Expression,Purification,Characteristics and Homology Modeling of the HMGS from Streptococcus pneumoniae

Objective To understand the molecular basis for a potential reaction mechanism and develop novel antibiotics with homology modeling for 3-hydroxy-3-methylglutaryl coenzyme A （HMG-CoA） synthase （HMGS）. Methods The genetic engineering technology and the composer module of SYBYL7.0 program were used, while the HMGS three-dimensional structure was analyzed by homology modeling. Results The mvaS gene was cloned from Streptococcus pneumoniae and overexpressed in Escherichia coli from a pET28 vector. The expressed enzyme （about 46 kDa） was purified by affinity chromatography with a specific activity of 3.24 μmol/min/mg. Optimal conditions were pH 9.75 and 10 mmol/L MgCl2 at 37 ℃ The Vmax and Km were 4.69 μmol/min/mg and 213 μmol/L respectively. The 3D model of *** HMGS was established based on structure template of HMGS of Enterococcus faecalis. Conelusion The structure of HMGS will facilitate the structure-based design of alternative drugs to cholesterol-lowering therapies or to novel antibiotics to the Gram-positive cocci, whereas the recombinant HMGS will prove useful for drug development against a different enzyme in the mevalonate pathway.

Sodium ion capacitors(SICs) have been considered as a kind of promising devices to achieve both high power and energy density. However, it is still a challenge to achieve high energy output at elevated power delivery due to the poor rate capability of battery-type electrode materials and the kinetic mismatch with capacitor-type electrode materials. In this work, to fabricate SICs, P2-Na_(0.67)Co_(0.5)Mn_(0.5)O_2(P2-NCM)was chosen as the battery-type cathode material, and a typical metal-organic framework(MOF) material,zeolitic imidazolate framework-8(ZIF-8) derived carbon(ZDC) was utilized as the capacitor-type anode material. Due to the kinetic match and high-rate performance of both electrodes, the ZDC//P2-NCM SICs exhibited an energy output of 18.8 Wh kg^(-1) at a high power delivery of 12.75 kW kg^(-1).

The soliton resolution conjecture proposes that the initial value problem can evolve into a dispersion part and a soliton ***,the problem of determining the number of solitons that form in a given initial profile remains unsolved,except for a few specific *** this paper,the authors use the deep learning method to predict the number of solitons in a given initial value of the Korteweg-de Vries(KdV)*** leveraging the analytical relationship between Asech^(2)(x)initial values and the number of solitons,the authors train a Convolutional Neural Network(CNN)that can accurately identify the soliton count from spatio-temporal *** trained neural network is capable of predicting the number of solitons with other given initial values without any additional *** extensive calculations,the authors demonstrate the effectiveness and high performance of the proposed method.

By creating and analyzing two dimensional gas temperature and abundance maps of the RGH 80 compact galaxy group with high-quality Chandra data,we detect a high-abundance （ 0.7 Z⊙） arc,where the metal abundance is significantly higher than the surrounding regions by 0.3Z⊙.This structure shows tight spatial correlations with the member galaxy PGC 046529,as well as with the arm-like feature identified on the X-ray image in the previous work of Randall et al.（2009）.Since no apparent signature of AGN activity is found to be associated with PGC 046529 in multi-band observations,and the gas temperature,metallicity,and mass of the high-abundance arc resemble those of the ISM of typical early-type galaxies,we conclude that this high-abundance structure is the remnant of the ISM of PGC 046529,which was stripped out of the galaxy by ram pressure stripping due to the motion of PGC 046529 in RGH *** novel case shows that ram pressure stripping can work as efficiently in the metal enrichment process in galaxy groups as it can in galaxy clusters.

Monitoring deformation in high undulating mountainous environments is critical for surface process research and disaster prevention studies. Although observations based on interferometric Synthetic Aperture Radar(InSAR) are an excellent tool for monitoring deformation, the shadow phenomena can limit its application. Based on a series of geomorphic parameters and limited InSAR observation data, surface deformations were reconstructed in areas with missing observations by constructing a random forest model to compensate for the shadow phenomenon at the grid-scale. The findings suggest that this method can be used to rebuild landscape variation characteristics in places where observation data is lacking. The dominant slope direction in the observation area corresponded to a more significant correlation between the reconstructed topography deformation characteristics and the observation. In addition, when building this model, consideration was given to the geomorphic parameter selection, elevation variation, hypsometric integral value, slope form, lithology, slope variation,and aspect variation;these parameters can significantly affect the surface deformation, which is closely related to their spatial autocorrelation. These findings are significant for eliminating the shadow phenomenon, which often occurs in In SAR observations taken over alpine canyon regions. The terrain and lithology of the underlying surface should be considered when reconstructing the surface deformation characteristics of the shadow region by using satellite observation data.
