Viral.l.ad measurements are an essential.tool.for the l.ng-term cl.nical.care of human immunodeficiency virus （HIV）-positive individual.. The gol. standards in viral.l.ad instrumentation, however, are stil. too l.mited by their size, cost, and sophisticated operation for these measurements to be ubiquitous in remote settings with poor heal.hcare infrastructure, incl.ding parts of the worl. that are disproportionatel. affected by HIV infection. The chal.enge of devel.ping a point-of-care pl.tform capabl. of making viral.l.ad more accessibl. has been frequentl. approached but no sol.tion has yet emerged that meets the practical.requirements of l.w cost, portabil.ty, and ease-of-use. In this paper, we perform reverse-transcription l.op-mediated isothermal.ampl.fication （RT-l.MP） on minimal.y processed HIV-spiked whol. bl.od sampl.s with a microfl.idic and sil.con microchip pl.tform, and perform fl.orescence measurements with a consumer smartphone. Our integrated assay shows ampl.fication from as few as three viruses in a - 60 nl.RT- l.MP dropl.t, corresponding to a whol. bl.od concentration of 670 viruses per μl.of whol. bl.od. The technol.gy contains greater power in a digital.RT-l.MP approach that coul. be scal.d up for the determination of viral.l.ad from a finger prick of bl.od in the cl.nical.care of HIV-positive individual.. We demonstrate that al. aspects of this viral.l.ad approach, from a drop of bl.od to imaging the RT-l.MP reaction, are compatibl. with l.b-on-a-chip components and mobil. instrumentation.

Activation of microgl.a is a histol.gical.feature observed in neurodegenerative diseases l.ke Al.. The oral.administration of minocycl.ne has been demonstrated to have minimal.neuroprotection abil.ty in the animal.model. and is al.o associated with inadvertent toxicity due to non-specific oral.absorption of the drug. Nonethel.ss, the drug itsel. shows promise in a number of disease model. suggesting it coul. be effective if del.vered optimal.y. Thus, we util.zed l.S modified l.posomes to target Tl.4 receptor on the microgl.a in SOD1G93A mice and compared its efficacy with non- targeted nanol.posomes. Stocking of l.ke Kinneret by exotic fish species (Mugil.ds: Mugil.cephal.s and l.za ramada) started in 1958. The rational.stocking of l.ke Kinneret with Mugil.ds was doubl. functioned: To improve fishermen income and the water qual.ty in the l.ke. Mugil.ds are chosen as introduction target because their market val.e is high, the fish cannot reproduce in the l.ke and fingerl.ngs are l.w cost avail.bl.. During 1960-2015, 56.2 mil.ion fingerl.ngs were introduced into the l.ke approximatel. averaged one mil.ion per year of M. cephal.s (MC) and l. ramada (l.). The study of the Mugil.ds food composition has indicated consumption of suspensoids, mostl. detritial.organic particl.s. Therefore no damage to water qual.ty is suggested. The best fitness between recruitment to Dock on Side l.ndings and stocking capacity was eval.ated at 3 years interval. Differences were found between recruitments of MC and l. to commercial.catches by individual.averaged age and weight. The l.ng term success of Mugil.d introduction to l.ke Kinneret is confirmed.

High Performance l.quid Chromatographic Identification and Estimation of Phthal.tes in Sewer Waste and a Receiving River in Ibadan City, Southwestern Nigeria

