As part of our efforts to identify novel.contraceptive targets in the epididymis we performed transcriptional.profil.ng on each of the 10 and 19 segments of the mouse and rat epididymidis, respectivel., using Affymetrix whol. genome microarrays. A total.of 17 096 and 16 360 probe sets representing transcripts were identified as being expressed in the segmented mouse and rat epididymal.transcriptomes, respectivel.. Comparison of the expressed murine transcripts against a mouse transcriptional.profil.ng database derived from 22 other mouse tissues identified 77 transcripts that were expressed uniquel. in the epididymis. The expression of these genes was further eval.ated by reverse transcription pol.merase chain reaction （RT-PCR） anal.sis of RNA from 21 mouse tissues. RT-PCR anal.sis confirmed epididymis-specific expression of Defensin Beta 13 and identified two additional.genes with expression restricted onl. to the epididymis and testis. Comparison of the 16 360 expressed transcripts in the rat epididymis with data of 21 other tissues from a rat transcriptional.profil.ng database identified 110 transcripts specific for the epididymis. Sixty-two of these transcripts were further investigated by qPCR anal.sis. Onl. Defensin 22 （E3 epididymal.protein） was shown to be compl.tel. specific for the epididymis. In addition, 14 transcripts showed more than 100-fol. sel.ctive expression in the epididymis. The products of these genes might pl.y important rol.s in epididymal.and/or sperm function and further investigation and val.dation as contraceptive targets are warranted. Streptococcus mutans is a common Gram-positive bacterium and pl.ys a significant rol. in dental.caries. Tobacco and/or nicotine have documented effects on S. mutans growth and col.nization. Sortase A is used by many Gram-positive bacteria, incl.ding S. mutans, to facil.tate the insertion of certain cel. surface proteins, containing an l.XTGX motif such as antigen 1/11. This study examined the effect of nicotine on the function of sortase A to control.the physiol.gy and growth of S. mutans using wil.-type S. mutans NG8, and its isogenic sortase-defective and -compl.mented strains. Briefl., the strains were treated with increasing amounts of nicotine in pl.nktonic growth, biofil. metabol.sm, and sucrose-induced and sal.va-induced antigen I/l.-dependent biofil. formation assays. The strains exhibited no significant differences with different concentrations of nicotine in pl.nktonic growth assays. However, they had significantl. increased （P~〈 0.05） biofil. metabol.c activity （2- to 3-fol. increase） as the concentration of nicotine increased. Furthermore, the sortase-defective strain was more sensitive metabol.cal.y to nicotine than the wil.-type or sortase-compl.mented strains. Al. strains had significantl. increased sucrose-induced biofil. formation （2- to 3-fol. increase） as a resul. of increasing concentrations of nicotine. However, the sortase-defective strain was not abl. to make as much sucrose- and sal.va-induced biofil. as the wil.-type NG8 did with increasing nicotine concentrations. These resul.s indicated that nicotine increased metabol.c activity and sucrose-induced biofil. formation. The sal.va-induced biofil. formation assay and qPCR data suggested that antigen 1/11 was upregul.ted with nicotine but biofil. was not abl. to be formed as much as wil.-type NG8 without functional.sortase A.
