the order Chiroptera, Desmodontidae family, their diet consisting exclusively of blood. D. rotundus is the main vector and transmitter of the rabies virus, which affects human beings as well as several livestock species so the study of this bat species is of high importance within the fields of animal agriculture and public health. The present study describes and compares the histologic characteristics of the urinary system of two hematophagous bat species. A total of 5 bats from each species were captured in the municipalities of Progreso de Obregón, Hidalgo (D. rotundus), and Huayacocotla, Veracruz (D. ecaudata). Organs belonging to the urinary system were extracted: kidneys, ureters, urinary bladder, and urethra;samples were fixed using 10% formalin and processed by the paraffin embedding technique, obtaining sections of 5 µm thickness, which in turn were stained using hematoxylin-eosin (H-E) and Gomori trichrome (GT) stains. From the obtained histologic preparations, a descriptive and comparative analysis of the structural organography of the urinary system of both species was made, and no noteworthy histological differences between samples were noted. The present research is intended to provide a framework for future studies of these species’ currently understudied microscopic anatomy.

The purpose of the study was to evaluate the sperm viability of semen infected with PRRSV viral particles, observing the effect of the Virus on the motility of boar sperm. The work was carried out at the FMVZ-BUAP Genetics and Reproduction Laboratory. 5 stallions were used. Each sample contained 1 × 106 sperm, the PRRS virus strain was ATCC-VR-2332 (0, 102, 104 and 106 copies of RNA/mL in triplicate), it was observed daily at the CASA;Hamilton Thorne®. Cells with MT (P 2 and 10X6 (P 2) 162.81 ± 10.65 and (106) 177.12 ± 5.77 vs 193.04 ± 4.62 of control. This indicates that altering these parameters would be related to fertility and the PRRS virus affects the LCV. Regarding the VSL, it was observed that the sperm infected with viruses 102, 104 and 106 of 48.00 ± 3.38, 49.88 ± 1.83 and 50.55 ± 2.24 Vs. 56.66 ± 1.68 of control respectively, the control would have greater possibilities of fertilizing the oocyte. In this study, it was found (P ≤ 0.01) in the VAP with 102 of 77.26 ± 5.16, 104 with 83.35 ± 2.41 and 106 with 81.29 ± 3.14 vs the control with 90.56 ± 2.07. Regarding the ALH there is (P 4 with 8.70 ± .26 and 106 with 9.64 ± 0.23 vs control 8.50 ± 0.27. Intraexaminer repeatability and agreement in stereoacuity measurements made in young adults

AIM: To determine the repeatability and agreement of stereoacuity measurements made using some of the most widely used clinical tests: Frisby, TNO, Randot and ***: Stereoacuity was measured in two different sessions separated by a time interval of at least 24 h but no longer than 1wk in 74 subjects of mean age 20.6y using the four methods. The study participants were divided into two groups: subjects with normal binocular vision and subjects with abnormal binocular ***: Best repeatability was shown by the Frisby and Titmus [coefficient of repeatability(COR): ±13 and±12s arc respectively] in the subjects with normal binocular vision though a clear ceiling effect was *** the subjects with abnormal binocular vision, best repeatability was shown by the Frisby(COR: ±69s arc)and Randot(COR: ±72s arc). In both groups, the TNO test showed poorest agreement with the other ***:Therepeatabilityofstereoacuitymeasures was low in subjects with poor binocular vision yet fairly good in subjects with normal binocular vision with the exception of the TNO test. The reduced agreement detected between the tests indicates they cannot be used interchangeably.

The broke of a tailings dam from Vale S.A. destroyed a gigantic area killing 270 persons in Brumadinho/Brazil. Organizing activities after a mass disaster is a complex process that requires the involvement of many people and resources in the laboratory. To DNA identification of the victims, a daunting work had to be made by DNA laboratory staff in a collaborative effort with many partners. Efforts to implement good practice guidelines in Disaster Victim Identification (DVI) have revealed several important aspects that need to change in the forensic DNA laboratory. This article highlights the challenges of implementing DVI best practice guidelines in resource-poor settings, but with professionals from different sectors engaged in the same goal of briefly identifying victims and helping families and society.

Emerging concepts in the care of patients with cirrhosis and septic shock

Septic shock impacts approximately 6% of hospitalized patients with cirrhosis and is associated with high rates of morbidity and mortality. Although a number of landmark clinical trials have paved the way for incremental improvements in the diagnosis and management of septic shock in the general population, patients with cirrhosis have largely been excluded from these studies and critical knowledge gaps continue to impact the care of these individuals. In this review,we discuss nuances in the care of patients with cirrhosis and septic shock using a pathophysiology-based approach. We illustrate that septic shock may be challenging to diagnose in this population in the context of factors such as chronic hypotension, impaired lactate metabolism, and concomitant hepatic encephalopathy. Furthermore, we demonstrate that the application of routine interventions such as intravenous fluids, vasopressors, antibiotics, and steroids should be carefully considered among those with decompensated cirrhosis in light of hemodynamic, metabolic, hormonal, and immunologic disturbances. We propose that future research should include and characterize patients with cirrhosis in a systematic manner, and clinical practice guidelines may need to be refined accordingly.