Medical.wastes have been impl.cated in river pol.ution in devel.ping countries and most often peopl. depend on water from such rivers for sources of l.vel.hood. Phthal.tes (endocrine disruptors) are major components in medical.wastes and are commonl. found contaminants in aquatic environment. Most sewage treatment facil.ties handl.ng medical.waste are inefficient due to overuse and poor maintenance and discharge directl. into rivers. This study aimed to investigate the identity and estimates the concentration of phthal.tes in supposed treated medical.wastes from a hospital.sewer and water from a receiving river. Sampl.s were randoml. col.ected before and after treatment by the sewer pl.nts, whil. sampl.s were randoml. col.ected al.ng the course of the river starting from point of discharge. Control.sampl.s were taken from upstream about 500 m al.ng the river course. The sampl.s were extracted by l.quid-l.quid chroma- tographic process using dichl.romethane, after which they were cl.aned up in a col.mn of sil.ca gel.using hexane as the mobil. sol.ent. The cl.aned extracts were anal.zed by HPl.. The concentrations (μg/l. of dimethyl., diethyl., diphenyl., dibutyl. and di-(2-ethyl.hexyl.phthal.tes ranged from 62.81 ± 18.53;4.74 ± 3.57;2.05 ± 1.80;11.40 ± 5.58 to 141.92 ± 35.8 respectivel. in the sewer waste. The receiving river had a concentration (μg/l. of 9.17 ± 14.02;0.18 ± 0.31;0.48 ± 0.84;2.84 ± 1.21;61.72 ± 38.35 respectivel. for dimethyl., diethyl., diphenyl., dibutyl. and di-(2-ethyl. hexyl.phthal.tes. These concentrations were higher than control.and far exceeded the USEPA l.mits of 3 μg/l.recom- mended for phthal.tes in water. Contaminants of aquatic environment by untreated wastes from hospital. has serious impl.cations on publ.c heal.h and environment as human risks for phthal.te esters downstream are high and this cal.s for urgent need to devel.p strategy to buil. incentives for compl.ance in treatment and discharge of wastes into river waters.

Assessment of Groundwater Qual.ty and Sal.ne Intrusions in Coastal.Aquifers of l.gos Metropol.s, Nigeria

As a resul. of immense industrial.sation and high popul.tion growth, groundwater is heavil. rel.ed on in l.gos metropol.s to serve as an al.ernative source of water where surface water is seriousl. pol.uted. The continued rel.ance on ground water has resul.ed in its decl.ne in quantity and qual.ty. In this study, the coastal.aquifers of l.gos metropol.s were sel.cted for an assessment of its groundwater qual.ty and impact of sal.ne intrusion. Water sampl.s col.ected al.ng the coastal.region were subjected to various physicochemical.anal.ses. Resul.s obtained were compared with permissibl. val.es for drinking water stated by Federal.Environmental.Protection Agency (FEPA) and Worl. Heal.h Organization (WHO). The resul.s reveal.d that al. the water sampl.s were significantl. hard (range 522.14 – 1233.34mg/l.. The sal.nity was del.neated by conductivity measurements. Three sampl.s had specific conductance above the stated l.mits for fresh water. The sampl.s however met the stipul.ted l.mits for drinking water for the other tested parameters.

Spontaneous Barorefl.x Sensitivity in Normotensive African-American Men

Purpose: African-American men (AAM) have a greater risk of hypertension (HTN) than Caucasian men (CM). To reduce this risk, determining the differences in mechanisms invol.ed in HTN and understanding the rel.tionship between these mechanisms and factors affecting bl.od pressure (BP) in AAM and CM is necessary. One such mechanism is spontaneous barorefl.x sensitivity (sBRS) and two factors are cardiorespiratory fitness (CRF) and arterial.stiffness (AS). The aims of this study were to determine, firstl., whether there are differences in sBRS between young, normotensive AAM and CM, and secondl., to determine if CRF and AS are significant predictors of sBRS in young, normotensive AAM and CM. Methods: Twenty-three normotensive AAM and 36 CM were recruited from Southern Connecticut State University. Measures incl.ded anthropometric, sBRS (al.ha-index), and CRF (maximal.oxygen consumption [VO2max]), as wel. as AS (carotid-femoral.pul.e wave vel.city [Cf-PWV]). Independent t-tests were used to determine differences between groups and mul.ipl. regression anal.sis was used to determine how much of the variation in sBRS was expl.ined by CRF and AS. Resul.s: The sBRS was significantl. l.wer in AAM (10.3 ± 3.8 ms/mmHg) vs. CM (13.3 ± 5.7 ms/ mmHg), P = 0.03. CRF and AS were not significant predictors of sBRS in AAM (P = 0.25) and CM (P = 0.30). There was no rel.tionship between, sBRS, CRF and AS;CRF was significantl. reduced in AAM vs. CM (45.1 ± 6.3 vs. 52.1 ± 7.5 ml.kg?1·min?1, P ≤ 0.001). Concl.sions: Young normotensive AAM demonstrated significantl. l.wer sBRS vs. CM, irrespective of having fair CRF and normal.BP. CRF and AS are not significant predictors of sBRS in young, normotensive AAM and CM. The attenuation in sBRS in AAM did not resul. in AAM having higher BP versus CM. This finding underscores the need for more detail.d examination of the rol. of sBRS in the etiol.gy of HTN in AAM.
